    Notes: Aim : To test whether α-methylacyl-CoA racemase (AMACR) is a sensitive and specific marker of prostate cancer.Methods and results : The expression levels of AMACR mRNA were measured by real-time polymerase chain reaction. A total of 807 prostatic specimens were further examined by immunohistochemistry specific for AMACR. Quantitative immunostaining analyses were carried out by using the ChromaVision Automated Cellular Imaging System and the Ariol SL-50 Imaging System, respectively. AMACR mRNA levels measured in prostatic adenocarcinoma were 55 times higher than those in benign prostate tissue. Of 454 cases of prostatic adenocarcinoma, 441 were positive for AMACR, while 254 of 277 cases of benign prostate were negative for AMACR. The sensitivity and specificity of AMACR immunodetection of prostatic adenocarcinomas were 97% and 92%, respectively. Both positive and negative predictive values were 95%. By automatic imaging analyses, the AMACR immunostaining intensity and percentage in prostatic adenocarcinomas were also significantly higher than those in benign prostatic tissue (105.9 versus 16.1 for intensity, 45.7% versus 0.02% and 35.03% versus 4.64% for percentage, respectively).Conclusions : We have demonstrated the promising features of AMACR as a biomarker for prostate cancer in this large series and the potential to develop automated quantitative diagnostic tests.

    Notes: Aims:  There is no well-established positive immunomarker for urothelial carcinoma. We evaluated the diagnostic utility of high molecular weight cytokeratin (HMWCK) antibody clone 34βE12 in differentiating high-grade invasive urothelial carcinoma from prostate cancer.Methods and results:  Formalin-fixed paraffin-embedded sections from 28 cases of high-grade invasive urothelial carcinoma (20 not otherwise specified (UC-NOS), eight with glandular differentiation) and 20 cases of poorly differentiated prostate carcinoma were immunostained with a monoclonal antibody to carcinoembryonic antigen (CEA), clone 85A12 and with HMWCK antibody clone 34βE12 after microwave pretreatment or protease 24 predigestion. All cases of UC-NOS expressed HMWCK on 34βE12 immunostaining after microwaving or enzyme predigestion. Immunoreactivity was intense and diffuse in all the cases after microwave pretreatment, whilst with enzyme predigestion immunoreactivity was sometimes patchy with 〈50% tumour cells positive in 20% of cases. In comparison with 34βE12, 85A12 was insensitive with 15% of UC-NOS cases totally CEA-negative and 〈50% tumour cell immunoreactivity in 60% of cases. Rare positive cells were present in two (10%) cases of prostate cancer with monoclonal anti-CEA and 34βE12 on microwaved sections, but all the cases were HMWCK-negative using 34βE12 on sections pretreated by enzyme digestion.Conclusions:  HMWCK antibody clone 34βE12, particularly when used with microwave heat retrieval, is a very sensitive positive marker for high-grade invasive urothelial carcinoma.

    Notes: Summary Poly(vinyl oxazoline ester) and poly(stearyl methacrylate) were found to be good alien polymers for grafting of methyl methacrylate by suspension polymerization whereas poly(tridecyl methacrylate) was not a good backbone. Azobisisobutyronitrile was found to be an inefficient grafting initiator where azocumene that produces highly resonance and hyperconjugated stabilized dimethyl phenyl methyl radical was a superior initiator to benzoyl peroxide for grafting. Poly(stearyl methacrylate) was found to be a suitable polymeric stabilizer for nonaqueous suspension polymerizations.

    Notes: Empirical equations have been developed to predict the mechanical properties of greenhouse films when exposed to natural weather conditions for long durations. Tensile strength and percent elongation data obtained for the Dhahran site was used to develop empirical equations. Prediction models were developed with exposure time as independent parameter for both tensile strength and percent elongation, which were taken as dependent parameter. The mean deviations for tensile strength and percent elongation were found to be 2.5 and 1.2%, respectively. Weather data of Dhahran for the period of natural exposure is also presented. © 1995 John Wiley & Sons, Inc.

    Notes: Natural and artificially accelerated weathering trials were carried out on low-density polyethylene samples used for agricultural and disposable purposes. Modifications in polymer characteristics were studied using Fourier transform infrared (FTIR) spectroscopy, different scanning calorimetry (DSC), gel permeation chromatography (GPC), scanning electron microscopy (SEM), and mechanical properties testing. The natural exposure trials were conducted at five geographical locations representing varying ambient temperature and humidity conditions and receiving different doses of solar radiation. The artificially accelerated exposure trials were carried out in an Atlas weatherometer for 5,000 hours, whereas the natural exposure was for 24 months. Rates of photo-oxidative and thermal degradation have been determined by measuring the formation of non-volatile carbonyl oxidation products which absorb in the infrared region with a maximum absorbance level at 1710cm-1. Thermal characteristics were noticed to vary in terms of the shape and size of melting peak for both the natural and artificial weathering. This implies a change in crystallinity, which has indicated an increasing trend with exposure time. The crystalline melting temperature (Tm) remains almost steady for both exposure tests. The drop in mechanical properties and the modification in polymer chain length were also monitored. The GPC analysis indicated the change in molecular weight distribution (MWD), which shows the formation of lower molecular-weight species. Surface modifications were revealed in terms of abrasion effects apparent from SEM micrographs. A correlation between natural and artificial weathering was considered for lifetime prediction in a short exposure time. It was found that the confidence level of predicting lifetime on the basis of artificially accelerated exposure trials is dependent on many parameters, which include time, material, equipment, etc. © 1995 John Wiley & Sons, Inc.

    Notes: Photo-oxidative degradation reactions are mainly responsible for reduced lifetime in polymers. These reactions are responsible for the decay in useful physical and chemical properties of polymers. In addition to high ultraviolet radiation doses, hostile environments have increased temperature, high humidity, and air-borne sand. In this work white pigmented polyvinyl chloride (PVC) pipe sections were exposed to natural weather at seven different locations. Characterization studies were made on exposed samples using Fourier transform infrared (FTIR) spectroscopic, gel permeation chromatographic (GPC), and scanning electron microscopic (SEM) studies. Performance characteristics were determined by testing changes in the mechanical properties. Functional groups introduced during the processing of PVC and irregularities in the polymer chain can be held responsible for the initiation and propagation of photo-oxidative degradation reactions catalyzed by high surface temperature and increased humidity.
