    Notes: The mechanical properties of sintered timing wheel in contact with chain wheels were analysed using Finite Element Methods(FEM), in which the timing wheel is modelled as a metal powder. The mechanical properties of sintered timing wheel wereinvestigated as a function of sintered density.Tensile strength and Young’s modulus increased with a decrease in porosity. Current methods of calculating gear contact stressesuse Hertz’s equations, which were originally derived for contact between sintered timing wheel and chain wheels. The results of the2D dimensional FEM analyses from ANSYS are presented.The relationship between relative density of P/M steels and mechanical behavior is also obtained from FEM and compared with theexperimental data. Good agreement between the experimental and FEM results is observed, which demonstrates that FEM cancapture the major features of the P/M steels behaviour during loading. This indicates that the FEM model is accurate

    Notes: The aim of this study was to determine immunogenetic markers of susceptibility in Crohn's disease (CD), taking the different features of the clinical course of the disease into account. HLA class I, HLA class II and TAP transporter gene polymorphisms were studied using DNA typing methods. Gene and antigen frequencies were analysed and compared in a group of 102 CD patients and 200 unrelated healthy controls from the same area. Analysis of the whole CD patient population revealed no definite association with either HLA or TAP gene alleles, with the exception of an association with DRB1*1302 (Pc 〈 0.05). However, when clinical subgroups of patients were considered, specific associations with some genetic markers were found. The most definitive results involved a genetic association in the group of patients who did not respond to glucocorticoid therapy. This group was characterized by a high frequency of HLA-DRB1*04 (P 〈 0.05). Conversely, a positive association with the TAP2-A allele was found in cortico-responder patients (Pc 〈 0.03). Furthermore, analysis of the distribution of HLA class II alleles in relation to the presence of extra-intestinal manifestations revealed an association with the DQB 1*0501 or *0503 suballele of DQ5 (P 〈 0.05). Finally, patients with lesions in the small bowel were more frequently HLA DRB1 *07 (P 〈 0.05). The present study supports the concept of clinical heterogeneity in Crohn's disease associated with a background of genetic heterogeneity.

    Notes: Abstract In this report, we establish that 3′-azido-3′-deoxythymidine (AZT) treatment of melanoma cells greatly alters the pattern of glycosphingolipid biosynthesis. In SK-MEL-30 cells, synthesis of the gangliosides GM3 and GD3 was significantly inhibited (60% and 50% of control, respectively) and the production of their precursor, lactosylceramide, was stimulated by 2.5-fold. Control experiments established that phospholipid synthesis was not affected by AZT treatment, consistent with AZT treatment only affecting lipid biosynthetic reactions that involve glycosylation. Likely as a consequence of decreased rates of ganglioside synthesis, AZT treatment of SK-MEL-30 cells also significantly suppressed the amount of gangliosides shed from the membranes of these cells. Since shedding of gangliosides has been proposed to allow melanoma cells to avoid destruction by the immune system and alterations of glycosphingolipid levels are likely important for the malignant cell phenotype, these results may have important implications regarding the potential use of AZT or related glycosylation inhibitors as cancer chemotherapeutics.
