    Notes: A genome scan, searching for quantitative trait loci (QTL) for the traits cold tolerance and body weight in tilapia, was performed on a cross between a (Oreochromis niloticus×Sarotherodon galilaeus) male and a (O. mossambicus×O. aureus) female. Fifty-four microsatellites and 23 amplified fragment length polymorphism (AFLP) primer combinations were genotyped and tested for marker–trait associations. Sex-specific linkage maps were constructed from this data. Twenty-three point-wise significant marker–trait associations were found in the genome scan, and putative QTL were subsequently tested in another (On×Sg) × (Om×Oa) family. None of the putative QTL from the first experiment were significant in the second experiment. However, one microsatellite, UNH130, found to be associated to weight in the first experiment, was found to be strongly associated to cold tolerance in the second experiment. Since QTL for cold tolerance and body weight were recently found on the linkage group containing UNH130 (linkage group 23) in another study, this linkage group was investigated more closely using interval mapping. The results provide indications, but not conclusive evidence, of a QTL for cold tolerance on linkage group 23.

    Notes: Abstract Unusual phenotypic distributions at the muscle-specific, duplicate aspartate aminotransferase (AAT) loci were found in wild populations of brook trout (Salvelinus fontinalis), a species of the tetraploid-derivative Salmonidae. Analysis of these phenotypic distributions ruled out disparate gene frequencies, nonrandom association between the two loci, and inbreeding as possible explanations; however, models incorporating a null allele fit the data. Inheritance data from hatchery populations of brook trout also indicated a null allele polymorphism. This proposed AAT null allele, along with other null allele polymorphisms in salmonids, is evidence that loss of duplicate gene expression is still occurring. In contrast, there is no such evidence of ongoing loss of duplicate gene expression in the Catostomidae, another tetraploid-derivative lineage. We interpret this and other differences between salmonids and catostomids as reflecting an autotetraploid origin for salmonids and an allotetraploid origin for catostomids. The significance of these findings is also considered with respect to current models of the rate of loss of duplicate gene expression in tetraploid-derivative organisms.

    Notes: Abstract The results of 107 pairwise examinations of joint segregation of biochemical and skin color loci in rainbow trout, Salmo gairdneri, are presented. Three examinations revealed significant nonrandom assortment: Idh-3 with Me-2, Ada-1 with G3p-3, and Mdh-3 with Mdh-4. We believe the first two instances to be cases of classical linkage and the latter instance to reflect pseudolinkage based on similar findings in Salvelinus. All known salmonid linkage associations are reviewed. The results of this summary indicate a high degree of genome conservatism among genera within the Salmonidae, which would seem to be in contrast to the highly plastic genome of this family based on karyotypic data. Data are presented which negate the view that Robertsonian fusion of homologous acrocentric chromosome arms was the preferred mode of metacentric formation in salmonid evolution.

    Notes: Abstract Electrophoretic variation is described for malic enzyme (ME) for the first time in brook trout (Salvelinus fontinalis). Since the quaternary structure of ME was not clear from examination of banding patterns in brook trout alone, ME phenotypes in rainbow trout (Salmo gairdneri) × brook trout hybrids as well as in esocid species demonstrated that ME is tetrameric. A model of two duplicated loci is proposed to account for the observed variation. One locus (ME-2) is fixed and one locus (ME-1) is variable with three electrophoretically distinct alleles; the protein products of ME-1 are reduced in activity relative to the protein products of ME-2. Joint segregation was examined between ME-1 and ten other biochemical loci in brook trout, and between ME-1, ME-2, and nine other biochemical loci in a splake—lake trout (Salvelinus namaycush) × brook trout hybrid—backcross. All pairwise examinations showed random assortment except ME-2 with an isocitrate dehydrogenase locus (IDH-3), which showed complete linkage in the splake backcross. This may be due to a chromosomal aberration.

    Notes: Abstract In this paper we report the results of an electrophoretic examination of genetic variation in the common meadow mushroom, Agaricus campestris. An examination of variability at 10 biochemical loci in a single natural population revealed an average heterozygosity of 0.281. Mendelian segregation was verified for 13 loci in single-spore-derived lines from individual sporophores. One linkage was found (Gpt-2 with Pep-GL-1, r = 0.065) among 47 pairwise examinations for joint segregation.

    Notes: Abstract Starch gel electrophoresis of allozymes extracted from mycelium grown in submerged culture was used to identify and catalog lines ofLentinula edodes. Variability at 11 multiallelic loci was used to separate 91 lines collected from worldwide sources into 35 genotypic clases. Genotypic class 2 was the largest, with 18 lines; 18 other genotypic classes contained more than one line; and 16 genotypic classes contained only one line each. Three hybrids, with potential commercial value, were produced by crossing single-spore-derived monokaryons of different genotypes. Each of the three hybrids could be differentiated from the above 35 classes based on their multilocus allelic combinations. Multilocus enzyme analysis should aid the description, registration, and licensing of improved shiitake cultivars.

    Notes: Abstract In second-generation sparctics (Salvelinus fontinalis × Salvelinus alpinus) backcrossed toS. fontinalis, we have identified tight classical linkage of phenotypic sex withLdh-1, Aat-5, andGpi-3. We designate this locusSex-1 and suggest that it may be the primary sex-determining locus in salmonids. Cumulative salmonid gene-to-centromere map distances for the three biochemical loci put the order as centromere—Ldh-1—(Aat-5 andGpi-3), with the latter two loci being tightly linked. An absence of association of phenotypic sex (presumably Sex-1) with these same three loci and other loci known to be linked to these loci is shown in splakes (S. fontinalis × Salvelinus namaycush) and cutbows (Salmo gairdneri × Salmo clarki). These data imply that the linkage ofSex-1 with these loci is found only inS. alpinus and support the view thatSex-1 lies across the centromere from these three loci inS. alpinus, representing a Robertsonian fusion not found in any of the other four species. A similar specific Robertsonian fusion is argued forS. gairdneri, whereSex-1 may be linked across a centromere to another biochemical locus (Ha). These linkage results and chromosomal observations of other investigators suggest thatSex-1 lies on an information-depauperate arm.
