    Notes: The problems of prevention and therapy of cancer are far from being solved. Regarding therapy, it is beyond doubt that surgery and radiotherapy cure a certain percentage of cancer patients. Cancer chemotherapy with the conventional cytostatic agents is a further means by which oncologists can help patients. Immunotherapy, including the newest methods of treatment with thymosin, and particularly interferon, may bring further progress. In the field of prevention of cancer mortality, early detection of precancerous and cancerous lesions and elimination of carcinogenic agents are up to now the most successful means. The continuing high mortality of cancer, even in the most developed countries, underlines the still unsatisfactory medical methods at our disposal. New approaches to the cancer problem are badly needed. It is the purpose of this conference to acquaint us with the basic research work done in the field of retinoids. The retinoids (vitamin A and analogs) represent a fairly new development in the cancer field, offering a new approach differing markedly in their more physiological mode of action from the hitherto existing approaches in prevention and therapy of cancer.

    Notes: Zusammenfassung Es wird über Versuche an Ratten berichtet, die mit 4-dl-Methyl-tryptophan und 5-dl-Methyltryptophan behandelt wurden. TÄgliche Injektionen von 25, 50 bzw. 100 mg führten regelmÄ\ig eine NierenschÄdigung herbei. Mit kleinen Tagesdosen lie\ sich nach einiger Zeit eine herdförmige Glomerulotubulonephrose erzielen. Grö\ere Mengen führten zu atrophischen Prozessen sowie cystischen Erweiterungen einer Anzahl von Nephronen. In fortgeschrittenen FÄllen standen die Ektasien von Tubuli und Glomeruli im Vordergrunde des pathologischen Geschehens, so da\ man von einer Nephrohydrose sprechen konnte. Die Pathogenese dieser Art von NierenschÄdigung wird diskutiert.

    Notes: Summary The early and recent investigations in the field of retinoids and cancer are reviewed. The retinoids, including natural vitamin A compounds and their synthetic analogs, present a new class of substances exerting a prophylactic and a therapeutic effect both in certain experimental tumor models and in certain clinical conditions of preneoplastic and neoplastic lesions. Because of a particular physiological mechanism of action, the retinoids offer a new approach to the cancer problem, which is different from those of surgery, X-ray therapy, conventional chemotherapy, and immunotherapy.

    Notes: Summary A review of recent investigations in the retinoid field is presented. Retinoic acid exerts a prophylactic and a therapeutic effect on chemically induced benign and malignant epithelial tumors in mice. In clinical studies positive therapeutic results have been obtained in patients with preneoplastic and neoplastic epithelial lesions. However, treatment with retinoic acid is limited by serious side effects (hypervitaminosis A syndrome). Therefore, the synthesis of analogs of retinoic acid (retinoids) possessing a more favorable therapeutic ratio has been initiated. Among a large series of synthesized compounds, certain aromatic analogs proved to have a particularly favorable therapeutic ratio. The structure-activity relationship of the most active retinoids is discussed including some biological data concerning prophylaxis and therapy of epithelial tumors. The total synthesis of retinoids according to various building schemes is discussed in detail. Methods for the synthesis of the cyclic end group, of the polyene chain component, and of the full retinoid skeleton are described. Metabolic studies of retinoic acid and of the most active retinoid, as well as the synthesis of some isolated metabolites are outlined. Suggestions concerning the mechanism of action of retinoids are made. Some clinical results on the treatment of acne, psoriasis and precancerous conditions are reported.

    Notes: Abstract Topically applied retinoic acid induces partial or complete clinical regressions of senile or actinic keratoses as well as of basal cell carcinomas of the skin. 15 out of 16 treated basal cell carcinomas regressed by more than 50%, regressions being complete in 5 cases. The mode of action of retinoic acid on premalignant and malignant lesions is discussed.

    Notes: Summary Male and female albino rats were treated with Myleran (CH3SO2OCH2CH2CH2CH2OSO2CH3). Particular attention was paid to the histological examinations of the gonads of these animals. The testes as well as the ovaries were badly damaged. The male germinal epithelium was completely destroyed. Oocytes and follicles were no longer present in the ovaries.

    Notes: Zusammenfassung 1-(3,4-Dichlorphenyl)-5-isopropyl-biguanid (I) besitzt cytostatische Eigenschaften, die speziesabhängig sind. I übt eine stark depressive Wirkung auf die Hämatopoese von Meerschweinchen, Katze und Mensch aus, während das Knochenmark von Maus, Ratte, Kaninchen und Hund nicht beeinflusst wird. Transplantable Tumoren von Maus und Ratte werden in ihrem Wachstum nicht gehemmt. Hingegen liessen sich beim Menschen objektive Regressionen einiger Tumorarten beobachten. Die Wirkung auf die Meerschweinchenleukopenie kann durch Formyltetrahydrofolsäure aufgehoben werden. I hemmt die Dihydrofolatreduktase der auf I empfindlichen Spezies Meerschweinchen, nicht aber das entsprechende Ferment der auf I resistenten Spezies Ratte. Der Folsäureantagonismus ist speziesabhängig, was auf eine verschiedene Empfindlichkeit der Dihydrofolatreduktasen und nicht auf einen verschiedenen Metabolismus zurückzuführen sein dürfte. Borverbindungen von I besitzen ähnliche cytostatische Eigenschaften.
