    Notes: Summary Attempts were made to demonstrate the presence of specific oestrogen binding proteins (“receptors”) in bone cells. High speed cytosol preparations of bone were incubated with several concentrations of radioactive oestradiol alone and with radioactive oestradiol in the presence of a specific antioestrogen, Nafoxidine. Separation of bound and free oestradiol was carried out by dextran coated charcoal treatment and by sucrose gradient ultracentrifugation. Several types of bones likely to be oestrogen-sensitive were investigated: human femoral heads, human phalanx, rat and rabbit calvaria, humeri and femora of female rats. In all experiments we were unable to demonstrate the presence of specific oestrogen receptors in bone cell cytosol indicating that the direct effect of oestrogens on bone, if present, is not mediated by specific oestrogen receptors.

    Notes: Abstract Aims/hypothesis. Type II (non-insulin-dependent) diabetes mellitus is a multifactorial disease in which pancreatic islet amyloid is a characteristic histopathological finding. Islet amyloid fibrils consist of the beta-cell protein “islet amyloid polypeptide” (IAPP)/“amylin”. Unlike human IAPP (hIAPP), mouse IAPP cannot form amyloid. In previously generated transgenic mice, high expression of hIAPP as such did not induce islet amyloid formation. To further explore the potential diabetogenic role of amyloidogenic IAPP, we introduced a diabetogenic trait (“ob” mutation) in hIAPP transgenic mice. Methods. Plasma concentrations of IAPP, insulin and glucose were determined at 3.5 (t1), 6 (t2), and 16–19 months of age (t3). At t3, the mice were killed and the pancreas was analysed (immuno)histochemically. Results. In non-transgenic ob/ob mice, insulin resistance caused a compensatory increase in insulin production, normalizing the initial hyperglycaemia. In transgenic ob/ob mice, concurrent increase in hIAPP production resulted in extensive islet amyloid formation (more often and more extensive than in transgenic non-ob/ob mice), insulin insufficiency and persistent hyperglycaemia: At t3, plasma insulin levels in transgenic ob/ob mice with amyloid were fourfold lower than in non-transgenic ob/ob mice (p 〈 0.05), and plasma glucose concentrations in transgenic ob/ob mice were almost twofold higher (p 〈 0.05). In addition, the degree of islet amyloid formation in ob/ob mice was positively correlated to the glucose:insulin ratio (r s = 0.53, p 〈 0.05). Conclusion/interpretation. Islet amyloid is a secondary diabetogenic factor which can be both a consequence of insulin resistance and a cause of insulin insufficiency. [Diabetologia (1999) 42: 427–434]

    Notes: Summary The aim of this study was to investigate the seasonal behaviour of the plasma free testosterone: cortisol ratio (FTCR) and to relate hormonal changes to daily training volume and performance parameters on a rowing ergometer in elite female rowers. During 9 months of training preceding the 1988 Olympic Games the resting values of the FTCR in six elite female rowers were regularly (ten times) studied. Daily training volume was analysed in terms of rowed distance (l rowad) and time (t). In addition, two performance parameters, the power at 4.0 mmol·l−1 lactate concentration in the blood and the maximal power, were determined by a test on a rowing ergometer. The results indicated that the mean FTCR test value did not differ significantly from the level of the initial test or from the mean value of the directly preceding test. A significant negative correlation (r=−0.98, P〈0.01) between FTCR and l rowed was found in a period i.e. at a training camp, when there was a sudden increase in training volume. When FTCR was related to t a significant positive correlation (r=0.88, P〈0.05) was found only for the period at the training camp. Our data further suggested that the FTCR alone was not an adequate indicator for the anabolic/catabolic balance in elite female rowers. This finding was contrary to previous findings in elite male rowers. However, in training practice the FTCR seems useful as an indicator of the hormonal training status of elite female rowers when complemented with data about total and free testosterone, performance parameters and knowledge concerning cyclic variations of the FTCR.

    Notes: Summary The Breast Cancer Mucin (BCM) enzyme immunoassay utilizes two monoclonal antibodies (Mab), M85/34 and F36/22, for the identification of a mucin-like glycoprotein in serum of breast cancer patients. We have compared BCM with CA 15-3, another member of the human mammary epithelial antigen family. Serum BCM was evaluated in 151 and CA 15-3 in 134 patients with breast cancer, in 30 normal controls, in 9 pregnant women, and in 13 cancer patients (non-breast). Neither the normal controls nor the pregnant women had BCM levels 〉 25 U/ml. In contrast, 87 of 115 patients (75%) with metastatic breast cancer had BCM levels 〉 25 U/ml. All control persons had CA 15-3 levels 〈 25 U/ml, but 2 out of 9 pregnant women (22%) had levels 〉 25 U/ml. Seventy-four out of 97 patients (76%) with metastatic breast cancer had CA 15-3 levels 〉 25 U/ml. A statistically significant correlation was found between BCM and CA 15-3 in the breast cancer patient group (r = 0.883, p 〈 0.001, n = 134) and in the normal control group (r = 0.743, p 〈 0.001, n = 30). BCM and CA 15.3 both showed no correlation with CEA in breast cancer patients (r = 0.060, n = 81; and r = 0.146, n = 78, respectively). BCM had a range of sensitivity similar to that of the CA 15-3 RIA. Our results suggest that BCM may be a useful new marker for monitoring the clinical course of patients with breast cancer. Furthermore, in the evaluation of breast cancer patients, marker pands depending on disease stage may be a better choice than any single parameter in the evaluation of breast cancer patients.
