    Notes: Abstract: The protein kinase C (PKC) family is composed of at least four conventional (α, βI, βII, and γ) and several related novel (δ, ε, η, and ζ) isoforms with different distribution and sensitivity to Ca2+ and phorbol esters. The enzyme is known to be present in cerebral endothelial cells. We have investigated the occurrence of seven isoforms (α, β, γ, δ, ε, η, and ζ) by using reverse transcriptase-polymerase chain reaction in rat brain, in a freshly isolated brain microvessel fraction, in primary cultures of rat brain endothelial cells, in an immortalized rat brain endothelial cell line, and in aortic endothelial cell cultures. Brain tissue contained all seven investigated isoforms. A similar expression pattern was seen in freshly purified microvessels, but the PKC-γ isoform could not be detected. Primary cultures of endothelial cells expressed PKC-α, -β, -δ, -η, and -ε isoenzymes, whereas the immortalized cell line expressed PKC-α, -δ, -ε, and -η. The rat aortic endothelium contained only PKC-α and -δ isoforms. The variety of expression patterns of PKC family members in endothelial cells of different type may reflect differences in the functional responsiveness to environmental stimuli. Because PKC has been shown to be involved in the regulation of the blood-brain barrier permeability, the presence of different isoforms may confer a sophisticated intracellular regulatory mechanism to the brain endothelial cells.

    Notes: Periodic line structures with a period of 167 nm and linewidths varying from 35 to 100 nm have been produced on polyimide by direct ablation with a KrF laser using an interferometric technique. Since ablation is a nonlinear process, the resolution can exceed that expected from the wavelength and numerical aperture of the system and the linewidth can be controlled by varying the laser fluence. This externally generated period of 167 nm prevents the spontaneous growth of periodic surface structures due to radiation remnants.

    Notes: An ultrahigh-brightness ArF excimer laser system is described that is capable of generating pulse energies of 60 mJ with a pulse duration of ∼700 fs. The system utilizes a newly developed seed pulse generation scheme based on spectrally compensated sum-frequency mixing in beta-barium metaborate (BBO), and a double-pass discharge pumped ArF excimer preamplifier followed by an electron beam pumped power amplifier.

    Notes: A case of Waldenström's macroglobulinaemia presenting with non-specific dermatological symptoms (cold urticaria and cold purpura) is described in a 37-year-old female patient. Progressive telangiectasia and spider naevi were also observed. Involvement of the skin may be explained by the presence of macroglobulins with cryoprotein-like qualities. This facilitated an early diagnosis.

    Notes: In this paper we have examined the possibility of identifying those membrane structural variables (polar head groups and the nature of hydrocarbon tails) that modulate membrane ionic permeability. Altering the bilayer lipid composition produces variations in physical parameters (surface potential, partition coefficient, and mobility) governing the conductance mediated by neutral carriers of anions and cations. Specifically, the effects of the charged polar head groups are shown to be understandable in terms of the surface potential they produce through the formation of a diffuse double layer, whereas the effects of the viscosity may be demonstrated by “freezing” the membrane. The effects of membrane composition on membrane conductance are illustrated by a third, less well understood, example of how cholesterol alters bilayer conductances. The results indicate the possibility of using positive and negative permeant species as probes of membrane structures.

    Notes: A series of overlapping clones coding for L-glutamic acid decarboxylase was purified from a mouse brain cDNA library, the longest of which contains a 1869 bp open reading frame and 913 bp of non-coding sequence. By comparison with the corresponding sequences from the mouse genome, it was determined that the first methionine in the longest cDNA represents the initiation codon. Expression of this cDNA in eukaryotic cells produces a 62 kd protein that is recognized by antiserum against rat GAD and which displays GAD activity commensurate with the amount of protein produced. Antibodies raised against the purified product of this cDNA recognize a 62 kd protein from mouse brain on immunoblots, specifically stain GABA-ergic neurons in brain sections, and are capable of immunoprecipitating most GAD activity from mouse brain extracts. These results provide the first definitive identification of a cDNA coding for the larger of two forms of GAD in mouse brain, and suggest that the two forms are closely related.

    Notes: Olfactory bulb interneurons are continuously generated throughout development and in adulthood. These neurons are born in the subventricular zone (SVZ) and migrate along the rostral migratory stream into the olfactory bulb where they differentiate into local interneurons. To investigate the differentiation of GABAergic interneurons of the olfactory bulb we used a transgenic mouse which expresses green fluorescent protein (GFP) under the control of the glutamic acid decarboxylase 65 kDa (GAD65) promoter. During development and in adulthood GFP was expressed by cells in the SVZ and along the entire length of its rostral extension including the distal portion within the olfactory bulb. The occurrence of GAD65 mRNA in these zones was confirmed by PCR analysis on microdissected regions along the pathway. Polysialic acid neural cell adhesion molecule, a marker of migrating neuroblasts in adults, was coexpressed by the majority of the GFP-positive SVZ-derived progenitor cells. Cell tracer injections into the SVZ indicated that ≈ 26% of migrating progenitor cells expressed GFP. These data show the early differentiation of migrating SVZ-derived progenitors into a GAD65–GFP-positive phenotype. These cells could represent a restricted lineage giving rise to GAD65-positive GABAergic olfactory bulb interneurons.
