    Notizen: Summary We report attempts to isolate and purify sialic acid-containing glycolipids (gangliosides) from etiolated hypocotyls of soybean (Glycine max) using methods developed for rat liver. The maximum amounts of ganglioside sialic acid present was found to be less than. 0.021 nmole/g fresh weight or less than 1:100,000 the amounts present in rat liver. We conclude that this tissue lacks gangliosides.

    Notizen: Summary Maize root tips were fixed in glutaraldehyde fixatives containing tannic acid and then processed for electron microscopy. Under these conditions, tannic acid selectively stained the contents of the Golgi apparatus secretory vesicles of some outer root cap cells, the cell walls of all cells, and substances in, and adjacent to, intercellular connections of mature primary walls and of secondary walls. Intercellular connections of the young primary walls were not stained. Plasma membranes, and substances associated with the outer leaflets of the plasma membranes, were also stained. Tannic acid-positive material was associated with the cell plate vesicles of forming walls but very little, or none, was associated with the Golgi apparatus vesicles of dividing cells.

    Notizen: Summary An ultrastructural study of the pollen tubes of Lilium and Clivia has demonstrated three different classes of longitudinal structures which could influence patterns of protoplasmic streaming and/or serve as “guide elements” in the vectorial migration of secretory vesicles: (a), cortical and noncortical microtubules; (b), microfilaments; and (c), subcortical tubules and cisternae of the endoplasmic reticulum (“subsurface cisternae”). Morphological details of these structures are described. Colchicine concentrations which lead to the complete disappearance of the microtubules affect neither germination of the pollen nor cytoplasmic streaming and tip growth of the elongating pollen tubes. Tip growth is initially uninhibited by cycloheximide, and cytoplasmic streaming is insensitive to this inhibitor. However, both of these processes are sensitive to cytochalasin B and vinblastine. Our results suggest that neither microtubules nor subsurface cisternae are essential for cytoplasmic streaming and directional secretion of cell surface materials in the pollen tube but would be consistent with an involvement of microfilamentous structures in these processes. Additionally, the possible importance of the lateral cross-link elements interconnecting all three types of structures is discussed.

    Notizen: Abstract. Cobalt chloride at a rate of 16 oz/acre when combined with the auxin herbicide triclopyr at 1 oz/acre synergistically enhanced the activity of the herbicide. A similar response was elicited by the addition of ammonium nitrate. Mg2+ or Mn2+ was antagonistic. The enhancement of activity by Co2+ or NH+ 4 appeared not to be the result of a simple enhancement of triclopyr uptake by the cations. Triclopyr uptake, as determined by gas chromatography/microwave-induced plasma/chemical reaction interface mass spectroscopy, was unaffected by the cation additions.

    Notizen: Abstract Retinoic acid inhibits the reduction of diferric transferrin through the transplasma membrane electron transport system on fetal rat liver cells infected with a temperature-sensitive SV40 virus when the cells are in the nontransformed state cultured at 40°C. When the cells are in the transformed state (grown at the permissive 33°C temperature), retinoic acid does not inhibit the diferric transferrin reduction. Inhibition of activity of nontransformed cells is specific for retinoic acid with only slight inhibition by retinol and retinyl acetate at higher concentrations. Isolated rat liver plasma membrane NADH diferric transferrin reductase is also inhibited by retinoic acid. The effect of transformation with SV40 virus to decrease susceptibility to retinoic acid inhibition stands in contrast to much greater adriamycin inhibition of diferric transferrin reduction in the transformed cells than in nontransformed cells.

    Notizen: Summary Methods are described for isolating and identifying subcellular membranes from walled hyphae ofGilbertella persicaria. Differences in thickness and symmetry of membranes and in contents of vesicles were used to distinguish different types of membranes. Mitochondria, vacuoles, plasma membrane, and vesicles with attached ribosomes from homogenized germlings equilibrated at the 1.2/1.4 M interface in discontinuous sucrose gradients. Accelerated flotation in centrifuged Ficol-sucrose gradients resulted in the additional separation of the mixed membranes into three fractions: one contained predominantly intact mitochondria, another was composed of vacuoles and vesicles coated with ribosomes, and a third was enriched in plasma membranes. Based upon morphometric analysis, these fractions contained 92% mitochondria, 53% vacuoles, and 89% plasma membranes, respectively. The source of vesicles coated with ribosomes was investigated since rapidly growing hyphae ofG. persicaria contained little rough endoplasmic reticulum as compared with other classes of membranes. Reconstruction from electron micrographs of mitochondrial fragmentation and vesiculation suggested that most of the ribosome-coated vesicles originated from disrupted mitochondria rather than from rough endoplasmic reticulum. The study demonstrates the utility of ultrastructural markers to identify membranesin vitro independent of, or as an adjunct to, cytochemical and biochemical markers.

    Notizen: Summary Epidermal and outer rootcap cells of maize root tips were treated with the sodium selective ionophore, monensin, and the ultrastructural changes were studied. In the presence of 10−5 to 10−3 M monensin, dictyosomes became distorted, cisternae separated from the stack, and secretory vesicles were released. Released secretory vesicles disappeard from the cytoplasm suggesting that their transport to, and fusion with, the plasma membrane was unaffected. Monensin did not inhibit cytoplasmic streaming of the outer rootcap cells. No new secretory vesicles were formed on the remaining dictyosomes or dictyosome fragments. In contrast to results with animal cells, swelling of plant dictyosome cisternae was observed only after fixation in glutaraldehyde-osmium tetroxide and not after fixation in potassium permanganate. Other cell components were not altered structurally by monensin. The effects of monensin on the Golgi apparatus were reversible, and dictyosomes were either repaired or new dictyosomes were formed after the root tips were removed from the monensin. Dictyosomes in epidermal cells reacted in the same manner as those in the rootcap except that numerous secretory vesicles remained in the cytoplasm, mostly in association with dictyosome fragments. Some secretory vesicles increased in size but no evidence of vesicle-vesicle fusion was noted. Cell plate formation was partially inhibited or blocked by monensin.

    Notizen: Summary Purified plasma membrane vesicles isolated both by aqueous twoPhase methods and by free-flow electrophoresis from homogenates prepared in the presence of 10 mM ascorbate, oxidized external NADH at rates of about 15 nanomoles/min/mg protein. The rate in the isolated vesicles was accelerated, without perceptible lag, 1.5-to 2-fold by 1 to 10 μM auxin (2,4-dichlorophenoxyacetic acid or indole-3-acetic acid). The reaction would be expected to result in acidification of the vesicle interiors and is proposed as a mechanism to account for auxin-induced acidification of cytoplasmin vitro.
