Notes: We have previously distributed a software package, SIMLA (SIMulation of Linkage and Association), which can be used to generate disease phenotype and marker genotype data in three-generational pedigrees of user-specified structure. To our knowledge, SIMLA is the only publicly available program that can simulate variable levels of both linkage (recombination) and linkage disequilibrium (LD) between marker and disease loci in general pedigrees. While the previous SIMLA version provided flexibility in choosing many parameters relevant for linkage and association mapping of complex human diseases, it did not allow for the segregation of more than one disease locus in a given pedigree and did not incorporate environmental covariates possibly interacting with disease susceptibility genes. Here, we present an extension of the simulation algorithm characterized by a much more general penetrance function, which allows for the joint action of up to two genes and up to two environmental covariates in the simulated pedigrees, with all possible multiplicative interaction effects between them. This makes the program even more useful for comparing the performance of different linkage and association analysis methods applied to complex human phenotypes. SIMLA can assist investigators in planning and designing a variety of linkage and association studies, and can help interpret results of real data analyses by comparing them to results obtained under a user-controlled data generation mechanism.A free download of the SIMLA package is available at http://wwwchg.duhs.duke.edu/software.

Notes: Candida dubliniensis is a phylogenetically closely related species to Candida albicans. So far virtually nothing is known about the virulence factors of C. dubliniensis. Cell surface hydrophobicity (CSH) plays a critical role in adhesion of microorganisms to phagocytic cells; hydrophobic cells of C. albicans have been reported to be less sensitive to phagocytic killing than hydrophilic cells. C. dubliniensis displays CSH at 37°C in contrast to C. albicans. To elucidate this issue, we determined levels of phagocytosis, oxidative burst and killing by human neutrophils of C. dubliniensis (n=10) compared to C. albicans (n=10) both cultured at 37°C. Obtained test results revealed no statistically significant differences between these two yeast species for the level of phagocytosis (77.3 vs. 76.2% after 60 min), evoked oxidative burst (64.5 vs. 67.3% after 30 min) and killing (72.7 vs. 73.1% after 240 min). Therefore, human neutrophils can be considered to be equally efficient against these two yeast species.

Description / Table of Contents: Summary Attachment theory, originally developed by John Bowlby, becomes increasingly relevant in the field of psychotherapy (research). This review starts with a definition of central concepts and hypotheses of this theory and summarizes results regarding the development as well as correlates of attachment, mainly from research in developmental psychology. Related to adults, attachment theory has to be reconceptualized which is done in another part of this paper. Finally, some common methods for the assessment of adult attachment are described which play an important role in clinical studies (these will be the content of the part 2 of this review).

Description / Table of Contents: Summary Whilst the first part of the review about the relevance of attachment theory covered the basic principles and methods of attachment research, this second part deals with possible implications of the theory within the field of psychotherapy and psychosomatics. Consideration of its general clinical relevance and of the relationship between attachment theory and psychoanalysis forms the basis for a description of the usefulness results of attachment research might have in psychotherapeutic practice. Summaries of studies dealing with the importance of attachment characteristics for the development of psychosomatic disorders and for coping with illness and stress and studies combining attachment styles with psychopathology will follow. The last part of the review refers to the importance of the theory for psychotherapy and psychotherapy research.
