    Notes: The construction and performance characteristics of a monolithic quad-pixel Ge detector designed specifically for fluorescence x-ray absorption spectroscopy (XAS) at synchrotron radiation sources is described. The detector semiconductor element has an active surface area of 4.0 cm2 that is electrically separated into four 1.0 cm2 pixels, with little interfacial dead volume. The spatial response of the array demonstrates that cross-talk between adjacent pixels is less than 10% for 5.9-keV photons that fall within 0.5 mm of the pixel boundaries. The detector electronics system utilizes preamplifiers built at LBNL with commercial Tennelec Model TC 244 amplifiers. Employing an 55Fe test source (Mn Kα , 5.9 keV), energy resolution of better than 200 eV is achieved with a 4 msec peaking time. At 0.5 msec peaking time, pulse pileup results in a 75% throughput efficiency for an incoming count rate of 100 kHz. Initial XAS fluorescence measurements at the beamline 4 wiggler end stations at SSRL show that the detector system has several advantages over commercially available x-ray spectrometers for low-concentration counting applications. © 1996 American Institute of Physics.

    Notes: A new crystal bender for dispersive X-ray absorption fine-structure spectroscopy (XAFS) has been built and its performance characterized. The bender employs an adjustable four-point bending mechanism and a contoured crystal, which makes it possible to produce an excellent approximation of the ideal elliptical bend from 8000 to 20000 eV with a single such Si (220) crystal. With the near-elliptical optics, it is possible to achieve a focus size of 175 μm with a 5 mm source and simultaneously to achieve a useful bandpass of 700 at 9000, 1200 at 11500 and 2000 eV at 18000 eV. The upper limit on the resolution, which is dictated by the finite source size in this configuration, is 2–5 eV for all of the energy ranges studied.

    Notes: The local structures of Am, Nd and Er-Benzimidazole (Biz) in solution were determined by EXAFS. The BIZ molecule coordinated to Am and Nd through two nitrogen atoms in a bidentate fashion. Two nitrogen atoms of BIZ ligated to Am and Nd with the bond distances RAm-N = 2.63 Å and RNd-N = 2.65 Å, respectively. The total coordination number of the Am BIZ complexes (at a molar ratio of metal ion to ligand of 1:20) was 1̃0 but that of Nd BIZ complex was 9̃.

    Notes: Summary The black Portuguese millipede, Ommatoiulus moreletii, an exotic species first reported in Australia in 1953, shows a pattern of initial eruption and subsequent decline in abundance following its introduction to sites in South Australia. Comparative sampling of new, erupted populations and older, declined populations was done in an attempt to find testable hypotheses to account for the decline. We report on laboratory and field experiments which show that a native rhabditid nematode appears to be the causal agent for the decline of populations of O. moreletii in South Australia. Implications for the biological control of introduced species are discussed in terms of this work.

    Notes: Unfertilized eggs of the sea urchin Arbacia punctulata contain pigment granules distributed throughout their cytoplasm. During the first 15 minutes after fertilization, these vesicles move out to the cortex where they become firmly anchored. We have used time-lapse video differential interference microscopy to analyze the motility of these organelles in unfertilized and fertilized Arbacia eggs. Pigment granules exhibit saltatory movement in both unfertilized and fertilized eggs. Quantitation of vesicle saltations before and after fertilization demonstrates that while there is no significant difference in the speed or pathlength of vesicle movement, there is a dramatic change in the orientation of these saltations. Saltations in the unfertilized egg are very non-radial and are as likely to be directed toward the cortex as away. In contrast, saltations in the fertilized egg are more radially oriented and more likely to be cortically directed. This transition must reflect underlying changes in the cellular structures necessary for pigment granule saltations. The change in the orientation of pigment granule saltations following fertilization requires both a transient increase in the cytoplasmic concentration of Ca2+ and an elevation of cytoplasmic pH. Similarly, the ability of pigment granules to adhere to the cortex requires both the transient elevation of cytoplasmic Ca2+ and the alkalinization of the cytoplasm. As the reorganization of cortical actin at fertilization is regulated by these ionic fluxes, and both movement and adhesion are sensitive to cytochalasins, we hypothesize that the alterations in directed motility and adhesion reflect underlying changes in the actin cytoskeleton.

    Notes: We have determined the activities, protein, and mRNA abundances as well as the level of transcriptional activation of two intracellular forms of the free radical metabolizing enzyme superoxide dismutase in 29 human skin fibroblast lines established from donors of different ages. SOD-1 (a copper and zinc containing from of SOD) and SOD-2 (a manganese containing form of the enzyme) activities were both observed to be significantly lower in cell lines derived from fetal skin than in lines established form postnatal skin (ages 17-94 years). The percent of total activity contributed by SOD-1 decreased in an age-associated manner from approximately 50% in the fetal lines to less than 20% in lines established from old tissue donors. All of the cell lines were screened to exclude the possibility that they contained a polymorphism known to influence SOD-2 activity. Northern blot analysis revealed three SOD-1 mRNA transcripts that were 0.5, 0.7, and 1.9 kb in length. Although SOD-1 protein abundance was lower in fetal lines than in lines derived from postnatal donors, SOD-1 mRNA abundance did not differ between fetal cells and cell lines derived from young donors. SOD-2 protein abundance and mRNA abundance were both significantly lower in fetal lines than in postnatal lines. No postnatal age-dependent differences were observed in any of the SOD-2 parameters examined. Nuclear run-on analysis revealed that fetal cell lines exhibited a lower level of transcriptional initiation for SOD-1 than postnatal lines. The transcription of SOD-2 was readily detected in postnatal lines, but undetectable in fetal lines. These results are consisten with multiple levels of control of SOD-1 expression and with a strong transcriptional influence on SOD-2 expression. © 1995 Wiley-Liss Inc.
