    Notes: Density functional theory (DFT) has been applied to the study of the fluid–solid transition in systems with realistic potentials (soft cores and attractive forces): the purely repulsive WCA Lennard-Jones reference potential (LJT), the full Lennard-Jones potential (LJ) and the exponential-6 potential appropriate for helium and hydrogen. Three different DFT formalisms were used: the formulation of Haymet and Oxtoby (HO) and the new theories of Denton and Ashcroft (MWDA) and of Baus (MELA). The results for the melting pressure are compared with recent simulation and experimental data. The results of the HO version are always too high, the deviation increasing when going from the repulsive Lennard-Jones to the exponential-6 potential of H2. The MWDA gives too low results for the repulsive Lennard-Jones potential. At low temperatures, it fails for the full LJ potential while at high temperatures it is in good agreement. Including the attraction as a mean-field correction gives good results also for low temperatures. The MWDA results are too high for the exponential-6 potentials. The MELA fails completely for the LJT potential and the hydrogen exponential-6 potential, since it does not give a stable solid phase.

    Notes: A quasi-isochoric scanning method has been used to study the phase diagram of nitrogen from 150 to 550 K and up to 85 kbar in a diamond anvil cell in order to make a comparison with previous measurements of the binary phase diagram He–N2. It has been confirmed that there is only one solid-solid-fluid triple point in N2 up to 85 kbar. However, both the δ–β transition line and the melting line have shifted appreciably towards lower pressures. The present experiment shows that, as a result of this, the triple point is located at 555±5 K and 80±2 kbar, which is 20% lower in pressure than previous data.

    Notes: We have measured the melting curve of neon to 54.5 kbar and 328 K using a diamond-anvil cell. The measured points together with earlier low-pressure data are fitted accurately with a Simon-Glatzel function. Theoretical calculations of the melting curve using lattice dynamics and variational fluid theory with exponential-six potentials fitted to solid isotherm data are in good agreement with the experimental data. The law of corresponding states is tested for the melting curves of He, Ne, and Ar, and is found to be obeyed.

    Notes: We studied 44 cases of Hodgkin's disease for the presence of Epstein-Barr virus (EBV) DNA, its localization and the expression of the EBV receptor on the tumour cells. EBV DNA was found in 52% (16/31) of the Hodgkin's lymphomas using the polymerase chain reaction. With a very sensitive non-radioactive DNA in situ hybridization technique in combination with immunohistochemistry for CD 30 or CD 15 antigens, EBV DNA was localized to Reed-Sternberg cells and its mononuclear variants. The relationship between the presence of EBV DNA and the expression of the EBV-receptor CR2 (CD 21) on Reed-Sternberg cells was studied using the same techniques and two different monoclonal anti-CD 21 antibodies. CR2 could be detected on a substantial number of the Reed-Sternberg cells in EBV DNA positive Hodgkin's lymphomas (9/12; 75%), whereas in EBV negative cases positivity with anti-CD 21 was rare (1/13; 8%). The results indicate that CR2 expression on Reed-Sternberg cells and the presence of EBV DNA sequences are frequently associated in Hodgkin's lymphomas.

    Notes: Epstein-Barr virus (EBV) is frequently found in Hodgkin and Reed-Sternberg cells in Hodgkin's disease. Epstein-Barr virus has transforming properties in vitro and might be involved in the pathogenesis of certain types of Hodgkin's disease. One of the possible mechanisms is the upregulation of the human proto-oncogene bcl-2 by the latent membrane protein 1 of EBV in vitro. Another possibility might be the expression of the viral ‘bcl-2 homologue’, BHRF-1. In the present study of 64 cases of Hodgkin's disease we investigated the expression of bcl-2 at the protein level in relation to the presence of EBV. Moreover, in 10 EBV positive cases we investigated, the expression of the bcl-2 homologue, BHRF-1, by reverse-transcriptase PCR. bcl-2 was detected in 14 of 22 (64%) EBV positive and in 37 of 42 (88%) EBV negative cases. In 17 of 22 (77%) EBV positive cases Reed-Sternberg cells were negative (n = 8) or expressed the bcl-2 protein in a very low percentage (〈 5%) of cells (n = 9), whereas in 20 of 42 (43%) of the EBV negative cases the majority (〉 50%) of the neoplastic cells were bcl-2 positive. Using the reverse-transcriptase PCR with primers amplifying transcripts of BHRF-1 we were able to detect BHRF-1 transcripts in only one of the 10 tested cases of EBV positive Hodgkin's disease. Our data indicate that in EBV positive Hodgkin's disease growth advantage of Reed-Sternberg cells is not obtained by upregulation of bcl-2 or by the EBV homologue BHRF-1.

    Notes: Aim: We studied the expression of TCR ζ-chain on tumour-infiltrating lymphocytes in EBV-positive and EBV-negative cases of Hodgkin's disease (HD), to assess whether downregulation of TCR ζ-chain on tumour-infiltrating lymphocytes might be a mechanism for immune escape of the neoplastic cells.〈section xml:id="abs1-1"〉〈title type="main"〉Methods and resultsBy immunohistochemistry we investigated tissue of 27 cases of primary HD, both paraffin embedded and frozen, for the presence of T-cell receptor complex ζ-chain and other T-cell markers on the reactive cells. Strong membranous staining of TCR ζ-chain was present in all cases in frozen tissue. In contrast, in paraffin-embedded material substantial loss of TCR ζ-chain was detected in old (〉 6 years) tissues. However, no differences in either the number of positive cells or their staining intensity were observed in EBV-positive and negative cases of HD as detected in frozen tissue. Storage of paraffin-embedded tissue leads to a rapid and substantial loss of TCR ζ-chain reactivity compared to frozen material of the same HD cases. Staining reactivity of other T-cell markers (CD3, CD4 and CD8) on paraffin-embedded material remained unaffected. Immunofluorescent double-staining confirmed colocalization and coexpression of TCR ζ-chain and CD3.〈section xml:id="abs1-2"〉〈title type="main"〉ConclusionsIn frozen biopsies of primary HD TCR ζ-chain was expressed on all reactive CD3-positive cells, both in EBV-positive and EBV-negative cases. This suggests that ζ-chain downregulation is not a likely mechanism whereby neoplastic cells of HD can escape immune surveillance.

    Notes: The dynamics of concentrated, charge-stabilized colloidal silica suspensions was studied over a wide range of wave-vectors. The short-time diffusion coefficient, D(Q), was measured for concentrated suspensions up to their solidification points by photon correlation spectroscopy with coherent X-rays and compared to free particle diffusion D0, studied by Dynamic Light Scattering (DLS) in the dilute case. Small angle X-ray scattering (SAXS) was used to determine the static structure factor S(Q). D0/D(Q) peaks for Q values corresponding to the maximum of the static structure factor showing that the mostly likely density fluctuations decay the slowest. The data allow one to estimate the diffusion coefficient D(Q) in the Q → 0 and Q → ∞ limits. Thus, hydrodynamic functions can be derived free from any modeling of the static or dynamic properties. The effects of hydrodynamic interactions on the diffusion coefficient in charge-stabilized suspensions are presented for volume fractions 0.075 〈 Φ 〈 0.28.

    Notes: Lactose-specific enzyme IIA of the phosphoenol:pyruvate-dependent sugar phosphotransferase system from Lactococcus lactis has been crystallized in phosphate buffer. The crystals belong to space group P41212 or its enantiomorph P43212 with unit-cell axes a = b = 90.9 and c = 82.4 Å. The packing parameter (Matthews parameter) Vm of 2.48 Å3 Da−1 is consistent with one trimer per asymmetric unit and non-crystallographic threefold symmetry has been confirmed by calculating a selfrotation function. The crystals diffract X-rays to at least 2.3 Å resolution, are stable in an X-ray beam and are therefore appropriate for structure determination. Native data to 2.3 Å resolution have been collected using a MAR image-plate system at a synchrotron source. One isomorphous heavy-atom derivative has been identified and the presence of an isomorphous signal in the data has been confirmed by Patterson methods.

    Notes: Synchrotron small-angle X-ray scattering (SAXS) experiments have been performed at the ESRF on colloidal dispersions. The samples are optically multiply scattering and are aimed at so-called photonic applications. Scattering from dilute suspensions yields the particle form factor, which shows many oscillations. From these results, the radius, size distribution and information about the internal structure of the particles in situ can be determined, in contrast to most studies on colloids. Using the form factor, structure factors can be extracted from the diffraction patterns of the colloidal crystals. The structure factors clearly reveal sharp Bragg peaks. The crystal structure, lattice parameter and average orientations were determined as a function of height in sedimented samples. This provides important information for the interpretation of optical experiments.
