    Notes: : Bacterial populations in nontreated and herbicide-treated waters were subjected to three different herbicides. Diuron, dichlobenil, and diquat were added (100 mg/l) to water samples from two fresh water lakes and two herbicide-treated ponds. Total numbers of bacteria were monitored. Bacterial populations in fresh lake water decreased after herbicide additions; however, final populations were significantly greater than the controls. Similar observations were recorded for bacteria in dichlobenil- and diuron-treated waters. Selective enrichment is probably expressed in these experiments.

    Notes: Abstract An adaptation of the d-ribulose-1,5-bisphosphate carboxylase (RuBPCase) activity to changing CO2 concentrations in the growth medium in the chemostat was observed in the obligate chemolithotroph Thiobacillus neapolitanus. RuBPCase activity has been separated in a soluble and particulate fraction. The activity of the particulate fraction appeared to be associated with the carboxysomes. The total activity of RuBPCase of CO2 limited cultures was about 5-fold higher than the activity of thiosulphate limited cultures grown in the presence of 5% CO2 whilst the particulate activity and the soluble activity were about 8- and 1.5-fold higher, respectively. The fluctuation of the total and particulate RuBPCase activity correlated with the changes in volume density of carboxysomes in the cell. An inverse correlation between maximal CO2 fixing capacity by whole cells and the volume density of carboxysomes was observed. The change in ratio of soluble RuBPCase activity to particulate RuBPCase activity paralleled the change in maximal CO2 fixation by whole cells during the different growth conditions.

    Notes: Abstract The carboxysomal polypeptides of Thiobacillus neapolitanus with apparent molecular masses of 85 and 130 kDa were isolated and subjected to N-terminal sequencing. The first 17 amino acids of the two peptides were identical. The sequence perfectly matched the deduced amino acid sequence of an open reading frame in the carboxysome operon. The gene was subsequently named csoS2. Expression of the gene in Escherichia coli resulted in the production of two peptides with apparent molecular masses of 85 and 130 kDa. Immunospecific antibodies generated against the smaller peptide recognized both peptides; the peptides were named CsoS2A and CsoS2B, respectively. A digoxigenin-hydrazide glycosylation assay revealed that both CsoS2A and CsoS2B are post-translationally modified by glycosylation. CsoS2 was localized to the edges of purified carboxysomes by immunogold electron microscopy using the monospecific CsoS2A antibodies. The molecular mass of CsoS2A calculated from the nucleotide sequence was 92.3 kDa.

    Notes: Abstract A homogenous preparation of carboxysomes was isolated from Thiobacillus neapolitanus by means of density gradient centrifugation and preparative electrophoresis through agarose. Analysis of the carboxysomes by denaturing sodium dodecyl sulfate polyacrylamide gel electrophoresis (SDS-PAGE) revealed the presence of 12–15 polypeptides. Approximately 62% of the total protein was found to consist of the large and small subunits of ribulose-1,5-bisphosphate carboxylase-oxygenase (RubisCO). Two polypeptides were found to be components of the carboxysome shell. Ribose-5-phosphate isomerase, ribulose-5-phosphate kinase, and fructose-1,6-bisphosphatase could not be detected in the carboxysome. Purified carboxysomal and cytoplasmic RubisCO were shown to be similar if not identical by several criteria including specific activity, carboxylase/oxygenase activity, and electrophoretic mobility.

    Notes: Abstract To date only marine strains of the genus Beggiatoa have been proven to have the capacity for lithoautotrophic growth. The two best characterized freshwater strains (B18LD and OH-75-2a) — previously tested with no evidence of autotrophic potential — were reinvestigated in the current study. Heterotrophically grown cells from exponential or early stationary phase showed extremely low ribulose-1,5-bisphosphate carboxylase/oxygenase (RuBisCO) activities (0.002–0.2 nmol CO2·mg prot-1 min-1). After acetate limitation terminated growth both strains demonstrated increased RuBisCO activity with a maximum achieved 1–3 days into stationary phase. For a single growth cycle, activity could increase by more than 100-fold (strain OH-75-2a), but the highest activity achieved was less than 5% of that observed in a lithoautotrophic control. Phosphoribulokinase activity was approximately an order of magnitude greater than the corresponding RuBisCO activity and showed a similar pattern of regulation. A gene probe derived from the RuBisCO large subunit gene of Anacystis nidulans hybridized strongly with DNA from strain OH-75-2a and the autotrophic marine control but not with DNA from strain B18LD.
