    Notes: Abstract  An HPLC procedure is described which enables both identification and quantitation of chlorophyll a and chlorophyll b and their major degradation products which can occur during solvent extraction of chloropigments from green plant leaves or during storage and canning of green vegetables. The method has been applied to visualize enzymatic as well as chemical degradation of chloropigments from spinach. Complex mixtures of phytyl esters of chlorophyllides, pheophorbides, pyrochlorophyllides, and pyropheophorbides and their respective hydrolysis products were separated, identified and quantitated in the presence of carotenoids in a process lasting less than 40 min.

    Notes: Abstract The incubation of 13-hydroperoxy-9Z,11E,15Z-octadecatrienoic acid (13-HPOT) with a hydroperoxide lyase containing extract of mung bean seedlings (Phaseolus radiatus L.) led not only to the formation of 2E-hexenal, but also to the generation of several non-volatile by-products (oxylipins). These oxylipins, generated by the catalysis of other 13-HPOT metabolizing enzymes, were analyzed by high performance liquid chromatography (HPLC) without time-comsuming derivatization procedures, which would be necessary for their volatilization and stabilization during gaschromatography (GC). Different detection systems such as an evaporative light-scattering detector (ELSD), diode-array detector (DAD), and particle beam-mass spectrometry (PB-MS) were applied. The utilization of an ELSD represented an improvement of sensitivity compared to a DAD, especially in the case of substances with low UV-activity.

    Description / Table of Contents: Abstract The mass balance of volatile flavours of the industrial processing of strawberry juice to juice concentrate and aroma concentrate was determined. Thirty-eight volatile indicator compounds were quantified. Conventional fractional distillation (A), when compared with a novel, three-stage plate condenser/scrubber (B), yielded the sensorially and analytically preferred aroma concentrate. Variant A transferred approx 26% (w/w) of the recorded volatiles into the aroma concentrate, whereas variant B recovered approx 15% (w/w). The calculated overall processing loss including the singlings was 66.7% (A), and 79.2% (B) of the genuine aroma pool, respectively.
    Notes: Zusammenfassung Die Massenbilanz der Aromastoffe bei der industriellen Verarbeitung von Erdbeersaft zu Saftkonzentrat und Aromakonzentrat wurde erstellt. Achtunddreißig flüchtige Indikatorsubstanzen wurden quantifiziert. Das konventionelle Rektifikationsverfahren (A) lieferte im Vergleich zu einem neuartigen, dreistufigen Plattenkondensator/Gaswäscher (B) das sensorisch und analytisch bevorzugte Aromakonzentrat. Verfahren (A) überführte ca. 26% der erfaßten Aromastoffe in das Aromakonzentrat; Verfahren (B) ca. 15%. Das rechnerische Prozeßdefizit einschließlich der Lutterwässer betrug 66,7% (A) bzw. 79,2% (B) des genuinen Aromabestandes.

    Notes: Abstract Many hospitals today have implemented widely disparate information systems on mainframe and mini-computer hardware. The advent of network technology in hospitals has made it possible to access information in these systems. Unfortunately, the user interfaces to applications on these systems are unique and difficult to learn, which makes them unsuitable for use by clinical services. In this paper we describe the development of a Physician's Workstation which integrates information from multiple existing information systems and discuss how the workstation makes it possible to move from the departmental systems of the present to the computer-based medical record system of the future.

    Notes: Abstract Three out of five cultivars of citrus species tested convert exogenous valencene via the 2-hydroxy-derivative (nootkatol) to nootkatone. The effect of various valencene concentrations and the time course of the biotransformation were examined. The transformation capability of the cells runs parallel with growth up to the middle of the logarithmic phase and remains constant until the carbon source is completely exhausted.

    Notes: Abstract The synthesis of thioethyl 2-methylpropanoate, butanoate, 3-methylbutanoate, hexanoate, and of thiobutyl propanoate, butanoate, and pentanoate was achieved via esterification of ethanethiol or butanethiol with short-chain acids using an immobilised lipase from Rhizomucor miehei. High acid conversions were obtained (47% for thiobutyl pentanoate). The analysis of educts and products was carried out by reverse-phase liquid chromatography (HPLC), gas chromatography (GC), GC/mass spectrometry, and GC/olfactometry. All of the thioesters imparted a similar, onion-like smell, which remained unchanged on dilution. The thresholds of thioesters derived from the same thiol were loosely correlated with the size of the acid moiety: the larger the molecular mass the higher the threshold. Surprisingly, higher odour thresholds were obtained for the branched-chain thioesters than for their linear analogues.

    Notes: Summary The volatile constituents of Pleurotus euosmus, grown on synthetic liquid medium, have been enriched and analysed by gas chromatography, sniffing analysis and coupled gas chromatography-mass spectrometry. Linalool and coumarin were identified as main compounds (900 resp. 2460/ug/l), and were found to be responsible for the intensive sweet flowery odor of the cultures.

    Notes: Abstract Pellets of Tyromyces sambuceus were cultured in standard liquid media that contained a natural triacylglycerol as an additional carbon source. Parallel to a drop of pH below 2.0 and the formation of R(+)-4-decanolide, the oil droplets disappeared. Assays based on 3-naphthyl ester showed that most of the lipolytic and esterolytic enzyme activities were membrane-associated. The time course of enzyme activities after addition of castor oil suggested an inducibility of both membrane-bound and intracellular lipases, while only the intracellular esterases responded significantly. The results could contribute to improving the biotechnological generation of lipid-derived flavours and other compounds.

    Notes: Abstract The biotechnological generation of natural aroma compounds is rapidly expanding. Aroma chemicals, such as vanillin, benzaldehyde (bitter almond, cherry) and 4-(R)-decanolide (fruity–fatty) are marketed on a scale of several thousand tons per year. Their possible production by single-step biotransformations, bioconversions and de novo synthesis using microorganisms, plant cells or isolated enzymes is shown. The perspectives of bioprocesses for the oxifunctionalisation of lower terpenes by genetically modified organisms and economic aspects are discussed.

    Notes: Abstract The basidiomycete Cystoderma carcharias transformed citronellol into 3,7-dimethyl-1,6,7-octanetriol as the main product. 3,7-Dimethyl-6,7-epoxy-1-octanol was identified as important intermediary product of the biotransformation, and the allylic diols 2,6-dimethyl-2-octene-1,8-diol, 3,7-dimethyl-5-octene-1,7-diol and 3,7-dimethyl-7-octene-1,6-diol were found to be minor products. Microbial formation of rose oxide, a flavour-impact component, was observed for the first time. The formation of the main products was inhibited by 70% after addition of 0.1 mmol l−1 cytochrome monooxygenase inhibitors. Formation of 3,7-dimethyl-1,6,7-octanetriol was effective in a bioreactor with aeration over a coil of a hydrophobic microporous polypropene capillary membrane. Production rates of up to 150 mg l−1 day−1 were reached and led to a product concentration of 866 mg l−1 (conversion rate: 52%). The total loss of the added volatile substrate via the exhaust air was 4.5% when this aeration method was used.
