    Notes: Solid phase epitaxial growth of amorphized InP has been investigated at temperatures of 145–335 °C. The epitaxial recrystallization kinetics have been determined for both thermal and ion beam annealing using ion channeling and time-resolved reflectivity measurements and post-anneal, residual disorder has been characterized with transmission electron microscopy. The twin density and size distribution were significantly smaller for ion beam annealed samples, thus clearly demonstrating that ion beam annealing effectively suppresses the onset of highly defective (twinned) regrowth characteristic of thermal annealing. The substantially better crystalline quality of ion beam annealed samples contrasts with observations in GaAs where only a slight difference in post-anneal disorder was apparent for the two annealing regimes.

    Notes: The passage of the alkali metals Li, Na, K, Rb, and Cs through saturated phosphatidylcholine membranes has been measured using particle induced gamma ray and x-ray emission to observe the ions. Simultaneous measurements of these five cations has not been possible with more traditional methods involving ion specific electrodes or radioactive tracers. To the authors' knowledge this is the first time this technique has been used in lipid bilayer research. The ion leakage was determined for an incubation period of 30 min at several temperatures which spanned the bilayer melting temperature. The dependence of this permeability on temperature, acyl chain length, and ion size is described theoretically in terms of the density fluctuations in the bilayer. A leakage rate which depends on the mass of the ion was observed. This dependence shows that the heavier the ion the faster it diffuses out of the vesicle. The effect of this selectivity is shown to be most pronounced near the melting temperature of the lipid. Moreover, this size dependence suggests that the permeating entity is a hydrated ion, rather than a naked ion or a neutral, bound ion pair.

    Notes: Abstract. Positron emission tomography (PET) has been utilized to obtain dynamic images of long distance nutrient translocation in plants. Positron emitting 18F, produced by a Van de Graaff accelerator using the reaction 18O(p,n)18F, was fed in solution to excised stems of Glycine max positioned vertically in a large-aperture PET detector system. Images of tracer activity were recorded with a time resolution of 0.5 min and a spatial resolution of 4 mm. Maximum tracer activities at stem sites were obtained within 3 min of the pulse feed. A model is presented enabling evaluation of regional values for tracer flow, tracer binding, flow speed and flow volume. Analysis of data for one stem position yielded a flow volume of 2.1mm3 min−1 and a flow speed of 36cm min−1. Comparison with the distribution of 14C-inulin, which was simultaneously fed to the cut stems, indicates the 18F is suitable for use as an apoplastic tracer; 92% of the tracer activity accumulated in the leaves. The fraction of 18F that remained bound was most concentrated at stem nodal regions, an observation consistent with the existence of transfer cells at these sites. Advantages and limitations of PET applied to plant physiological investigations are discussed.

    Notes: Abstract Laser light scattering has been employed to determine the swimming speed distribution and the fraction of motile cells in samples of bovine spermatozoa. As predicted from theory, average trajectory velocities determined by laser light scattering were approximately four times the average translational speed estimated using light microscopy. The proportion of motile spermatozoa decreased with time at the same rate when samples were prepared in either HEPES or phosphate buffers. However, whereas the mean swimming velocity declined slowly in HEPES buffer, it dropped rapidly when phosphate buffer was used. Dilution (in the range 40–0.4×106 spermatozoa·ml-1) in either of these two buffers reduced the fraction of motile spermatozoa in the sample, but the mean swimming velocity of the remaining active spermatozoa was unchanged. Lowering the temperature from 37° C to 15° C reduced the mean swimming speed by a factor of 2–3 and the fraction of motile cells by a factor of 4–5.

    Notes: Abstract The design and construction of a laser light scattering spectrometer suitable for diffusion and flow measurements on biological materials is described. The type of quantitative information that can be obtained using this instrument in conjunction with photon correlation signal processing includes: the diffusion coefficient of a macromolecule; the average size and dispersity of a sample of subcellular particlesin vitro; the average velocity and velocity profile of streaming cytoplasmin vivo. The turntable unit and readily adjustable collection optics enable a variety of samples to be investigated at a range of scattering angles.

    Notes: A quasielastic light-scattering system has been constructed to study human fibrinogen. The first phase of the investigation was an attempt to clarify the shape of the firbinogen molecule using diffusion methods. The translational diffusion coefficient was measured as 2.04 ± 0.09 × 10-7 cm2 sec-1. Aggregation is suggested as the reason for a lower value previously obtained using this technique. Diffusion indicated the molecule was rigid and did not dissociate at low concentration, low ionic strength, or 37°C. Thermal denaturation was observed at 40°C. At 3°C, a second thermal instability was discovered.

    Notes: The shape of fibrin intermediate polymers as well as the rate of fibrinogen polymerization was studied using diffusion measured by quasielastic light scattering. After their length distribution was narrowed by gel filtration, the polymers yielded translational and rotational diffusion coefficients of 0.37 ± 0.05 × 10-7 cm2 sec-1 and 142 ± 32 sec-1, respectively. Theoretical considerations indicated the polymers to be rigid rods. The rate of polymerization of fibrinogen monitored by diffusion paralleled that provided by simulataneous intensity measurements. Both monitors indicated polymerization occurs most rapidly at 30°C.
