    Notes: Radiofrequent magnetron sputtering was used to produce calcium phosphate coatings on metal and plastic substrates. Scanning electron microscopy showed that the deposited films had a uniform thickness and a dense columnar structure. Energy-dispersive X-ray analysis, X-ray crystal diffraction and atomic absorption spectrometry demonstrated that the sputtered layer was well-crystallized calcium phosphate ceramic with a Ca/P ratio varying between 1.9 and 2.5. The biocompatibility of the coatings was determined by in vitro and in vivo experiments. It was found that the coatings were biocompatible without any sign of adverse tissue reaction. It was concluded that magnetron sputtering is a promising method for forming a biocompatible ceramic coating onto an implant material. Nevertheless, several problems have to be solved before magnetron sputtering can be used on a routine basis for the production of Ca/P coatings.

    Notes: Abstract Four different implant materials were installed into the mandibular corner of goats to investigate the trabecular bone response in a mainly unloaded model. The implants were installed using a standardized technique and were left in situ for 12 weeks. One goat had to be sacrificed after surgery because of a broken rib; the other animals healed uneventfully. After sacrifice of the animals, the bone response to the uncoated and the three different Ca–P implants was evaluated histologically and histomorphometrically. Four sections of each implant were evaluated; two were located in the cortical and two in the trabecular bone. Of the 44 retrieved implants, 20 implants appeared to be installed partially in the mandibular canal, as clearly visible on the X-rays. These implants were not used in the histomorphometrical measurements. Histological evaluation showed that the trabecular and cortical bone reactions were similar; there was no significant difference in the percentage of bone contact nor in the amount of bone in contact with the implants. In conclusion this study showed that the mandibular corner is an unsatisfactory model for the installation of implants because of anatomical restrictions. Also, the experiment remained inconclusive about the influence of loading conditions on bone behaviour. Nevertheless, the histological results confirmed the bioactive properties of Ca–P coatings.

    Notes: A newly developed saw technique was developed to produce sections circa 10 μm or thicker from fresh bone or dentine and from plastic embedded undecalcified bone tissue with or without implant materials. The method comprises only one step because grinding or polishing to make the sections thinner is not necessary. The bone slices can be decalcified rapidly without using aggressive solvents and used for making ultrathin sections for electron microscopy. Sections of fresh dentine of 15 to 30 μm are transparent which makes it possible to study osteoclastic resorption in vitro. Sections, 10 μm thick, with an intact interface of bone and implant material can be observed for biocompatibility studies. © 1994 John Wiley & Sons, Inc.

    Notes: The aim of this study was to investigate the bone response to three different types of titanium (Ti) alloys and hydroxyapatite (HA) coated titanium alloy by histological and histomorphometrical analysis. Therefore, implants made of these materials were inserted into the tibia of rabbits. Implantation times were 6 and 16 weeks. The histological evaluation included measurement of the amount of bone apposition and analysis of the bone reaction and interface characteristics around the implants. The results demonstrated no marked differences in bony reaction to the different implant materials. In addition, the HA coatings showed loss of thickness. © 1993 John Wiley & Sons, Inc.

    Notes: This study reports on certain aspects of wound-healing around percutaneous implants. Plasma-sprayed and dense hydroxylapatite, titanium, and carbon test implants were inserted into the tibia and the cranium of 12 rabbits. Four and 8 months after insertion, the animals were sacrificed and the implants with their surrounding tissues were processed histologically. Light- and transmission electron microscopic investigations were performed. It is found that direct and indirect boneanchoring favors the longevity of percutaneous implants. No differences in tissue reaction between the various implant materials were observed.

    Notes: Previous studies suggested that surface topographic configurations of 1-3 μm influence cellular behavior and tissue response. They did not address which specific aspect of the configurations elicits the cellular response. We therefore investigated the effect of the orientation of several surface configurations. Seven different textures on polydimethyl siloxane (silicone; Dow Corning Silastic®) specimens were used to test the question of whether orientation into (down) or out of the surface (up) affected cellular response to a material. The textures were smooth and photoetched configurations of 2 μm up, 2 μm down, 5 μm up, 5 μm down, 10 μm up, and 10 μm down. The response of cultured fibroblasts on these surfaces was compared with that of a standard tissue culture material, polyethylene terephalate (ThermanoxTM). The cell density was measured over a 12-day period with the use of a colorimetric assay. The uptake of methylene blue was measured daily and compared as an absorbance in a destaining agent. Cells on the 2 and 5 μm up arrays showed increased rates of proliferation and cell density as compared with their down counterparts. This would indicate that textures of 2 and 5 μm have a significant influence on cell growth, and that the surface with hills has a greater effect than the surface with wells. In contrast, the 10 μm up and 10 μm down arrays did not prove to be statistically different from smooth ones. This indicates that the orientation effect is related to the configuration size and that this configuration size is not viewed differently from smooth silicone by the cells. The presented data are in agreement with results of this laboratory and others that fibroblasts recognize the dimensions of surface configurations and react accordingly. Specifically, they appear to react to the uppermost surface area presented to them, but conclusive data can only be obtained from a study of the focal adhesions. © 1994 John Wiley & Sons, Inc.

    Notes: This study reports on the soft-tissue response to titanium fiber mesh percutaneous implants. The implants were inserted in the backs of 15 rabbits. The surgical procedure was performed in two steps: implantation of the subcutaneous part of the implants and, after 3 months, fixation of the percutaneous part into the subcutaneous part. Two and 6 months after insertion the animals were sacrificed and the implants, with their surrounding tissues, were processed histologically. Light microscopic analysis demonstrated that only one implant showed considerable skin retraction in the percutaneous area. Around the rest of the implants no downgrowth or only a very limited epidermal downgrowth was observed. Furthermore, the fiber mesh material showed good biocompatible behavior. These results confirm the previous finding that sintered titanium fiberweb structures are effective for the stabilization of percutaneous devices located in soft tissues and can improve their performance. © 1994 John Wiley & Sons, Inc.

    Notes: The aim of this study was to investigate the bone response to calcium phosphate (Ca-P) plasma-spray and radiofrequency magnetron sputter-coated implants with comparable roughness. Therefore, tapered conical screw designed implants were installed in the trabecular bone of the femurs of nine goats. They were provided with two types of coatings, a plasma-spray dual coating of fluorapatite and hydroxyapatite (FA/HA-PS) and a titanium plasma-spray coating, covered with an amorphous Ca-P magnetron sputtercoating (TPS/Ca-P-a). These implants were evaluated histologically and mechanically after 3 months of implantation. A well-controlled method to apply and measure a torsional force to load the screw-type implants to the point of failure was introduced. All implants healed uneventful and were well fixed. No significant difference (Student t test, p 〉 0.05) for the torsional failure force was measured for both type of coatings. Nevertheless, SEM revealed differently situated fracture planes. Light microscopy showed intimate bone-implant contact for both types of coatings; original drill margins were still visible. A lamellar type of bone with some remodeling lacunae was shown. Histomorphometry revealed a higher percentage of bone contact for the FA/HA-PS-coated implants (student t test, p 〈 0.05). Measurement of the amount of bone revealed more bone mass around TPS/Ca-P-a-coated implants (analysis of variance and Tukey multiple comparison, p 〈 0.05). © 1997 John Wiley & Sons, Inc.

    Notes: The objective of this study was to evaluate a new type of percutaneous device (PD) designed to be implanted in soft tissues. The new PD consisted out of: (1) a flange-shaped subcutaneous component, made from sintered titanium fiber-web, and (2) a percutaneous component, made from dense sintered hydroxyapatite. The PDs were inserted in the back of 15 rabbits. The surgical procedure was performed in two steps. In the first session the subcutaneous component was placed. In the second session, after 3-4 months the percutaneous component was fixed in the subcutaneous component. The implants were left in situ for 1 and 4 months after the second implantation session. Clinical and histological investigations were performed. It is found, that there was only a limited epidermal downgrowth in the percutaneous area. No inflammatory reaction was observed in the dermal connective tissue. Histological analysis also demonstrated that titanium fiber mesh evokes minor adverse effects of the surrounding tissues. In conclusion, these experiments have shown that stabilization of the PD in the hypodermal area by using a sintered titanium fiber-web structure favors the longevity of PDs implanted in soft tissues.

Therefore, coated and uncoated titanium implants were inserted into the tibia of rabbits. Implantation times were at 3 and 12 weeks. The histological evaluation included measurement of the amount of bone apposition to the various implant surfaces. The results demonstrated at 3 and 12 weeks no marked differences in bony reaction at the cortical level to the different implant materials. However, compared with the 3-week specimens, at 12 weeks extensive maturation of the woven bone callus had occurred. In addition, all 12-week implants induced bone formation in the medullary cavity. It was also noted that all HA-coatings showed loss of coating thickness. Quantitative determination of bone contact demonstrated that all 12-week implants showed the same high amount of bone apposition. © 1993 John Wiley & Sons, Inc.
