    Notes: Summary Blastocysts were recovered from mice in experimentally delayed implantation 16 hrs after an injection of estrogen. When obtained from uterine horns with Pontamine Blue-positive sites, the blastocysts possessed a smooth embryonic pole and a rough abembryonic one as seen with SEM. In the embryonic pole, most of the trophoblast cells were flat while in the abembryonic region, the cells showed irregular cell extensions and rough imprints. TEM of Pontamine Blue-positive sites demonstrated that the embryonic pole had a rather smooth borderline to the epithelium, while the borderline at the abembryonic pole was more irregular with small parts of the uterine epithelium indenting into the trophoblast. The cytoplasm of the trophoblast contained a well developed endoplasmic reticulum, several mitochondria, large vacuoles, many multivesicular bodies and lipid granules. The difference in structure between the two poles of the blastocyst is presumably related to the differences in enzymatic activity between the poles as demonstrated by histochemistry and to the greater invasive capacity of the abembryonic trophoblast.

    Notes: Amniotic fluid cells provide important information about the fetus. Studies of the surface ultrastructure of such cells identified four cell populations in midgestational fluids. A parallel study of tissue surfaces in contact with or communicating with the amniotic fluid showed that some tissues made no contribution to the cell populations, while others contributed extensively. Two principal cell shedding mechanisms were found in mid-trimester—exfoliation of whole cells and detachment of cellular fragments. The latter mechanism was observed from skin and umbilical cords during a limited period in mid-pregnancy. The majority of cells were derived from exfoliation of squamous epithelia.

    Notes: Mononuclear cells cause disappearance of fibronectin synthesized and released by fibroblasts. This disappearance of extracellular fibroblast fibronectin is accompanied by the appearance of components of “lower” molecular weight indicating that a fibronectin-degrading enzymatic activity is responsible for the effect. Additional support for the existence of a fibronectin-degrading enzyme is that mononuclear cells degrade iodinated fibronectin attached to a collagen matrix, Furthermore, lymphocytes seem to mediale the degradation of fibronectin whereas monocytes rather act inhibitory.

    Notes: Borrelia hermsii, an agent of replapsing fever, avoids the host's immune response by means of multiphasic antigenic variation. Serotype specificity is determined by variable antigens called the Vmp lipoproteins. Through recombination between linear plasmids a formerly silent vmp gene replaces another vmp gene at a telomeric expression locus. We examined strain HS1 borreliae before and after a switch from serotype 7 to serotype 21. The nucleotide sequences of 5 regions of silent and expressed vmp7 and vmp 21 were determined. Silent and active vmp 7 and vmp 21 genes shared a block of homologous sequences surrounding their 5′ ends. Sequences upstream of silent vmp 7 and vmp 21 genes lacked the promoter and substantially differed from each other. In this antigenic switch a vmp gene was activated by a recombination that placed it downstream of a promoter.

    Notes: The relapsing fever agent Borrelia hermsii avoids the host's immune response by the strategy of multiphasic antigenic variation. A given Borrelia cell can express one of a number of alleles for polymorphic outer-membrane proteins, known as Vmp proteins. The genes for the variant-specific Vmp proteins of serotypes 7 and 21 of B. hermsii strain HS1 were sequenced. The genes, which were designated vmp7 and vmp21, were obtained from populations of borreliae before and after a switch in serotypes from 7 to 21. The analysis showed that vmp7 and vmp21 are 77% identical in terms of their coding sequence. The deduced translation products of vmp7 and vmp21 are polypeptides of 369 (37.2kD) and 364 amino acids (37.1kD), respectively. Vmp7 and Vmp21 have sequence features of prokaryotic lipoproteins and are processed as such during expression in E. coli. The secondary structure predictions of the Vmp proteins reveals analogous structures to the VSG proteins of the African trypanosome.

    Notes: The ospA and ospB genes encode the major outer membrane proteins of the Lyme disease spirochaete Borrelia burgdorferi. The deduced translation products from the ospA and ospB genes were: (OspA) 273 amino acids long with a molecular weight of 29334, and (OspB) 296 amino acids long with a molecular weight of 31739. The two Osp proteins showed a great degree of sequence similarity indicating a recent evolutionary event. Molecular analysis and sequence comparison of OspA and OspB with other proteins revealed a sequence similarity to the signal peptides of prokaryotic lipoproteins. These are the first sequences from Borrelia and provide interesting data on the evolutionary relationship between spirochaetes and other species as well as providing potential for spirochaete diagnostics and vaccines.
