    Notes: In suspension cultured cells of Nicotiana tabacum L. cv. Wisconsin 38 the .concentration of 18 nucleotides and 3 nucleosides have been determined using a new procedure developed for the extraction, purification and HPLC separation of these compounds from plant tissue. The different nucleotide pools increase in size immediately at the onset of the batch culture and reach distinct maxima at the very beginning of the proliferative phase. The main component are the uracil nucleotides with UDP-sugars as the predominant fraction followed by the adenine nucleotides; the energy charge is maintained at a high and constant value throughout the whole culture time. During the growth interval the increases in the nucleotide pools reveal that the cell proliferation phase is followed by an extensive phase of cell elongation. Whereas the concentration of the total nucleotides varies by a factor of about 3 along the growth curve, the ratio of uracil to adenine nucleotides is kept fairly constant indicating regulatory mechanisms for correlation of the individual nucleotide pools.

    Notes: Tobacco (Nicotiana tabacum L. cv. Samsun) plants were treated once with 2,4-dichlorophenoxyacetic acid (2,4-D) at the 8-leaf stage. The effect of the herbicide on leaf metabolism was followed over 7 days by determination of the ribonucleotide pools, including NAD+, NADP+ and UDP-sugars, by high-preformance liquid chromatography. 2,4-D treatment resulted in large changes in the nucleotide concentrations, the magnitude and sign of which were dependent upon the leafage. The nucleotide pools decreased in the apical tissue, but increased strongly in the mature leaves with the highest relative increase in the oldest leaf tested. The time course of the changes revealed a maximum on day 5 after 2,4-D treatment. The increase in the adenine nucleotide pools, energy charge and the NADVNADP+ ratio are interpreted to indicate a stress situation. The different responses of young, mature and senescent tissue to the synthetic auxin could reflect their different inherent sensitivity due to the natural auxin gradient.

    Notes: The content of the different ribonucleotides, nucleoside diphosphate sugars, NAD+ and NADP+ was determined in the leaves and roots of single plants of Nicotiana tabacum L. cv. Samsun using a new procedure. The determination makes use of extraction with HClO4 followed by extract purification and separation by a combination of anion-exchange and reversed phase high performance liquid chromatography. The largest pools were the adenine nucleotides and the uracil nucleotides with nucleoside diphosphate sugars as the main fraction. Leaf senescence was accompanied by a strong reduction of the nucleotide content of the leaves, but the adenine nucleotide-derived energy charge remained at a high level and even increased at the later stages of senescence. This indicates a balanced metabolism also in the older leaves and excludes the possibility that leaf senescence is triggered by the energy charge.

    Notes: A procedure for the determination of metabolites of the biochemical pathway ornithine to N-methyl-δ1-pyrrolinium salt (N-methylpyrroline) is described. Plant tissue was extracted with 0.5 M HCl and the extract purified on C18-cartridges. Ornithine was reacted with o-phthaldialdehyde, putrescine and N-methylputrescine with dansyl chloride and the products were separated by reversed-phase high-performance liquid chromatography (HPLC). N-methylpyrroline was determined by cation-exchange HPLC without derivatization. The metabolites in the roots of tobacco (Nicotiana) species with different nicotine-producing capacities were determined. Furthermore, the specific activities of the enzymes ornithine decarboxylase (EC 4.1.1.17), putrescine N-methyltransferase (EC 2.1.1.53) and N-methylputrescine oxidase were determined. Both the metabolite pools and the enzyme activities were correlated with the different nicotine-producing capacities of the different tobacco species.

    Notes: The activities of the three enzymes of the route ornithine to the N-methyl-Δ1-pyrrolinium salt and of the eight enzymes of the route quinolinic acid to nicotinic acid (pyridine nucleotide cycle) were determined in the roots of different Nicotiana tabacum L. cultivars and for comparison in tomato (Lysopersicon esculentum L. cv. Vollendung) roots. Enzyme activities were further followed in different plant organs, dedifferentiated tissues, root organ cultures and in the roots after decapitation of tobacco plants. The data are in accord with the concept that the two routes are predominantly regulated by the enzymes putrescine methyltransferase (EC 2.1.1.53) and quinolinic acid phosphoribosyltransferase (EC 2.4.2.19), respectively. Further regulation involves the enzymes NAD+-pyrophosphatase (EC 3.6.1.22) and nicotinic acid mononucleotide glycohydrolase; these findings confirm the suggestion that the transformation to nicotinic acid is performed along two routes: either via the synthesis and degradation of NAD+ or by a direct step through the action of a specific glycohydrolase.

    Description / Table of Contents: Zusammenfassung Es wird eine elektrophoretische Routinemethode zur Identifizierung von Weizensorten beschrieben. Die Trennung der Weizengliadine wird in Polyacrylamidgelen (PAG) mit einem kontinuierlichen Essigsäure/Glycin-Puffersystem durchgeführt und ist, bei gleicher Trennschärfe wie die derzeit üblichen Methoden mit Lactatpuffer, billiger und weniger störanfällig. Zur Veranschaulichung der erzielten Ergebnisse wird eine Auswahl Gliadinbandenmuster in Österreichverwendeter Weizensorten gezeigt.
    Notes: Summary An electrophoretic routine method for identification of wheat varieties is described. The separation of wheat gliadins is performed on polyacrylamidgels (PAG) in a continuous acetic acid/glycine buffer system. The method provides the same separation sharpness as present methods with aluminium lactate/lactic acid buffer, but is inexpensive and less susceptible to analytical problems. The results obtained are illustrated by a number of gliadin patterns of Austrian wheat cultivars.
