A procedure involving O-phenylendiamine monohydrochloride as a reagent is recommended for oxygen isotope analysis. In 3 hours at 300° this reagent converts the oxygen of a variety of organic compounds quantitatively to water, which can be equilibrated with isotopically normal CO2 suitable for mass analysis. Compounds to which the method is applicable include alcohols, ethers, epoxides, aldehydes, ketones, acids, esters, lactones, amides. peptides and nitro compounds. The method is not applicable to simple phenols and phenol ethers.Carbonyl and alkoxyl oxygen-labelled carboxylic esters may be differentiated by reaction with aniline magnesium bromide prior to analysis, since anilide formation occurs without affecting the tracer content of the carbonyl group.
