    Notes: The high molecular weight plasma glycoprotein pregnancy-associated alpha2-glycoprotein (α2-PAG) can be demonstrated on the surface of blood leucocytes and is synthesized by oestrogen-stimulated cells in culture. By the use of an indirect immunofluorescence technique, pretreatment of leucocytes with anli-α2-PAG IgG revealed α2-PAG on C3 rosette-forming cells and monocytes but not on E rosettes. Lymphocytes treated with anti-α2-PAG showed markedly diminished ability to form Fc rosettes. A percentage ranging from 15% to 20% of leucocytes from normal adults of both sexes, pregnant and oestrogen-treated females were positive for α2-PAG but no obvious correlation was observed between the incidence of α-PAG bearing cells and plasma levels of the protein.

    Notes: Histological studies on plaice tissues revealed mononuclear phagocytes emigrating through capillary endothelium in response to bloodborne foreign erythrocytes, with a 〉50% increase in circulating leucocytes within 2 h. Despite this rapid influx of leucocytes, cellular accumulation during peritoneal inflammatory responses was significantly slower than in mammals. In vitro studies with chemotaxis chambers revealed that random leucocyte migration was enhanced in the presence of endogenous chemostimulatory substances (inflammatory exudate fluid and endotoxin-activated plaice serum) although directional migration was not demonstrated. It is suggested that, compared with mammals, the slower accumulation of teleostean leucocytes at sites of injury is not due to a locomotory defect but possibly results from lower levels of endogenous chemoattractants being generated.

    Notes: When plaice were injected intraperitoneally with either oyster glycogen or live Vibrio alginolyticus an acute cellular inflammatory response was observed. The duration of these responses, 7 and 15 days respectively, exceeded the time course of the mammalian cellular inflammatory reaction. Peak leucocyte numbers were found at 2–3 days and neutrophils, which were phagocytic, were more numerous than macrophages. Although the increase in macrophage numbers was less marked, these cells appeared more actively phagocytic than neutrophils. Cortisol injections and environmentally-induced stress caused a significant reduction in the extent of inflammatory cell infiltrates, while endotoxin significantly enhanced the response.

    Notes: The influence of FK-506 and ciclosporin (CsA), either alone or in combination, on PHA- and alloantigen-induced human blood lymphocyte transformation was investigated. The concentrations of FK-506 and CsA required to cause 50% inhibition of mitogen-induced thymidine incorporation were 1.0 and 200 ng/ml respectively. Evidence of synergistic inhibition of DNA synthesis was obtained when doses of each drug below these concentrations were combined (FK-506, ≤0.5 ng/ml; CsA ≤50 ng/ml). In contrast, FK-506 and CsA failed to inhibit PHA-induced increases in cell size and granularity determined by flow cytometric analysis at 1 and 3 days of culture. Moreover, no significant changes in the expression of the interleukin 2 receptor (IL-2R; CD25), transferrin receptor (TR; CD7I) or HLA-DR were observed in terms of percentage positive lymphocytes or mean cell surface membrane fluorescence intensity. In primary mixed lymphocyte cultures, 50% inhibition of thymidine incorporation was obtained with FK-506 and CsA concentrations of approximately 0.25 and 25 ng/ml respectively. Evidence of synergy was obtained when lower doses of each drug were combined (FK-506, ≤0.1 ng/ml; CsA ≤2 ng/ml). On their own, these concentrations of CsA were ineffective. In contrast to corresponding PHA-stimulated cells, FK-506-treated alloactivated lymphocytes exhibited reductions in IL-2R, TR and HLA-DR antigen expression, which in the cases of IL-2R and TR were further reduced by combination of FK-506 with low-concentration CsA. These data provide further evidence of the potential of combined low-dosage FK-506 and CsA for the control of human T-cell activation and proliferation in response to allostimulation.

    Notes: Monoclonal antibodies were used to analyse adrenal, pancreas, thyroid, and gut samples from human fetuses (14–11) weeks estimated gestational age; EGA) for the presence of class II major histocompatibility complex (MHC) antigens (HLA-DR, DQ, DP) by immunohistochemistry. In the adrenal definitive and fetal cortex, HLA-DR+, DP−, DQ− cells were clearly demonstrated. These DR+ cells were identified, phentotypically, as predominantly tissue macro-phages and a small population of CD45R+, IgM+ lymphoid cells. Within the pancreas, numerous cell throughout the tissue were strongly DR+ but DQ−; DP+ cells were not observed until 17 weeks EGA. Using a double-labelling procedure, minor proportions of these DR+ cells were identified as macrophages or as (CD19+) B cells, while endocrine and endothelial cells were negative. Throughout the thyroid, small numbers of DR+ macrophages and small lmyphoid cells were detected, although the thyroid epithelial cells were DR+, DP+, and DQ−. Large numbers of DR+, DP+, DQ− cells were observed in the stomach wall and mucosa. In the intestine, DR+, DP+, DQ+ cells positive for all MHC class II loci products were abundant throughout the lamina propria and lymphoid aggregates. The class II antigens appeared in the proportion DR〉DF〉DQ and expression was most prevalent in the mid-gut region. A small proportion of epithelial cells of the villi along she gut were weakly DR+ but DQ−, DP−. These results show that DR+ cells in fetal endocrine tissue are mainly‘passenger leucocytes' and that, in contrast to recent reports concerning normal adult tissue, the adrenal and pancreatic endothelial cells are DR−.

    Notes: Administration of cyclophosphamide to Sprague-Dawley rats 2 days before immunization with ovalbumin in complete Freund's adjuvant resulted in a striking blood, bone marrow. and tissue eosinophilia. The number of circulating eosinophils reached a maximum (50-fold increase above normal) 2 weeks after immunization, and eosinophils were also prominent in bone marrow, lymph nodes (paracortical areas), spleen (while and red pulp), and liver. The eosinophilia could be inhibited by daily oral administration of cyclosporin A (CsA), although its effect was dependent both on the duration of treatment and on the dosage of CsA. A similar, inhibitory action of CsA was demonstrated with respect to methotrexate-induced eosinophilia. This experimental model may prove useful in the study of factors regulating eosinophil production and in examining the prospective value of CsA in the treatment of conditions where eosinophils play a central role.

    Notes: Administration of cyclophosphamide (Cy: 150 mg/kg i.p.) to rats 48 h before immunization with a T-dependent antigen (ovalbumin) resulted in a striking absolute eosinophilia in blood, bone marrow, and secondary lymphoid organs after 10 to 14 days. This eosinophilia was preceded by a significant increase in the W3/25+/OX-8+ (T helper/inducer to T cytotoxic/suppressor) ratio in lymph nodes and spleen and accompanied by a pronounced rise in splenic OX-12+ (B cell) numbers. There was also a concomitant increase in cells with the morphology and immunophenotype (OX-8+, OX-19−) of large granular lymphocytes (LGL). It is suggested that the eosinophilia linked with the B lymphocytosis may be due to cell-derived soluble factors, including a possible equivalent of eosinophil differentiation factor (EDF=interleukin 5), which also has B-cell growth factor activity (BCGF II) in mice. Ciclosporin (CsA; 25 mg/kg/day per os) from the time of immunization, did not affect the incidence of W3/25+ cells in spleen or lymph nodes, but abrogated Cy-induced eosinophilia and reduced the extent of B-cell proliferation. In addition, CsA caused a further, marked increase in the incidence of OX-8+, OX-19− LGL within the spleen. The functional role(s) of these latter cells remains to be defined.
