    Notes: The stimulation of macrophages is of importance to the defense against intracellularly replicating microorganisms such as Leishmania. In this study the direct effect of recombinant interleukin-10 (IL-10) on the leishmanicidal effector functions of murine peritoneal or bone marrow derived macrophuges was investigated. IL-10 almost completely inhibited the killing of intracellular leishmania at concentrations above 10ng/ml. This inhibitory effect was independent of the stimulus used as the activation of macrophages by IFN-γ and IL-7, recently shown to possess macrophage activating properties, were suppressed by IL-10. Kinetic experiments revealed that IL-10 must be present during the process of macrophage activation and that the leishmanicidal effector function of fully activated macrophages was not influenced. Furthermore, in the absence of exogenously added IL-10, the addition of neutralizing antibodies against IL-10 or IL-10-specific antisense phosphorothioate DNA-oligonucleotide led to an enhanced killing of parasites after stimulation with either IFN-7 or IL-7. In accordance with this, IL-10 mRNA was readily detectable in murine macrophages by PCR with reverse transcribed mRNA. These results indicate that IL-10, which is endogenously produced by macrophages, acts as an autocrine deactivating factor supporting the survival of the parasite.

    Notes: Summary Two patients with severe cerebral symptoms of toxemia of pregnancy were examined using computed tomography (CT) and angiography. CT disclosed extensive areas of low attenuation within the cerebral hemispheres. Angiography revealed constriction and narrowing of proximal and peripheral vessels suggesting vasculitis with extensive areas of impaired regional cerebral blood flow. A review of the known pathology and the theories regarding the pathophysiology of the cerebral effects of toxemia is presented.

    Notes: Abstract Purpose: The experiments aimed to determine if α-chemokine inhibitors are effective suppressors of the growth of adenocarcinomas, a neoplasm with a high mortality rate. Methods: Expression of growth-related oncogene (GROα) and interleukin-8 (IL-8) was determined by enzyme-linked immunosorbent assay. Inhibition of α-chemokine binding to tumor cells was assessed in the presence and absence of the hexapeptide, antileukinate. 3-(4,5-Dimethylthiazol-2-yl)-2,5-diphenylte-trazolium bromide assays were performed to determine the effect of α-chemokines, monoclonal antibodies (mAb), and antileukinate on cell proliferation. Finally, antileukinate inhibition of human, lung adenocarcinoma tumor growth, was determined in BALB/c nude mice. Results: All of the adenocarcinomas tested produced either GROα or IL-8 or both. Proliferation of lung, stomach and colon adenocarcinoma cells was inhibited by anti-GROα mAb and/or anti-IL-8 mAb while recombinant human GROα stimulated the proliferation of lung and stomach adenocarcinomas. Antileukinate inhibited GROα binding to specific receptors on adenocarcinoma cells and inhibited the proliferation of all adenocarcinomas tested. Colon-derived adenocarcinomas specifically bound IL-8 and this binding was also inhibited by antileukinate. Administration of antileukinate in vivo inhibited the tumor growth of adenocarcinoma A549. Conclusions: GROα and IL-8 are necessary for the growth of lung, stomach and colon adenocarcinomas, and can be inhibited by the hexapeptide, antileukinate. The findings suggest the possibility of using α-chemokine receptor inhibitors in the treatment of adenocarcinoma.

    Description / Table of Contents: Summary In most factor analytical studies of schizophrenic symptomatology a three-factor solution was found. The aim of this study was to investigate symptomatological dimensions in old age and to clarify whether the dimensions correlate differently with neuropsychological and motor parameters. A total of 131 DSM-III-R schizophrenics (mean age 68 years) were assessed using SANS, SAPS, a neuropsychological test battery and motor scales. A factor analysis yielded three dimensions (negative, desorganized, paranoid), two of which (negative, disorganized) showed different correlations with neuropsychological and motor phenomena. Thus, three symptomatological dimensions could also be demonstrated in a chronic, old-age schizophrenic sample. The pathophysiological significance of the different correlations with neuropsychological and motor parameters should be clarified in neuroimaging and neuropathological studies.
    Notes: Zusammenfassung In den meisten faktorenanalytischen Studien der schizophrenen Symptomatik wurde eine 3-Faktoren-Lösung gefunden. Ziel dieser Studie war es, die symptomatologischen Dimensionen der Schizophrenie im Alter zu untersuchen und zu prüfen, ob die verschiedenen Dimensionen unterschiedlich mit kognitiven und motorischen Parametern korrelieren. 131 gemäß DSM-III-R chronisch schizophrene Patienten (Alter x–=68 Jahre) wurden mittels SANS, SAPS, einer neuropsychologischen Testbatterie und motorischen Skalen untersucht. Die Faktorenanalyse der Symptomatik ergab 3 Dimensionen (negativ, desorganisiert, paranoid-halluzinatorisch). Der negative und desorganisierte Faktor korrelierten signifikant unterschiedlich mit neuropsychologischen und motorischen Auffälligkeiten. Auch bei chronischen, altgewordenen Schizophrenen lassen sich somit 3 symptomatologische Dimensionen nachweisen. Die pathophysiologische Bedeutung der unterschiedlichen Korrelationen mit den neuropsychologischen und motorischen Parametern sollte in bildgebenden und neuropathologischen Studien abgeklärt werden.
