    Notes: Abstract Intraperitoneal administration of tuftsin-M [Thr−Lys−Pro−Arg−NH−(CH2)2−NH−CO−C15H31] to Balb/C mice has been shown to induce a respiratory burst in the peritoneal exudate cells. The macrophages exhibited enhanced levels of O2 −, H2O2, NADPH oxidase and myeloperoxidase, but the activities of superoxide dismutase, catalase and glutathione peroxidase remained virtually unchanged. The magnitude of the oxidative burst depended directly on the dose of tuftsin-M; higher activity was observed at higher doses of the peptide. Tuftsin-M enhanced the generation of both O 2 − and H2O2 under in vitro conditions, as did phorbol myristate acetate. These results suggest that tuftsin-M could enhance non-specific defence against infections by activating the macrophages.

    Notes: Abstract Ascaridia galli, using 2-oxoglutarate as an acceptor, transaminates alanine and aspartate at significantly high rates. Among other amino acids valine, phenylalanine, leucine, isoleucine, arginine, tyrosine and methionine are metabolised at moderate rates while lysine, serine, threonine, cysteine, glycine, histidine, tryptophan, DOPA and GABA appear to be inert in this respect. Body parts mimic the whole worm in the pattern of transamination of various amino acids with the exception of methionine. alanine, aspartate and glutamate may transfer their amino group also to pyruvate and oxaloacetate. Alanine and aspartate: 2-oxoglutarate transaminases are located mainly in the cytosol and mitochondrial fractions.

    Notes: Abstract With [U-14C]glucose we confirmed that essentially all of the glucose carbon used aerobically by intactBrugia pahangi adults was accounted for in the recovered lactate, in spite of the presence of cristate mitochondria in these parasites. Approximately 0.2% of the glucose carbon that disappeared was recovered in CO2. However, aerobiosis was required for the incorporation of alanine, proline, and glutamate into both respiratory CO2 and protein, but the glucose was used on the order of 1000 times faster than the amino acids. Homogenate fractions were examined for amino acid use, and all except the 100 000g soluble fraction showed activity. However, all of the active fractions also contained living microfilariae and other developmental stages that arise from the adult uterus, since the females give birth to living larvae. Sonicates of the microfilariae formed CO2 from proline 30 times more rapidly than crude homogenates of the adult. Similarly, CO2 was formed from glucose 14 times more rapidly by intact microfilariae than by intact adults. The possibility arises that the low rates of CO2 formation from both the amino acids and glucose by intact adults may arise from the metabolism of the aerobic microfilariae and other developmental stages present in utero.

    Notes: We have previously reported that Ca2+ influx into the mitochondria of Hymenolepis diminuta, a rat intestinal eestode, takes place through an electrophoretic uniport system. Sodium and lithium were found to induce efflux of 45Ca2+ from the mitochondria of H. diminuta. The two cations induced the efflux in a hyperbolic and linear fashion, respectively. The efflux as well as an exchange of external Ca2+ with internal 45Ca2+ was inhibited by lanthanum. The type of Ca2+ transport system in the cestode organelle has been discussed and compared with that of the host (mammalian) counterpart.
