    Notes: Summary Three sequential chromatographic equipments are in operation at the University of Aston fractionating mixtures ranging from polymeric materials using continuous gel permeation techniques through to high boiling organic mixtures such as fatty acids and essential oils. This paper describes in detail the performance characteristics of one of these equipments made up of twelve columns, 61 cm long and 7.6 cm diameter that has been successfully used to separate halocarbon mixtures at feed rate up to 1 litre per hour. The experimental results obtained with the equipment have been used to substantiate some theoretical developments proposed in the paper.

    Notes: Abstract Human low-molecular-weight (2′, 5′)A synthetase is induced in certain human x mouse somatic hybrid cell lines when these cells are treated with mouse interferon. We have assigned the gene coding for this interferon-inducible antiviral enzyme to human chromosome 11 by somatic cell genetic techniques (1). Fluorescence-activated cell sorting for cells expressing or lacking 4F2 antigen in two independently derived, chromosome 11-containing hybrid cell lines separated the cells into subpopulations of cells that had retained or segregated chromosome 11, respectively (2). We used these subpopulations to confirm our gene assignment by demonstrating that retention of chromosome 11 was required for expression of human (2′, 5′) A synthetase.

    Notes: Abstract The human thyroglobulin structural gene (TG) was mapped to the long arm of chromosome 8 by blot hydridization of a TG cDNA probe to DNA from 21 human × mouse somatic cell hybrids containing overlapping subsets of human chromosomes. In situ hybridization of the TG probe to metaphase chromosomes from a karyotypically normal human lymphoblastoid cell line, JS, localized the TG gene to within the region 8q23 → q24.3. Thus, the TG and c-myc genes map to the same chromosome band in normal human cells. In a human colon carcinoma cell line (COLO 320 DM) which contains amplified c-myc, the TG gene is not amplified and hence it lies outside the amplification domain.

    Notes: High yields of the enzyme dextransucrase have been produced repeatedly by fed-batch fermentation techniques. Activities in excess of 21.9 U/cm3 have been obtained by culturing Leuconostoc mesenteroides NRRL B-512(F) under nonaerated fed-batch fermentation conditions. Aerobic fermentations carried out under identical conditions have consistently produced enzyme of less than 17 U/cm3, but with no difference in the final cell concentration in the broth. Different types of yeast extract have been found to have significant effect on the final cell concentration and more especially on the enzyme activity with enzyme yields varying by as much as 50% when different types of yeast extracts were used.

    Notes: Summary A sequential continuous chromatographic refiner (SCCR-2) for preparative-scale GLC up to 200°C is described. The separating capabilities and other characteristics of the SCCR-2 unit have been investigated using fatty acid ester mixtures of different separation difficulty and volatility. The feed mixtures selected had separation factors (α) between 1.1 and 1.5 and required operating temperatures between 110 and 200°C, while using FFAP (free fatty acid phase) on Chromosorb W or OV-275 (a cyanosilicone) on Chromosorb P. Initially the separation of a 50/50 v/v mixture of methyl chloroacetate/ethyl lactate (α≃1.5) was studied and the ability of the SCCR-2 unit to separate the mixture into two products with purities in excess of 99.8% has been demonstrated at feedrates up to 80 cm3h−1 and temperatures between 110 and 130°C. For the more difficult system ethyl chloroacetate/ethyl lactate (α≃1.2) the column was too short for successful separation, although purities of about 95% for one product stream were obtained at feedrates of 33 cm3 h−1 at 125°C. Preliminary studies on the recovery of γ-linolenic acid from fungal oil, at temperatures of 200°C are reported. Although this equipment has been demonstrated on the laboratory scale it is amenable to scale up to production levels. Partition coefficient data for several fatty acid solutes were determined using a comerical analytical scale GLC and some of the most favoured stationary phases for the GLC of fatty acids.

    Notes: Summary The construction and operation of a liquid preparativescale continuous chromatograph is described. The chromatographic bed is arranged in a series of interconnected vertical columns and is rotated whilst one end of the tube bundle is closed by a stationary plate through which liquids enter and leave the bed. The problem of maintaining an efficient seal is described. The chromatograph was commissioned by continuously fractionating dextran on porous silica beads with water as eluent. Results show that as the feed concentration was increased, the fractionation obtained deviated markedly from that expected from basic GPC theory, but that the relative molecular mass distribution of the feed was still narrowed with feed concentrations of 20% w/v.

    Notes: Summary A study has been made of the temperature and related solute concentrations in a 12 column (7.6 cm OD×61 cm) sequential continuous chromatograph at feed rates of up to 1400 cm3hr−1 of Arklone P-Genklene P. Moving the feed point from the middle of a column to the end cone was found to be particularly beneficial. Besides increasing throughput it enabled the system Arklone P and dichloromethane (separation factor 1.17) to be separated, not previously possible with a central feed point.

    Notes: Summary A semi-continuous, counter-current chromatographic refiner (SCCR) was packed with an anion resin in the bisulphite form and used to separate mixtures of sugars from different feedstocks. Products with purities of more than 99% were achieved and solid concentrations of the fructose products of 12% w/v were possible. The deterioration of the anion resin was controlled by simple precautions and thus its useful life was substantially prolonged.
