    Notes: Two types of molecular genetic markers were used for genetic identification of species and local stocks of palearctic coregonids (Coregonidae, Salmoniformes, Teleostei). Seven nominate species of whitefishes and ciscoes Coregonus, spp. of Eurasia Arctic Sea basin and inconnu Stenodus leucichthys nelma, represented by specimens from North America were studied. Using restriction analysis of PCR-amplified products of the ND-1 gene of mitochondrial DNA (mtDNA) and allelic composition at several allozyme loci discrimination was successful between C. lavaretus pidschian, Siberian whitefish, C. nasus, broad whitefish, C. autumnalis, Arctic cisco, C. migratorius, Baikal omul, C. peled, peled, and C. sardinella, least cisco. Muksun C. muksun, was indistinguishable from Siberian whitefish. Creatine kinase (CK) isozyme patterns and Rsa, I restriction patterns of ND-1 were the most effective markers allowing discrimination among species. Intra-specific differentiation in mtDNA was found in all species but was much less pronounced than inter-species variation. In several specimens composite haplotypes typical of another species were found that reflect probable gene introgression by hybridization. A combination of mtDNA and nuclear genetic markers is suggested for reliable identification of both typical species representatives and hybrids.

    Notes: Abstract. During the winter of 1993–1994, male lesser scaup (Aythya affinis) were collected on the heavily polluted Indiana Harbor Canal, East Chicago, Indiana, and examined for several bioindicators of chemical exposure. Livers were analyzed for activities of three cytochrome P450–associated monooxygenases and four measures of oxidative stress. Blood and spleen were analyzed by flow cytometry for chromosomal damage. In a concurrent study, scaup tissues were analyzed for organic and inorganic contaminants. Ethoxyresorufin-O-dealkylase (EROD) activity in livers of scaup collected in January 1994 was significantly higher than in livers of scaup collected in March 1994 or in livers of reference birds. Three hepatic monooxygenase activities were each significantly correlated with polycyclic aromatic hydrocarbon (PAH) concentrations in scaup carcasses. Thiobarbituric acid (TBA) activity in scaup livers was positively correlated with iron, boron, and lead concentrations in livers and polychlorinated biphenyl concentrations in carcasses. TBA activity was negatively correlated with protein-bound thiol activity and mercury concentrations in livers. The coefficient of variation of DNA content in scaup blood cells was correlated with PAH concentrations in scaup carcasses. This is the first field study with birds to demonstrate a correlation between liver monooxygenase activity and carcass PAH concentrations and to show a direct correlation between PAH concentrations in tissues and somatic chromosomal damage in blood.

    Notes: Abstract The flow cytometry method (FCM) was employed to determine cellular DNA content of black-crowned night-heron (Nycticorax nycticorax) embryos and 10-day-old chicks collected at sites differing in types of chemical contamination. The coefficient of variation of DNA content (CV) in blood collected from embryos suggested cytogenetic damage at a site in Louisiana known to be contaminated with petroleum. Blood CV from chicks suggested genetic damage at a site in Texas also known to be contaminated with petroleum. Spleen CVs in chicks were significantly lower than respective means from the reference site. The CVs of chick blood and liver and spleen negatively correlated, suggesting recovery of spleen and liver cells after exposure to a clastogenic compound. Thus, the lower CVs may also have been indicative of genetic damage. Based on the findings of this study, FCM is a potential indicator of certain environmental contaminants in black-crowned night-herons.

    Notes: The meiotic chromosomes of 4 species of turtles representing 3 genera (Clemmys, Mauremys and Geochelone) are described and compared. Clemmys differs from Mauremys and Geochelone in possessing one fewer bivalent in diakinesis and one fewer chromosome in prophase II. Mauremys differs from Clemmys and Geochelone in possessing a distinct heterochromatic knob on one small bivalent in late pachytene. The presence of this heterochromatic region in the batagurine turtle may be the ancestral condition and its loss may now characterize the emydines such as Clemmys and the testudinids such as Geochelone.

    Notes: Abstract Banded chromosomes of five species of testudinid turtles (Geochelone pardalis, G. elongata, G. elephantopus, Gopherus berlandieri, and G. polyphemus) reveal little variation within either genus, although there are differences in amount and distribution of heterochromatin between Geochelone pardalis and G. elongata. The chromosomal position and size of the nucleolar-organizer region differs between species of the two genera. Comparisons of standard karyotypes of these species and Malacochersus tornieri with data in the literature on other tortoises show a diploid number of 52 characterizes the family. These data are consistent with those for other families which show turtles are karyotypically conservative. G-banded chromosomes of Geochelone are identical to those of Chinemys reevesi, a karyotypically primitive batagurine emydid, supporting a derivation of the tortoises from a batagurine ancestor.

    Notes: Abstract Differential staining techniques were used to study the structure and variation of the NORs of 27 species of cryptodiran turtles. No species or individuals had more than a single pair of NORs. Extensive variation in NOR structure and chromosomal location was found among higher taxa and individual variation in NOR size was common. Thirty eight percent of all individuals studied were heterozygous for the size of the NOR. However, interspecific variation in chromosomal location and structure of the NOR within major taxa was relatively rare. It is concluded that ( I ) the pattern of variation of NORs is consistent with patterns of chromosomal evolution in turtles; (2) Turtles have only a single pair of NORs whereas other animals, such as some mammals, possess numerous NORs; (3) The heterochromatin associated with the NOR is involved in the structure of the nucleolus.
