    Notizen: The LAN-1 clone, a cell line derived from a human neuroblastoma, possesses muscarinic receptors. The stimulation of these receptors with increasing concentrations of carbachol (CCh; 1 -1,000 μM) caused a dose-dependent increase of the intracellular free Ca2+ concentration ([Ca2+]i). This increase was characterized by an early peak phase (10 s) and a late plateau phase. The removal of extracellular Ca2+ reduced the magnitude of the peak phase to ∼70% but completely abolished the plateau phase. The muscarinic-activated Ca2+ channel was gadolinium (Gd3+) blockable and nimodipine and ω-conotoxin insensitive. In addition, membrane depolarization did not cause any increase in [Ca2+]i. The CCh-induced [Ca2+]i elevation was concentration-dependently inhibited by pirenzepine and 4-diphenylacetoxy-N-methylpiperidine methiodide, two rather selective antagonists of M1 and M3 muscarinic receptor subtypes, respectively, whereas methoctramine, an M2 antagonist, was ineffective. The coupling of M1 and M3 receptor activation with [Ca2+]i elevation does not seem to be mediated by a pertussis toxin-sensitive guanine nucleotide-binding protein or by the diacylglycerol-protein kinase C system. The mobilization of [Ca2+]i elicited by M1 and M3 muscarinic receptor stimulation seems to be dependent on an inositol trisphosphate-sensitive intracellular store. In addition, ryanodine did not prevent CCh-induced [Ca2+]i mobilization, and, finally, LAN-1 cells appear to lack caffeine-sensitive Ca2+ stores, because the methylxanthine was unable to elicit intracellular Ca2+ mobilization, under basal conditions, after a subthreshold concentration of CCh (0.3 μM), or after thapsigargin.

    Notizen: The water relations of two durum wheat cultivars (Triticum durum Desf.) were studied throughout the growing season. Irrigated and unirrigated plants were compared from booting to milk stage; a period where water stress occurred naturally in the field. Modulus of elasticity (ε), turgid weight/dry weight ratio (TW/DW), relative water content at zero turgor (RWCo) and osmotic potential at full turgor (ε) declined throughout the season while average turgor (ψp) increased. Water stress induced a further decrease in ψπ100 and the TW/DW ratio. The elastic modulus varied greatly. During the first stages of growth, cv. Appulo (the more resistant cultivar) showed lower ε values than cv. Valforte. At the milk stage, ε was lower for the unirrigated than the irrigated plants. Correlation coefficients between the TW/DW ratio and the osmotic potential were significant for both cultivars. In cv. Valforte, TW/DW was also correlated with the average turgor and the bulk modulus of elasticity. Structural changes that affect the TW/DW ratio seem to be important factors influencing water relations and drought tolerance in durum wheat.

    Notizen: (Pro-Pro-Gly)10 is one of the most widely studied collagen polypeptide models. Microgravity crystal growth of (Pro-Pro-Gly)10 was carried out in the Advanced Protein Crystallization Facility aboard the Space Shuttle Discovery during the STS-95 mission. Crystals were successfully grown in all experiments, using both dialysis and free-interface diffusion methods. The quality of the microgravity-grown crystals and of ground-grown counterparts was assessed by X-ray synchrotron diffraction. Microgravity-grown crystals exhibited a significant improvement in terms of dimensions and resolution limit. As previously reported, crystals were orthorhombic, space group P212121. However, the diffraction pattern showed weak reflections, never previously measured, that were consistent with new unit-cell parameters a = 26.9, b = 26.4, c = 182.5 Å. This allowed the derivation of a new model for the arrangement of the triple-helical molecules in the crystals.

    Notizen: Crystals of alcohol dehydrogenase from Sulfolobus solfataricus were grown in the Advanced Protein Crystallization Facility during the Life and Microgravity Sciences Spacelab mission on the US Space Shuttle. Large diffracting crystals were obtained by dialysis, whereas only poor-quality crystals were obtained by vapour diffusion. The quality of both the microgravity and ground-based crystals was analysed by X-ray diffraction. There was some improvement in terms of size and diffraction resolution limit for the microgravity crystals. However, the twinning observed in the Earth-grown crystals was also present for those grown in microgravity.

    Notizen: The Advanced Protein Crystallization Facility is widely used by European and US teams to study protein crystal growth under microgravity conditions. This paper describes the laboratory equipment developed to observe the crystals, by means of a charge-coupled-device camera, during the growth process on the ground inside the hanging-drop reactors of the Advanced Protein Crystallization Facility. The apparatus was used during ground tests performed to prepare the microgravity experimentation, allowing a time reduction in determining the crystallization conditions to be used for the microgravity experiment.

    Notizen: Abstract During the last 13 years 110 children with duplex ureteroceles were treated. There was a 3 to 1 female-to-male predominance. The authors report their experience with a group of 25 patients with duplex ureteroceles diagnosed in utero. Endoscopic incision of the ureterocele was performed as the initial treatment in 18 (72%) of these patients; decompression of the upper pole of the ureterocele was achieved in 100% of cases. Preservation of the upper pole was possible in 8 patients (44%); vesicoureteral reflux was created at the level of the upper pole in 8 (44%). Reconstruction of the lower urinary tract was accomplished in 15 (83.3%) patients who had had endoscopic treatment. Our experience has led us to believe that endoscopic incision of the ureterocele deserves reconsideration and re-evaluation because, as our series has shown, this intervention offers the great advantage of allowing good functional recuperation of the obstructed moiety with a high rate of success in a totally asymptomatic group of patients diagnosed in utero.

    Beschreibung / Inhaltsverzeichnis: Sommario L'incorporazione di [14 C] colina, [14 C] etanolamina e [14 C] serina nei loro corrispondenti fosfolipidi è stata studiata in presenza o in assenza di TPA nella linea cellulare di neuroblastoma umano LAN-2. La presenza di TPA incrementava la radioattività incorporata nei fosfolipidi con una corrispondente diminuzione della radioattività nel compartimento citosolico. Non c'erano differenze tra le cellule esposte al TPA e quelle di controllo nella distribuzione della radioattività nella colina libera, nella fosforilcolina o nella CDP colina. Ciò indica che l'aumento di incorporazione del precursore marcato nei lipidi non è accompagnato da incremento di incorporazione nei composti intermedi della sinteside novo. I nostri risultati suggeriscono che una colina baseexchange enzyme potrebbe essere stimolata nelle cellule esposte al TPA. Inoltre l'aumentata incorporazione di serina, prodotta dal TPA, nel suo corrispondente fosfolipide implica la stimolazione del serinabase exchange enzyme, che è il responsabile della sintesi di fosfatidilserina nei mammiferi. Queste osservazioni sono suggestive di un più generale coinvolgimento deibase exchange enzymes nella modulazione dei fosfolipidi di membrana.
    Notizen: Abstract The incorporation of [14 C] choline, [14 C] ethanolamine, and [14 C] serine by LAN-2 cells into their corresponding phospholipids was investigated in the presence or absence of TPA. The presence of TPA increased the amount of radioactivity incorporated into the phospholipids with a corresponding decrease in the amount of radioactivity in the cytosolic compartment compared to control cultures. There were no differences between TPA-exposed and control cells in the distribution of radioactivity in free choline, phosphorylcholine or CDP-choline of [14 C] choline labeled cells. This indicates that the increased lipid labeling was not accompanied by enhanced labeling of the intermediates of thede novo pathway. These results suggest that a choline base exchange enzyme was stimulated in TPA exposed cells. In addition, the enhanced incorporation of serine by TPA into its corresponding phospholipid implies the stimulation of the serine base exchange enzyme which is responsible for phosphatidylserine synthesis in mammals. These observations suggest a more general involvement of the base exchange enzymes in the modulation of membrane phospholipids.

    Notizen: Abstract A form of CAT-like activity was found bound present in rat brain synaptosomal membranes which could be recovered in the Triton X-114 phase. The enzyme activity was slightly activated by NaCl, had a pH maximum around 8 and showed a temperature dependence with a Q10 of 2.28. It was inhibited 100% by 10−6 M naphthyl vinyl pyridinium but not by 10−5 M diisopropyl phosphofluoridate. The kinetics of this bound form of CAT were similar to the soluble form of the enzyme. TheK m was 405±58 μM for choline and 62±8 μM for AcCoA. Five isoelectric forms were found with pH's of 4.55, 6.05, 7.06, 7.36, and 8.00 which is in contrast to the three isoelectric forms found of the soluble enzyme in rat brain. The presence of a CAT-like activity in the plasma membrane was confirmed with experiments performed using intact synaptosomes and intact cells in culture. Acetylcholine, synthesized from radioactive AcCoA by intact rat brain synaptosomes, was recovered in the incubation medium and only in the presence of exogenous choline or when the production of choline was stimulated by oleate via the activation of phospholipase D. This was also seen in experiments with intact pheochromocytoma cell cultures (PC 12) which synthesize acetylcholine that was recoverved in the incubation medium. Acetylcholine formation in the presence of choline and AcCoA was stimulated in cells that had been grown in the presence of nerve growth factor (NGF). The localization of 1% of CAT activity in a transbilayer position in the plasma membrane, could suggest a possible role of this enzymatic form in the regulation of acetylcholine synthesis.
