    Notes: We have systematically investigated the influence of the three parameters (temperature of sintering, lead content, and duration of the sintering) on the growth of the (2:2:2:3) phase in ceramics of nominal composition Bi2−xPbxSr2Ca2Cu3O10−x/2, and have determined the compositions and thermal conditions leading to ceramics with Tc≈110 K and containing more than 95% of this phase. We show that this result can be obtained by usual calcination near 880°C in air, without recurring to the introduction of excess in the constituants, providing that intermediate grinding are effected, in order to reactivate the formation of the (2:2:2:3) phase which otherwise saturates for long durations of sintering. The suitable nominal compositions (0.3≤x≤0.4) correspond to those that yield inside the grains a lead content in the vicinity of the limit of solubility for lead. From the results of the preceding investigations and of experiments effected on lead-substituted single crystals of the (2:2:1:2) phase, we have clarified the role played by lead, at the microscopic level, in the enhancement of the formation of the (2:2:2:3) phase. The inferred role is the establishment of strong bonding between the slabs which are the buildings blocks of the structure of the superconducting phases, these slabs being almost independent in the lead-free compounds. The origin of the stronger bonding is discussed.

    Notes: We report the engineering of sets of mesas on lead-substituted single crystals of Bi2Sr2CaCu2Oy. Each mesa behaves as a series array of intrinsic Josephson junctions with, up to 70 K, "two states'' hysteretic I(V) characteristics, free of branching, whose RnIc products can be tuned through geometrical patterning and adjustment of the lead contents. RnIc values as high as 550 mV at 4.2 K have been reached. The tuning through lead substitution is attributed to the decrease of the anisotropy with increasing lead content.

    Notes: Abstract— A study was undertaken of the glycosphingolipids in cell cultures derived from cerebellum of Tay-Sachs disease fetal brain in order to determine the suitability of such cell strains as a model for Tay-Sachs disease. The glycosphingolipids in the Tay-Sachs disease cultured cerebellar cells were compared with those found in normal cultured cerebellar cells, normal and Tay-Sachs cultured lung cells, and normal and Tay-Sachs fetal brain. The glycolipids were separated by TLC, then analyzed by GLC of the trimethylsilyi derivatives of the methylglycosides of the sugar moieties. In the cultured cerebellar lines, the predominant gangliosides were GM2, GM3, and GD3. There was a 4-fold increase of GM2 in the Tay-Sachs as compared with the normal line. Only GM3 and GD3 gangliosides were found in the Tay-Sachs and the normal fetal lung cell cultures. The major neutral glycosphingolipids in all of the cultured cells which were analyzed were glucosylceramide, lactosylceramide, digalactosyl-glucosylceramide, and globoside. When the Tay-Sachs cerebellar cells were labelled with [1-14C]gluco-samine, some radioactivity was observed in the trihexosylceramide band, indicating the presence of a small amount of a galactosamine-containing trihexosylceramide which may be asialo-GM2 (GA2). The trihexosylceramide in Tay-Sachs fetal brain was identified as GA2 by GLC. Both Tay-Sachs and normal fetal brain gangliosides were more complex than those found in the cultured cells. Long chain fatty acids (C24:0 and C24;1) predominated in all of the glycosphingolipids of the Tay-Sachs and the normal cultured cerebellar cells. In contrast, the glycosphingolipids of Tay-Sachs and normal fetal brain contained mainly the shorter chain fatty acids (C16:0, C18:0, and C18:1). The cerebrosides in both the Tay-Sachs and normal fetal brains were mainly glucosylceramide with only small amounts of the galactosylceramide which predominates in infant brain.     Notes: —Quantitative analyses performed on the lipids of cerebral grey matter from brains of a normal child and a child with Tay-Sachs (T-S) disease were compared with such analyses on the brain of a 6-year-old, non-Jewish male with systemic GM2-gangliosidosis of a late infantile type (GM2-LI). Analysis of gangliosides showed a 3·5-fold increase of total gangliosides in the GM2-LI brain and a six-fold increase in the T-S brain, compared to normal brain. Both pathological brains had similar distribution patterns for gangliosides, with the GM2-ganglioside component constituting more than 80 per cent of the total. Lipid components in the T-S brain were below normal values except for lecithin and cholesterol, while in the GM2-LI brain there were increases in sulphatides, cerebrosides, sphingomyelin and cholesterol. Approximately twice as much ceramide trihexoside was present in the T-S brain as in the GM2-LI brain, and none could be detected in the normal brain. The clinical, pathological and biochemical data support the conclusion that this case represents a new variant of systemic late-infantile gangliosidosis in which there is an accumulation of the GM2-ganglioside like that in Tay-Sachs disease.

    Notes: Habituation of the alpha-blocking component of the orienting response (OR) was evaluated in depressed patients and normals. The method of feedback electroencephalography was used to elicit and control the OR.It was predicted that the depressed patients would show reduced initial ORs to visual stimuli compared to normals. The response of the patients to emotional verbal stimuli (related to their conflicts) was predicted to be more variable than the response to non-conflict words, compared to normals.The results indicated that depressed patients are less responsive than normals for the initial few presentations of visual stimuli. The hypothesis about greater response variability by depressives to emotionally significant words was partially confirmed, i.e., the response variability was increased for one of the two verbal stimuli. The study demonstrated that feedback EEG experiments and clinical assessments can be reliably carried out in a non-laboratory setting.

    Notes: The effects of waterfall-induced turbulence on the dissolved oxygen concentration of the Passaic River (New Jersey) at two mostly natural waterfalls were observed during a five-month period encompassing high to low temperature and flow. A parameter defined by past investigators as the “deficit ratio” was found to be useful for mathematically describing the results. The Great Falls system showed the deficit ratio to be an inverse function of temperature for all events, with good correlation. A relationship was developed for the Little Falls system under low flow showing the deficit ratio to likewise be an inverse function of temperature and a power function of flow.

    Notes: H-2Kb and HLA-A2 are MHC4 class I molecules with a similar overall structure. Important differences between these two class I molecules reside in the structure of the individual pockets in the antigenic-peptide-binding groove. H-2Kb, which has a deep C pocket, binds specifically peptides with a tyrosine or a phenylalanine at position 5. In contrast, HLA-A2 has a shallow C pocket, which cannot accommodate large side chains at position 5. Site-directed mutagenesis was used to generate a chimera between the murine H-2Kb and the human HLA-A2 [H-2Kb/HLA-A2(C′)]. The structure of this chimera is similar to H-2Kb except for the region around the deep C pocket, where residues at positions 9, 97 and 99 were substituted with those bulkier residues from HLA-A2. Peptide binding between this chimera and H-2Kb-binding peptides [VSV (52–59), OVA (257–264), and MCMV pp89 (168–176)], revealed that the deep C pocket of H-2Kb was crucial for high-affinity binding. While a peptide, VSV (52–59), was found to bind with severalfold lower ‘affinity’ to H-2Kb/HLA-A2(C′) than to the wild-type H-2Kb, a VSV analogue with the tyrosine in position 5 (Tyr5) substituted with an alanine was found to bind with a similar ‘affinity’ to both MHC class I molecules. Computer-aided modelling of the H-2Kb/HLA-A2(C′) complex indicates that the VSV (52–59) peptide probably binds to the chimeric MHC molecule with the peptide side chain of anchor residue Tyr5 pointing away from the groove. These results confirm a role of the individual pockets in determining peptide-binding affinity and specificity and suggest that this may be accomplished by changes in side-chain orientation.
