    Notes: 1. We investigated candidate loci for salt-sensitive high blood pressure (BP) in F2 progeny from crossing Wistar-Kyoto and stroke-prone spontaneously hypertensive rats.2. In female F2 progeny, systolic and diastolic BP on the 12th day and the seventh month after salt loading was strongly liked with the D3Mgh12 and D3Mgh6 loci on chromosome 3, respectively.3. These loci were linked with BP only in female F2 progeny, not in males.4. These results indicate that hormonal factors may influence salt sensitivity, particularly with respect to gender differences.

Peritoneal macrophages (Mø) were harvested from non-stimulated 2 month old SHRSP and WKY, and incubated with acetyl LDL or [14C]-oleate-albumin complex to check total, free and esterified cholesterol (Cho) accumulation as well as acy coenzyme A-cholesterol acyltransferase (ACAT) activity and its activation by hyperlipidaemic serum.2. Total and esterified Cho accumulation as well as ACAT activity were enhanced in Mø from SHRSP compared with those from WKY.3. To observe the dietary or pharmacological influence on atherogenesis, arteriolipidosis-prone SHR (ALR) were given a high fat cholesterol diet with 3% taurine or a Ca blocker (Nilvadipine, N; 30 mg/kg), both of which significantly decreased serum Cho level and clearly attenuated arterial fat deposition.

The purpose of the present study was to investigate how angiotensin 1 (AI), angiotensin II (AII), an angiotensin converting enzyme inhibitor (ACE inhibitor; ACE-I) and a serine proteinase inhibitor contribute to the protein metabolism of cultured newborn spontaneously hypertensive rats (SHR) heart cells. We examined the uptake of [3H]-uridine and [3H]-proline into cultured cardiac myocytes and fibroblasts, respectively.2. Both AI and AII increased the uptake of [3H]-uridine into myocytes in a concentration-dependent manner. However, the effect of AI was denied in the presence of the ACE-I with the concentration of 10−6 g/mL. Both AI and AII increased the uptake of [3H]proline into cardiac fibroblasts in a concentration-dependent manner. However, this effect was only partially abolished in the presence of 10−6 g/mL of the ACE-I, which was the maximal concentration that did not exert any effect on the [3H]-proline uptake. In the presence of AII receptor antagonist, [Sar1, Led8]- AII, the uptake of [3H]. proline into cardiac fibroblasts was completely inhibited. Moreover, the stimulatory effects of AI on the uptake of [3H]-proline into cardiac fibroblasts were completely inhibited in the presence of a serine proteinase inhibitor in addition to the ACE-I.3. These results suggest that an ACE-I has different effects on protein metabolism in the heart and also suggest the presence of serine proteinase in cultured cardiac fibroblasts from SHR.

Free Ca2+ ([Ca2+]i) and Mg2+ ([Mg2+]i) were measured in peripheral lymphocytes from stroke-prone spontaneously hypertensive rats (SHRSP) and normotensive Wistar-Kyoto rats (WKY) at the age of 5, 7 and 17 weeks, from various antihypertensive agents-treated SHRSP, and from secondary hypertensive WKY.2. At the age of 5 weeks, no difference was observed in systolic blood pressure (SBP), or lymphocyte [Ca2+]i and [Mg2+]i between SHRSP and WKY. At the age of 7 or 17 weeks, SBP and [Ca2+]i of SHRSP were significantly higher than in WKY, and at the age of 17 weeks, [Mg2+]i of SHRSP was significantly lower than in WKY. Further, [Ca2+]i or [Mg2+]i was positively or negatively correlated to SBP, and [Mg2+]i was negatively correlated to [Ca2+]i.3. SBP of SHRSP fell significantly after antihypertensive treatment with calcium antagonist, angiotensin-converting enzyme (ACE) inhibitor or hydralazine for 40 days. [Ca2+]i was significantly lower in calcium antagonist and hydralazine groups, and tended to be low in ACE inhibitor group. These four groups showed no difference in [Mg2+]i.4. After 40-day administration of NG-nitro-l-arginine (l-NNA), WKY developed severe hypertension, but there were no significant differences in lymphocyte [Ca2+]i and [Mg2+]i between the l-NNA treated and non-treated groups.5. These results suggested that increased lymphocyte [Ca2+]i and decreased [Mg2+]i observed in SHRSP are not only secondary to hypertension but possibly related to a basic genetic abnormality of divalent cation handling.

A significant rise of the systolic blood pressure was observed after the administration of NO2Arg with food (0.023% in weight, about 2.8 mg of NO2Arg per rat per day). Relaxation by acetylcholine decreased markedly in ring preparations of the thoracic aorta of NO2Arg-treated rats. However, glyceryltrinitrate-induced relaxation was not reduced after NO2Arg administration, suggesting that NO2Arg administration specifically inhibited endothelium-dependent relaxation.3. An increase of blood pressure may be because oral administration of NO2Arg inhibited endothelium-dependent relaxation in vivo suggesting that the release of EDRF is important in physiological control of blood pressure.

Effects of potassium (K) supplementation (100 mEq/day) on urinary sodium (Na) excretion and on the secretion of atrial natriuretic polypeptide (ANP) during salt loading (350 mEq/day) were studied in 12 healthy salt-resistant normotensives under strictly controlled metabolic ward conditions.2. Urinary volume and Na excretion on the first day of the high salt period (HSP) were significantly greater in the K-supplemented group (KG) than in the control group (CG).3. There was a significant gain in bodyweight after salt loading in both groups, with a significantly greater gain in CG on the second day of HSP. Haematocrit decreased significantly during salt loading in both groups, the degree of which was significantly greater in CG.4. Plasma norepinephrine decreased significantly during salt loading in both groups, the degree of which was significantly less in KG than in CG. A significant increase in plasma ANP was observed in CG on and after the second day of HSP, while a significant increase in plasma ANP was observed on the fifth day of HSP in KG.5. These findings indicate that K supplementation accelerates diuresis and natriuresis, resulting in moderate suppression of volume expansion induced by salt loading and that this accelerated diuresis and natriuresis is not a result of the action of ANP.

Significant rise of the systolic blood pressure was observed after 1 week administration of NO2Arg in food (0.023% in weight, about 2.8 mg of NO2Arg/rat per day) in female rats of stroke-prone spontaneously hypertensive rats (SHRSP) and normotensive Wistar-Kyoto rats (WKY). The rises were not different between SHRSP (21 mmHg) and WKY (23 mmHg).2. In ring preparations of the thoracic aorta of NO2Arg-administered rats of both strains, relaxation by acetylcholine decreased markedly compared with those of the control rats (to 43–44%). On the contrary, glyceryltrinitrate-induced relaxation was slightly but significantly increased in the aorta of WKY after NO2Arg administration and the same tendency was observed in SHRSP.3. The rise of blood pressure and the decrease of acetylcholine-induced relaxation suggested that NO2Arg inhibited the endothelium-dependent relaxation not only in WKY but also in SHRSP. The relaxation of the thoracic aorta preparation of SHRSP by acetylcholine was much less (ca 38%) than that of WKY; however, that of SHRSP by glyceryltrinitrate was slightly less (ca 74%), indicating that endothelium-dependent relaxation declined in vascular preparation of SHRSP.4. The present results suggest that endothelium-dependent relaxation has some contribution on blood pressure regulation in the hypertensive state, although a decline of endothelium-dependent relaxation is evident in vascular preparation of SHRSP compared with WKY.

The effects of extracellular Na+ concentration ([Na+]o) on serum-stimulated c-fos mRNA induction in vascular smooth muscle cells (VSMC) were studied to examine whether the activation of Na+/H+ exchange and the following intracellular alkalinization are necessary in the induction of c-fos in VSMC.2. When monitored with 2′, 7′-bis (carboxylethyl)-5(6)-carboxyfluorescein, the reduction in [Na+]o in HCO3−-free buffer caused a dose-dependent inhibition of Na+-dependent intracellular pH recovery in acid-loaded VSMC. In addition, serum-stimulated intracellular alkalinization in VSMC was completely blocked by the removal of extracellular Na+.3. By contrast, c-fos induction, quantified by slit blot hybridization of c-fos mRNA, was not fully inhibited by the reduction of [Na+]o. Furthermore, amiloride and ethylisopropyl-amiloride, inhibitors of Na+/H+ exchange, only partially blocked c-fos induction by serum.4. We concluded from the observation above, that the intracellular alkalinization caused by the activation of Na+/H+ exchange was unnecessary in serum-stimulated induction of c-fos mRNA in VSMC.

    Notes: The preventive effects of sesamin, a lignan from sesame oil and vitamin E on hypertension and thrombosis were examined using stroke-prone spontaneously hypertensive rats (SHRSP). Animals at 5 weeks of age were separated into four groups: (i) control group; (ii) vitamin E group, which was given 1000 mg alpha-tocopherol/kg diet; (iii) sesamin group, given 1000 mg sesamin/kg diet; and (iv) vitamin E plus sesamin group, given 1000 mg alpha-tocopherol plus 1000 mg sesamin/kg diet for 5 weeks from 5 to 10 weeks of age. Resting blood pressure was measured by the tail-cuff method once weekly. A closed cranial window was created in the right parietal bone of the rat and platelet-rich thrombi were induced in vivo using a helium-neon laser technique. The number of laser pulses required for formation of an occlusive thrombus was used as an index of thrombotic tendency. In control rats, systolic blood pressure and the amount of urinary 8-hydroxy-2′-deoxyguanosine (8-OHdG) became significantly elevated with age. However, the elevation in blood pressure and 8-OHdG were significantly suppressed in rats administered vitamin E, sesamin, or vitamin E plus sesamin. At 10 weeks, the number of laser pulses required to induce an occlusive thrombus in arterioles of the control group was significantly lower than in the other groups (P 〈 0.05). These results indicate that chronic ingestion of vitamin E and sesamin attenuated both elevation in blood pressure, oxidative stress and thrombotic tendency, suggesting that these treatments might be beneficial in the prevention of hypertension and stroke.
