    Notes: Summary In a national multicentre trial in the FRG patients with Hodgkin's lymphoma in stages CS/PS III B/IV were entered into the HD 3 protocol and received induction chemotherapy with 3× (COPP+ABVD). Patients in complete remission (CR) received consolidation therapy by either radiotherapy (20 Gy IF) or chemotherapy (COPP+ABVD). Patients not in CR received salvage therapy (40 Gy in case of persisting nodal disease, else 4× CEVD chemotherapy). Between July 1983 and May 1987 230 untreated patients aged 15 to 60 qualified for this HD 3 protocol. This analysis is based on the first 137 patients evaluable for response. Of these, 86 (63%) achieved CR after induction chemotherapy. Including salvage therapy a total of 104 patients (76%) achieved CR. Univariate and multivariate prognostic risk factor analyses were performed using freedom from treatment failure (FFTF) as endpoint. Sex, age, splenectomy, bone marrow, liver and bone involvement had no prognostic impact nor had stage according to the Ann Arbor classification. In contrast, a pretreatment erythrocyte sedimentation rate (ESR) above 80 mm/h and a serum alkaline phosphatase (AP) above 230 IU/ml appeared as significant risk factors (p〈0.01, relative risk 2.3). The two parameters were not independent. Comparing a group A (ESR ≤ 80 and AP ≤ 230) versus a pooled group B (ESR〉80 and/or AP〉230) increased the difference (p〈0.001, relative risk of 2.8) which was also significant for survival (p〈0.04).

    Description / Table of Contents: Zusammenfassung Zwei Methoden der Hauttransplantation bei der Maus werden beschrieben. Die eine dient zur Verpflanzung sehr großer Hautstücke direkt auf die Rückenfascie ohne Erhaltung des Panniculus carnosus, mit anschließender Fixation durch Michel Wundklammern. Diese Methode ist besonders für die Transplantation carcinogenbehandelter Haut geeignet. Bei der anderen Methode werden kleine Hauttransplantate auf den Panniculus carnosus der Rückenhaut aufgelegt und mit einem Stück Hansaplast abgedeckt, das an der Rückenhaut festgeklammert wird. Diese Technik eignet sich besonders für die Verpflanzung von Allotransplantaten, da die Abstoßung sehr gleichmäßig und für alle Transplantate zeitlich nahe beieinander liegend erfolgt. Die Vorzüge beider Methoden, wie geringe Mortalität, hohe Erfolgssicherheit und kosmetisch gute Ergebnisse liegen zum großen Teil in der Art der Fixation der Transplantate begründet. Diese erfolgt so kurz wie möglich, für 1 Tag bei großen und 4–5 Tage bei kleinen Transplantaten und erlaubt so früh eine dauernde ungehinderte Beobachtung.
    Notes: Summary Two methods of mouse skin transplantation are described. Large skin grafts are placed directly on the fascia of the back and fixed with wound clips. This method is suited primarily for grafting carcinogen treated skin. Small skin grafts are applied to the panniculus carnosus and simply covered with a small piece of “Hansaplast” clipped to the back skin. This method is appropriate in particular for allografting, as rejection for all animals occurs nearly simultaneously. For both methods the mortality rate is very low, the results are excellent and highly reproducible. This is mainly due to the method of fixation which is kept as short as possible, 1 day for the large and 4–5 days for the small grafts, thus allowing an early and continuous inspection of the grafts.

    Description / Table of Contents: Zusammenfassung Nach intragastrischer Initiierung mit dem Carcinogen DMBA kam es nach anschließender Promotion mit dem Cocarcinogen TPA zur Bildung einer großen Zahl von Papillomen. Hauttransplantate aus initiierten Mäusen heilten auf isologen Empfängern erfolgreich und auf Dauer ein. Dies widerlegt eine Hypothese, daß eine mit einem Carcinogen behandelte Haut als Ganzes sog. Neoantigene im Sinne von Transplantationsantigenen erwirbt. Die Transplantation selbst hat nur einen geringen tumorpromovierenden Effekt. Sie scheint darüberhinaus die Anzahl der initiierten Zellen im Transplantat zu vermindern, da die Zahl der Papillome in initiierten und autotransplantierten Tieren nach Dauerbehandlung mit TPA geringer war als in ähnlich behandelten, aber nicht transplantierten Kontrolltieren. Die Zahl der Papillome auf Autotransplantaten initiierter Mäuse lag etwas höher als auf Isotransplantaten initiierter Haut in normalen Empfängern. Der Verlust initiierter Zellen während der Transplantatheilung könnte durch eine spezifische Immunreaktion verursacht worden sein. Die lokalen Prozesse während der Anheilung erlauben vielleicht einen intensiveren Kontakt zwischen dem Immunsystem und prospektiven Tumorzellen. Die höhere Zahl an Papillomen auf Autotransplantaten könnte dann auf eine Immunsuppression durch das Carcinogen zurückgeführt werden. Die Problematik eines optimalen Kontaktes zwischen tumorassoziierten Transplantationsantigenen und dem Immunsystem könnte eine der Ursachen dafür sein, warum das Prinzip der “immunological surveillance” funktionieren oder versagen kann.
    Notes: Summary C3H mice initiated intragastrically with the carcinogen DMBA produced a large number of papillomas upon subsequent treatment with the cocarcinogen TPA. Skin from initiated mice could successfully be grafted to normal isologous recipients, the grafts remaining in good condition during an observation period of 8–12 months. This disproves a hypothesis that the cells of carcinogentreated skin as a whole might acquire new common transplantation antigens. Grafting had only a low tumor promoting effect. It seems to reduce the amount of initiated cells in the grafts as the number of papillomas in initiated and autografted animals after administration of TPA was lower than that in similarly treated but ungrafted controls. The amount of papillomas on initiated autografts was higher than that on initiated isografts in normal recipients. The loss of initiated cells during grafting could be caused by a specific immune reaction, the local processes during grafting allowing perhaps a more intense contact between immune system and prospective tumor cells. The higher amount of papillomas on autografts could then be due to immuno-suppression in these animals by the carcinogen. The optimals presentation of tumor-associated transplantation antigens to the immune system might be one of the keys to the understanding why immunological surveillance may fail or function.

    Notes: Summary The development of epidermis and epidermal appendages from dissociated cells of neonate mouse skin was examined by transplantation of cell suspensions to subdermally prepared, protected graft beds. Using Ficol gradients and culture procedures, we prepared subfractions of primary cell suspensions consisting of essentially pure epidermal cells or fibroblasts. Reformation of an epithelium structurally similar to the epidermis was observed from transplanted epidermal-cell suspensions, but formation of hair follicles and development of normal epidermal microarchitecture was observed only when epidermal cells were transplanted together with cells of dermal origin. This pattern was observed following transplantation of either fresh-cell isolates or cells cultured up to 7 days prior to transplantation.
