    Notizen: Normal rat hepatocytes were isolated and cultivated in vitro. Synthesis of secretory component was demonstrated by its accumulation in the culture medium, as measured by radioimmunoassay; by incorporation of 14C-leucine in the protein specifically precipitated with anti-secretory component antiserum; and by a positive precipitin reaction of concentrated culture medium with the same antiserum. The results explain the high levels of secretory component found in rat bile and render plausible a mechanism of hepatic IgA transfer involving secretory component as the hepatocyte membrane receptor for polymeric IgA.

    Notizen: Moncmeric (m-) and polymeric (p-) anti-DNP monoclonal (MC) rat IgA antibodies (Ab) were tested for precipitation with DNP-bovine serum albumin (DNP-BSA) and C3 conversion in rat serum, with rat MC anti-DNP igG2b used as reference. At equivalence, p-IgA rapidly precipitated DNP-BSA, with little antigen (Ag) left in the supernatant. In contrast, m-IgA at five-fold higher concentration precipitated Ag very slowly, with 〈50% of Ag precipitated at equivalence. The Ag/Ab weight ratio at equivalence was 0.13 for both m- and p-IgA, but the molar ratio was 0.3 for m-IgA and close to 1.0 for p-IgA, suggesting a higher avidity of p-IgA. Rat C3 conversion by rat IgA immune precipitates (IP) was about 20% with m-IgA and 40% with p-IgA. EGTA did not significantly affect these figures. Therefore, rat MC IgA IP activated the rat alternative C pathway. Neither rat nor mouse IgA anti-DNP IP activated C3 in normal human serum.

    Notizen: Secretory component (SC) was localized in the liver of guinea pigs, dogs, rabbits, rats, and mice. In rabbits, rats, and mice SC localized predominantly in bile canaliculi and on hepatocyte sinusoidal membranes but was doubtful in cholangincytes. In dogs and guinea pigs SC-staining was not detected in/on hepatocytes and canaliculi but was strong in/on cholangiocytes, as reported for humans. In guinea pigs IgA biliary output was small (0.23 mg/kg/day), as for dogs and humans, and below IgG output (1.4mg). in contrast to rats, whose IgA biliary output (38 mg/kg/day) was much larger than IgG output (2 mg). Biliary obstruction in guinea pigs induced only minor increases in serum IgA (+ 26% over 24 h), as reported for dogs and humans, in contrast to rats (+ 800% over 24 h) and rabbits. Hepatocyte SC expression correlates with IgA hepatobiliary excretion, being low in guinea pigs, dogs, and humans but high in rats, rabbits, and mice.

    Notizen: Groups of rats were submitted to end-to-side portacaval shunt, portacaval transposition or a control sham operation. There was an 18-fold increase of the IgA level in the serum of portacavally shunted rats (n=7) at 3–6 weeks after surgery. Gel filtration revealed that this increase was predominantly due to dimeric and polymeric IgA. The serum IgM of these shunted rats, but not the IgG, was also significantly increased (twofold) above that control values. In rats subjected to portacaval transposition (n=6), there was no significant increase of serum IgA, IgM or IgG when compared with their control values. These data confirm the active transfer, by the healthy rat liver, of IgA and, to a lesser extent, of IgM from the blood into bile. Impairment of this function, leading to the accumulation of polymeric IgA and IgM in the serum, in portacavally shunted but not portacavally transposed animals, may be related to impairment of hepatic blood Hot and hepatic atrophy, shown earlier to occur in portacavally shunted but not transposed rats. Such redaction in liver cell mass could also explain the fall in transferrin levels in shunted rats.

    Notizen: Proteins in hepatic bile from cannulated rabbits were analysed by gel filtration, electrophoresis, density gradient ultracentrifugation, and immunochemical methods. Bile-to-serum concentration ratios (no.=8) demonstrated that IgA and free secretory component (SC) were major constituents of rabbit bile. Relative concentration ratios, obtained by dividing the bile to serum level ratio tor each protein by the corresponding ratio for albumin, were 310, 1.6, 1.0, 0.82, and 0.54 for IgA, IgM, albumin, transferrin, and IgG respectively, suggesting that both IgA and IgM are selectively secreted into bile. Ligation of the bile duct (no.= 5) led to a selective increase of the serum levels of IgA and SC, the latter as secretory IgA. After intravenous injection (no.= 4) of human polymeric IgA, 37-69% of the injected dose was recovered in bile after 3 h, in contrast lo 4–5% for human monomeric IgA. The active transfer of both rabbit and human polymeric IgA into rabbit bile occurs via the same hepatic SC-mediated transport process as that described for the rat.

    Notizen: Pregnant rats were immunized with dinitrophenylated type III-pneumococcal vaccine by the intravenous, gastric, or intramammary routes. Anti-DNP antibody responses in the IgA. IgG and IgM isotypes were measured in serum, secretions and bile. Gastric intubation was most effective at eliciting IgA antibody responses in bile and secretion, whereas the other routes were more effective at inducing IgG responses in serum and bile. IgM antibody responses were infrequent and were found in fluids most closely associated with the immunization route. Isoelectric focusing studies of IgA antibodies appearing in secretions and bile revealed that the gastric route consistently elicited antibody spectrotypes with shared components. Intravenous and intramammary immunization resulted in IgA spectrotypes possessing less homology, suggesting that these protocols lead to independent antibody responses triggered in spleen, draining lymph nodes, or secretory sites. After gastric stimulation, the appearance of IgA antibodies with identical spectral components in secretions and bile favours the concept that IgA precursor cells with identical clonotype potential migrate from the gastrointestinal area to secretory sites. Antibody expression in bile appears to result from the selective transfer of IgA populations gaining access to serum after synthesis at a secretory site.

    Notizen: When mouse serum or ascites is applied on protein A-Sepharose columns and washed with enough phosphate-buffered saline, a second protein peak is often eluted with the same buffer after the first major peak of unbound proteins. This second peak is almost pure IgG1. More IgG I plus IgG2a, IgG2b and IgG3 are thereafter eluted with acid saline, 90% of the IgG1 which had been eluted with neutral buffer could be re-eluted at the same retarded position with the same buffer. When a gradient from 0 to 3 M sodium thiocyanate was started after the major peak of unbound proteins, all IgG I was eluted before IgG2 and IgG3. These results suggest that IgG1 has a much lower affiniiy for protein A than IgG2 or IgG3 and that normal mouse serum IgG1 can be purified by such a simple procedure.

    Notizen: The intracellular pathway of polymeric immunoglobulin receptor (pIgR) is governed by multiple signals that lead to constitutive transcytosis. In addition, in transfected polarized MDCK cells, polymeric immunoglobulin A (pIgA) binding stimulates rabbit pIgR-transcytosis, owing to phospholipase-Cγ1 activation and increase of intracellular calcium. Transcytosis of rat pIgR across hepatocytes is similarly accelerated by pIgA injection. In contrast we show here that human Madrin-Darby Canine Kidney (pIgR)-transcytosis, in human Calu-3 and human pIgR-transfected MDCK cells, is not promoted by pIgA, as monitored by a continuous apical release of its secreted ectodomain. However, the incubation of cells expressing human or rabbit pIgR with pIgA induces a comparable IP3 production, and pIgR-transcytosis of either species is accelerated by the protein kinase C (PKC)-activator phorbol myristate acetate. Without pIgA, mimicking phospholipase-C activation by combining low concentrations of phorbol myristate acetate with ionomycin, or high concentrations of ionomycin alone, stimulates the rabbit, but not the human, pIgR transcytosis. These data suggest that the species difference in pIgA-induced pIgR-transcytosis does not stem from the defective production of second messengers, but from a different sensitivity of pIgR to intracellular calcium. Our results outline the danger of extrapolating to humans the abundant data obtained from mucosal vaccination of laboratory animals.

    Notizen: Two intraperitoneal administrations of foreign red cells in Freund's complete adjuvant, or two intragastric intubations of erythrocytes, given to rats at 15 days interval, both elicit the appearance of specific antibodies in bile and serum. When bile was compared to serum, the selective predominance of IgA antibodies in this secretion was observed for both the intraperitoneally and intragastrically immunized groups, being more pronounced for the orally immunized group when related to IgG or IgM antibodies. The IgA content of upper intestinal washings was roughly ten-fold smaller in rats with bile duct cannulation than in sham-operated controls. Altogether, the data demonstrate that bile IgA may significantly contribute to the secretory IgA system of the gut.

    Notizen: A patient with an IgG(χ) monoclonal serum protein developed in the course of the disease a second monoclonal spike of the same light chain type and of the IgA class. The latter monoclonal protein progressively increased and eventually exceeded the first IgG(χ) protein. Antigenic analysis of the two myeloma proteins demonstrated that they shared idiotypic determinants. Immunofluorescence studies, carried out with anti γ and anti α reagents lagged with different fluorochromes revealed that the two isotypes were produced by different plasma cells. The data are discussed in the prospect of a possible transitional mechanism from IgG to IgA synthesis within a single B cell clone.
