Summary The cytopathogenic properties of 13 freshly isolated herpes simplex virus strains were studied in rabbit kidney and HeLa cell cultures. In rabbit kidney cells the only reaction was at first a rounding-off of single cells (EZ). From the 4th to 9th homologous passage onward all virus strains showed an additional giant cell type (RZ) of the cytopathic effect. We are here concerned with a mutant of the EZ type and not with the result of a selection from a mixture of both types, because virus from a single EZ plaque, which had been cloned in the presence of homotypic antibody, gave again rise to giant cells, when it was propagated vigorously. The mutation rate of the “Schott” strain was 2.2×10−4±0.35. In HeLa cultures and in HeLa clones, however, all virus strains showed only the EZ type during 11 homologous passages. Though the RZ variant had also arisen, it could be demonstrated in rabbit kidney cells only. By passages with end point dilutions in rabbit kidney cells it was possible to isolate a pure RZ type, which, however, produced only EZ plaques in HeLa cultures and clones. This type remained stable during 5 continuous HeLa passages and subsequently produced only giant cells in rabbit kidney cultures. These results tend to show that the kind of cytopathic effect depends on the genotype of the infecting virus, but is modified in its phenotypic expression by the host cells.

Zusammenfassung Es wurden 13 frisch isolierte Herpes-simplex-Stämme auf ihr zytopathisches Verhalten in Kaninchennieren- und HeLa-Zellkulturen untersucht. Bei Kaninchennieren-Zellen waren zunächst als einzige Reaktion nur Einzelzellabkugelungen (EZ) zu beobachten. Von der 4.–9. homologen Passage an kam bei allen Virusstämmen noch ein Riesenzell-Typ (RZ) hinzu. Hierbei handelte es sich um eine Mutante des EZ-Typs und nicht um das Ergebnis einer Selektion aus einer Mischung beider Typen im Ausgangsmaterial. Das Virus eines einzelnen EZ-Herdes — in Gegenwart von homotypischen Antikörpern kloniert — bildete bei intensiver Vermehrung wieder Riesenzellen, deren Mutationsrate bei dem Stamm „Schott“ 2,2×10−4±0,35 betrug. Auf HeLa-Zellkulturen und in HeLa-Zellklonen führten dagegen alle Virusstämme im Verlauf von elf homologen Passagen nur zum EZ-Typ. Die RZ-Variante war zwar ebenfalls entstanden, jedoch nur auf KN-Zellen nachweisbar. Durch Endverdünnungspassagen konnte auf KN-Zellen ein reiner RZ-Typ isoliert werden, der sich auf HeLa-Zellen und -Klonen nur als EZ-Typ auswirkte. Er blieb während fünf fortlaufender HeLa-Passagen stabil und bildete anschließend auf KN-Zellen wieder ausschließlich Riesenzellen. Diese Ergebnisse sprechen dafür, daß die Art des zytopathischen Effektes vom Genotyp des infizierenden Virus bestimmt wird, die phaenotypische Ausprägung jedoch von der Wirtszellart abhängig ist.
