    Notes: Abstract We have isolated and analysed a 2 kb region of the mitochondrial genome of Arabidopsis thaliana (Columbia) showing a high level of nucleotide identity with the mitochondrial (mt) rps14 small-subunit ribosomal protein gene from Oenothera berteriana and Vicia faba, as well as with an open reading frame (ORF) located upstream of the nad3 locus in O. berteriana. The rps14 locus is present as a single copy in the A. thaliana mt genome and has a translational stop codon located near the initiation codon, as well as a deletion of one nucleotide that disturbs the coding sequence. The cloning and sequencing of nine amplified mt rps14 cDNAs clearly demonstrated that this gene is transcribed and that the mRNA precursors are edited at three positions, all involving C-to-U conversions. No editing events changing the stop codon and restoring the correct coding sequence were witnessed within the 9 individual cDNA clones. Therefore, we conclude that the single rps14 sequence of the mitochondrial genome from A. thealiana is in fact a pseudogene that is transcribed and edited but not translated.

    Notes: Abstract The diagnosis ofToxoplasma gondii infection is currently based on immunological tests, but tests for IgG and IgM antibodies alone are often insufficient to assess the risk of active disease, especially during pregnancy and in immunodeficient subjects. The supplementary diagnostic value of testing for antitoxoplasmic IgA in cases of acute, chronic, congenital and reactivated toxoplasmosis, relative to classical immunological tests, was evaluated using two immunocapture tests, one based on tachyzoite agglutination and the other on an immunoenzymatic complex recognizing the membrane protein P30 ofToxoplasma gondii. A total of 4,541 sera from 395 uninfected subjects, 468 immunized subjects with chronic infection, 117 subjects with acute infection and 403 children, 103 of whom had congenital toxoplasmosis, was tested. Specific IgA tests were negative in the nonimmune population, but tests for this immunoglobulin subtype became positive very rapidly during primary infection, and IgA disappeared more rapidly than IgM. In the children infected in utero, specific IgA was detected more frequently than IgM. In contrast, in a population of HIV-seropositive subjects with clinical toxoplasmosis, tests for IgA were poorly sensitive. The two tests for specific IgA produced similar results, except in the early stages of primary infection, in which immunoenzymatic testing for anti-P30 IgA was less sensitive than the agglutination method.

    Notes: Abstract Although the involvement of tumor necrosis factor-α (TNF-α) during Toxoplasma gondii infection has largely been described in mouse models, only a few studies in human models have been reported. We demonstrated the role of TNF-α during the process of invasion by T. gondii in human monocytic cells. Cotreatment of cells with interferon-γ (IFN-γ) and lipolysaccharide (LPS) during T. gondii infection induced TNF-α production and decreased the number of parasitized cells (invasion index). Neutralization of the production of TNF-α using a specific monoclonal antibody or pentoxifylline enhanced the invasion index. The relationship between TNF-α production and protection of monocytic cells against T. gondii invasion was confirmed by treatment of infected cultures with exogenous TNF-α. Thus, in contrast to the results obtained in murine models but in accordance with those observed in other human models, the present study shows for the first time in human monocytic cells that T. gondii does not induce any TNF-α secretion and inhibits TNF-α production induced by IFN-γ/LPS.

    Description / Table of Contents: Zusammenfassung Wir haben ein Schnellprotokoll zur Geschlechtsbestimmung forensischer Proben mit Hilfe der „Polymerase Chain Reaction” etabliert. Hierzu werden 3 Primer-Paare verwendet; 2 hiervon sind spezifisch für repetitive Sequenzen auf dem Y-Chromosom, ein Paar ist spezifisch für repetitive Sequenzen auf dem X-Chromosom. Die Methode wird detailliert beschrieben, ebenfalls die durchgeführten Kontrollen. Die Anwendungsmöglichkeiten an Blutflecken und Vaginalabstrichen werden vorgestellt. Erwähnung findet auch die Empfindlichkeit des Tests und die Problematik, welche durch Verunreinigungen der Spur entsteht.
    Notes: Summary A rapid protocol has been established for sexing forensic samples by the Polymerase Chain Reaction method. Three sets of primer were used, two specific for Y chromosome repetitive sequences and one specific for X chromosome repetitive sequences. Detailed procedures of experiments, the controls and the applications to testing bloodstains and a vaginal swab are presented. The sensitivity of the test and problems due to contamination are discussed.

    Description / Table of Contents: Zusammenfassung Viele Geologen und Geographen betrachten den Jura als den Typus eines Faltengebirges mit regelmäßig hintereinander gestaffelten Synklinalen und Antiklinalen in der Form von Sinusoiden, obwohl bereits frühere Forschungen — in der bewundernswerten Bibliographie vonEmm. de Margerie (44) zusammengefaßt — ein viel naturnäheres Bild gaben. Die Untersuchungen der letzten 10 Jahre haben dieses Bild vertieft und namentlich die Vorgeschichte der Faltung als besonders wichtig hervorgehoben. Verschiedene Gruppen von Brüchen, die sich schon vor der Faltung bewegten, haben einen großen Einfluß auf Anordnung und Gestalt der Falten. Die Ergebnisse der Untersuchungen werden nach den tektonischen Erscheinungen, von den Querverschiebungen und den Randverwerfungen des Rheintales bis zum Alter der Faltung, zusammengefaßt und gruppiert. Das naturnähere Baubild, besonders aber die neueren Anschauungen über die Entstehung der Alpen, bedingen auch neue Deutungen des jurassischen Bewegungsbildes. Die Rolle des kristallinen Sockels ist zur Zeit eines der interessantesten Probleme. Geophysikalische Untersuchungen sind zu seiner Lösung notwendig.
    Notes: Abstract For most geologists and physiographers the Jura Mountains are the prototype of a folded chain with very regular anticlines and synclines. The more detailed picture based on the research of the last years is somewhat different. Faults older than the folding play an important rôle. The resulting structural phenomena and the different modern interpretations are exposed. A general survey of the earlier literature is found in the admirable monography of the Jura Mountains by EMM.     Notes: Abstract A case of cystic rectal duplication revealed on day 2 of life by a low intestinal occluding syndrome is reported. Radiologic imaging (ultrasonography, cystography, rectography) showed a large, retrorectal liquid formation in the pelvis and abdomen, with pelvic compression of the terminal alimentary canal and lower urinary tract. Magnetic res‐onance imaging demonstrated a liq‐uid formation with clearly defined edges and no medullary involvement, thus ruling out the possibility of a previous meningeal hernia. Biological markers were within normal limits. On day 4, a 9 × 6-cm cystic rectal duplication was removed, followed by a temporary colostomy. Pathologic examination demonstrated typical rectal architecture with ciliated cells. Radiologic and clinical findings at 2-month follow-up were reassuring. This case report is exceptional for the following reasons: (1) As a rule, rectal duplications are relatively rare (70 cases reported in the literature); (2) The means of disclosing a neonatal rectal duplication is unusual (4 cases reported in the literature); (3) The volume of the malformation was considerable; and (4) Heterotopic ciliated epithelium was present.
