    Notes: Abstract Anaerobic dehalogenation of aromatic compounds is a well-documented phenomenon. However, the effects of operating parameters such as pH have received little attention despite their potential impact on treatment processes using dehalogenating organisms. In this work the effect of pH on the dehalogenation of 2,4,6-trichlorophenol (2,4,6-TCP) was studied using defined media containing one of several non-fermentable buffering agents (MOPS, TRICINE, BICINE, CHES), and no chloride ions. The dechlorination process was followed by monitoring the disappearance of 2,4,6-TCP, as well as the appearance of its dehalogenation products, i.e., 2,4-dichlorophenol (2,4-DCP), 4-chlorophenol (4-CP), and chloride ions. The results indicate that dechlorination occurs only if the pH is within the range 8.0–8.8. The newly formed 2,4-DCP was also dehalogenated in the process. However, even within this pH range dechlorination ceased when all 2,4,6-TCP and 2,4-DCP was converted to 4-CP. Stoichiometric amounts of all dehalogenation products (including chloride) could be recovered at any stage during the process. In addition, the biomass concentration was measured. After an initial lag phase, it appeared that the rate of dechlorination per unit biomass (proportional to the Cl− concentration divided by the biomass concentration) went through a rapid increase and then remained constant throughout the process. This indicates that the dechlorinating organism(s) either make up the entire population or constitute a stable fraction of it.

    Notes: A laser-Doppler velocimetry (LDV) apparatus and a computational fluid dynamic (CFD) software package (FLUENT) were used to experimentally determine and numerically predict the velocities in a baffled vessel agitated by one or two 45° pitched-blade turbines. The flow characteristics in the impeller regions were measured by LDV and used as boundary conditions in the numerical computations. Turbulence effects were simulated using either the k-ε model or algebraic stress model (ASM). The CFD predictions were compared to the LDV measurements in terms of average velocities in all three directions as well as turbulent kinetic energies. Predictions based on ASM were typically in closer agreement with the experimental data than those based on the k-ε model. Flow patterns in both configurations were dominated by the axial and tangential components. The presence of the upper impeller altered the flow considerably, producing a strong vertical recirculation pattern between the impellers and significantly reducing the circulation flow below the lower impeller.

    Notes: The Monod or Andrews kinetic parameters describing the growth of Pseudomonas sp. CPE2 strain on 2,5-dich!orobenzoic acid and 2-chlorobenzoic acid, and Al-caligenes sp. CPE3 strain on 3,4-dichlorobenzoic acid, 4-chlorobenzoic acid, and 3-chlorobenzoic acid were determined from batch and continuous growth experiments conducted in the presence or absence of yeast extract (50 mg/L). Strain CPE2 displayed inhibitory growth kinetics in the absence of yeast extract and a noninhibitory kinetics in the presence of yeast extract. Similar results were obtained for CPE3. The presence of yeast extract also resulted in a significant increase in the affinity of the strains for the chlorobenzoic acids they degraded. © 1995 John Wiley & Sons, Inc.

    Notes: An improved mathematical model was developed to design a continuous sterilizer for liquid fermentation media containing suspended solids. Unsteady-state energy balances were used to determine the temperature distribution in the liquid medium and in the solid particles as a function of time and position within the particle, as the medium flows through the sterilizer. Such temperature profiles were used to determine the level of microbial reduction achieved by each component of the sterilizer and by the entire sterilization process. A considerable difference exists between the temperature in the particle core and in the surrounding liquid. This has a significant impact on the degree of sterility achieved by the process. The level of microbial reduction in the particles was found to be tens and or even hundreds of order of magnitude lower than the corresponding level achieved in the liquid. The role of the particle material on the degree of sterilization was also investigated. Solid materials typically found in fermentation media, such as wood or flour clumps were found to offer considerable resistance to the sterilization of the organisms lodged inside them. © 1993 Wiley & Sons, Inc.

    Notes: The biodegradation of 2,4,6-trichlorophenol and 2,4,5-trichlorophenol by the white rot fungus Phanerochaete chrysosporium was studied in batch and continuous reactor systems. Experiments were conducted in shake flasks as well as in packed-bed reactors in which the fungus was immobilized. The degradation rates in the packed-bed reactors were found to be two orders of magnitude greater than those obtained in the shake flasks in which the fungus was just suspended. The degradation rate was found to be influenced by the concentrations of the carbon and nitrogen sources, pH, and fluid shear stress. Optimal ranges of these parameters to maximize biodegradation were determined. A mathematical model was developed in which the degradation process was assumed to consist of two sequential reaction steps, the first catalyzed by an extracellular enzyme system and the second requiring the presence of the mycelium. The deactivation of the extracellular enzyme system was also accounted for in the model. The Michaelis-Menten and the enzyme deactivation parameters were determined independently. Good agreement between the experimental data and the results produced by the regression was found. © 1995 John Wiley & Sons, Inc.
