    Notes: The influenza virus haemagglutinin has an important role in the infectious cycle of the virus and carries multiple B and T cell epitopes. It is synthesized as a single polypeptide chain but viral infectivity depends on its post-translational enzymatic cleavage. The cleavage site of a trypsin-like enzyme responsible for this modification is found in the most conserved intersubunit region of the molecule. In this study the role of this region in antibody recognition was investigated. Synthetic peptides comprising the intact and cleaved forms of the intersubunit segment were used to examine the specificity of virus- or peptide- induced antibodies. The immune response elicited by viral infection resulted in the appearance of antibodies capable of neutralizing the virus without interfering with its binding to the receptor. A monoclonal antibody (MoAb) of such functional properties was shown to recognize the intact intersubunit region both in the uncleaved haemagglutinin molecule and in a 25-mer synthetic peptide comprising the intact intersubunit region. Specificity and functional studies revealed the conformation dependent recognition of the C-terminal segment of the haemagglutinin 1 subunit by this MoAb. The binding of the antibody was shown to inhibit the trypsin-mediated cleavage of the haemagglutinin molecule and the membrane fusion event. The enzymatic cleavage of the haemagglutinin was demonstrated to abolish antibody recognition of the infective virus suggesting an escape mechanism mediated by the functional destruction of this highly conserved region. The synthetic peptide corresponding to the intact intersubunit region is characterized by an ordered structure and is able to elicit an antibody response in BALB/c mice while its subfragments are nonimmunogenic. Furthermore, this peptide elicited a protective immune response demonstrated by in vivo experiments.

    Notes: The functional role of VR1, which we and others have recently identified on several epithelial and mesenchymal human skin cell populations, was investigated in the human hair follicle (HF), as a prototypic epithelial–mesenchymal interaction system. VR1 immunoreactivity was confined to distinct epithelial compartments of HFs in anagen and catagen, while dermal papilla fibroblasts and HF melanocytes were VR1 negative. In organ culture, VR1 activation by capsaicin resulted in a dose-dependent and VR1-specific inhibition of hair shaft elongation, suppression of proliferation, promotion of apoptosis, and induction of catagen transformation, possibly due to upregulation of a potent hair growth inhibitor TGFβ2. Cultured outer root sheath (ORS), as well as HaCaT, keratinocytes also expressed functional VR1, whose stimulation inhibited proliferation, induced apoptosis, and elevated intracellular calcium concentration. Finally, VR1 stimulation of cultured ORS keratinocytes upregulated the expression of recognized endogenous hair growth inhibitors (IL-1β and TGFβ2) and downregulated the expression of stimulators (HGF, IGF-1, and SCF), while key differentiation markers (CK17, CK14, filaggrin, and involucrin) remained unaffected. In conclusion, VR1 is a significant novel player in human hair growth control underscoring that its physiological functions in human skin far extend beyond sensory neuron-coupled nociception.

    Notes: Although until relatively recently assumed to be devoid of innervation, there is now ample proof that the adrenal cortex receives specific neurones of several types. A general interpretation of their roles in the regulation of adrenocortical function has not been forthcoming, probably because of the variety of the different experimental approaches which have been used, and the heterogeneous observations which have been made. We here summarize the evidence which is available, and offer the view that neural inputs may provide fine tuning of the responses to systemic factors such as ACTH, through direct actions on specific adrenocortical cells. However, neural regulation also provides an integrative function, through actions on the flow of blood through the gland, which itself exerts a powerful influence on adrenocortical function.

    Notes: In the present study, the transneuronal transport of neurotrophic virus technique was used to identify cell groups of the spinal cord and the brain that are transsynaptically connected with the testis. Pseudorabies virus was injected into the testis and after survival times of 3–6 days, the spinal cord and brain were processed immunocytochemically using a polyclonal antibody against the virus. Virus-infected perikarya were detected in the preganglionic neurones of the spinal cord (T10-L1, L5–S1) and in certain cell groups and areas of the brain stem, the hypothalamus and the telencephalon. In the brain stem, the cell goups and areas in which labelled neurones were present included, among others, the nucleus of the solitary tract, the caudal raphe nuclei, the locus coeruleus and the periaqueductal grey of the mesencephalon. In the hypothalamus, virus infected perikarya were observed in the paraventricular nucleus and in certain other cell groups. Telencephalic structures containing labelled neurones included the preoptic area, the bed nucleus of the stria terminalis, the central amygdala and the insular cortex. These data identify a multisynaptic circuit of neurones in the spinal cord and in the brain which may be involved in the control of testicular functions.

    Notes: The substrates of ion- and lipld-stimulated protein kinase activity in extracts of Escherichia coli were purified by chromatography. Subsequent N-terminal sequencing suggests that these substrates include the following: a novel 80kDa protein co-purifying with RNA polymerase but partially homologous to elongation factor G; a protein with an apparent molecular weight of 65kDa identified as the ribosomal protein S1; and a 32 kDa protein identified as succinyl CoA synthetase, a key enzyme in the tricarboxylic acid cycle. The phosphorylation of these three proteins was markedly stimulated by the addition of manganese, and occurred on threonine, serine or tyrosine residues as indicated by the stability of the phosphoresidues during acid treatment. In addition, a calcium-stimulated protein of 70kDa was identified as the heat-shook protein DnaK, and a 17kDa lipid-stimulated phosphoprotein as nucleotide diphosphate kinase.

    Notes: Colletotrichum coccodes is the causal agent of the potato blemish disease black dot. Two PCR primer sets were designed to sequences of the ribosomal internal transcribed spacer (ITS1 and ITS2) regions for use in a nested PCR. The genus-specific outer primers (Cc1F1/Cc2R1) were designed to regions common to Colletotrichum spp., and the species-specific nested primers (Cc1NF1/Cc2NR1) were designed to sequences unique to C. coccodes. The primer sets amplified single products of 447 bp (Cc1F1/Cc2R1) and 349 bp (Cc1NF1/Cc2NR1) with DNA extracted from 33 European and North American isolates of C. coccodes. The specificity of primers Cc1NF1/Cc2NR1 was confirmed by the absence of amplified product with DNA of other species representing the six phylogenetic groups of the genus Colletotrichum and 46 other eukaryotic and prokaryotic plant pathogenic species. A rapid procedure for the direct extraction of DNA from soil and potato tubers was used to verify the PCR assay for detecting C. coccodes in environmental samples. The limit of sensitivity of PCR for the specific detection of C. coccodes when inoculum was added to soils was 3·0 spores per g, or the equivalent of 0·06 microsclerotia per g soil, the lowest level of inoculum tested. Colletotrichum coccodes was also detected by PCR in naturally infested soil and from both potato peel and peel extract from infected and apparently healthy tubers. Specific primers and a TaqMan fluorogenic probe were designed to perform quantitative real-time (TaqMan) PCR to obtain the same levels of sensitivity for detection of C. coccodes in soil and tubers during a first-round PCR as with conventional nested PCR and gel electrophoresis. This rapid and quantitative PCR diagnostic assay allows an accurate estimation of tuber and soil contamination by C. coccodes.

    Notes: The relationship between contamination of potato seed tubers with Erwinia carotovora ssp. atroseptica (Eca), blackleg disease development, and the incidence and level of progeny tuber contamination in field grown crops was studied in 1998, 1999 and 2000. Seed tubers were inoculated by vacuum infiltration at three levels (low, intermediate and high) with a streptomycin-resistant marker strain of Eca (SCRI1039Str) and planted in the field. Blackleg disease development was directly related to the level of seed tuber contamination. The higher the level of seed tuber contamination, the earlier in the season blackleg disease appeared and the greater the final level of disease, which continued to rise as the season progressed. High and low levels of seed tuber contamination were related to high and low incidences of progeny tuber contamination, respectively, at all sampling times. However, an intermediate degree of seed tuber contamination tended to be associated with a low level of blackleg disease, a variable incidence of progeny tuber contamination early in the season but a high incidence later in the season. The level of progeny tuber contamination, derived from seed tubers inoculated at the three different levels of Eca, was categorized into four contamination classes (〈 102, 102–103, 103–104 and 〉 104 marker strain colony-forming units/mL peel extract). At the lowest level of seed tuber contamination, progeny tuber contamination tended to be in the two lower categories. However, as seed tuber contamination increased, the proportion of contaminated progeny tubers in the two higher categories also increased. Overall, the results suggest that progeny tuber contamination is related to seed tuber contamination and blackleg disease, and that the threshold level of seed tuber contamination remains an important factor in predicting both blackleg disease and tuber health.
