    Notes: A new therapeutic approach for inflammatory bowel diseases is based on the administration of probiotic bacteria. Prokaryotic DNA contains unmethylated CpG motifs which can activate immune responses, but it is unknown whether bacterial DNA is involved in the beneficial effects obtained by probiotic treatment. Peripheral blood mononuclear cells (PBMC) from healthy donors were incubated with pure DNA of eight probiotic strains and with total bacterial DNA from human feces collected before and after probiotic ingestion. Cytokine production was analyzed in culture supernatants. Modification of human microflora after probiotic administration was proven by polymerase chain reaction analysis. Here we show that Bifidobacterium genomic DNA induced secretion of the antiinflammatory interleukin-10 by PBMC. Total bacterial DNA from feces collected after probiotic administration modulated the immune response by a decrease of interleukin-1β and an increase of interleukin-10.

    Notes: Abstract In order to produce l(+)-lactic acid to be employed in poly-l-lactic acid polymer production, for biomedical applications, the strain Lactobacillus casei subsp. casei DSM 20011 was studied in a conventional batch mode using different initial concentrations of glucose. The results obtained showed that the initial glucose concentration exerts an influence on the fermentation pattern, modifying the different fermentation parameters. Nevertheless, the product yield remained at a constant value of 0.86 g·g−1. The proposed novel system of product recovery, based on the use of ion-exchange resins, gave high yields of pure lactic acid.

    Notes: Summary dl-Ethionine-resistant mutants of Saccharomyces uvarum ATCC 26602 were found to overproduce exogenous l-methionine. dl-Ethionine-resistant mutant ER 108, carrying a mutation to chloramphenicol resistance was converted to petite form, and protoplasts obtained from it were fused with protoplasts from antibiotic-sensitive S. cerevisiae X2928 carrying six auxotrophies. The resulting fusants maintained four auxotrophies and were capable of overproducing l-methionine. These fusants were stable after ten passages on complete medium.

    Notes: Summary It is well known that molasses stillage is difficult to dry because of its high hygroscopicity. This investigation was made to try to affect the drying capability of beet molasses stillage by the addition of gelling agents. Increase in crude protein and essential amino acid content of beet molasses was obtained by growing Brevibacterium flavum and Candida utilis. The results obtained showed that drying performance is probably due to an “optimum” combination of the chemico-physical properties of the raw material.

    Notes: Summary Transformation of intact cells of Bacillus licheniformis 5A24 with plasmids pLM6 (2.8 kb), pC194 (2.9 kb) and pCP49 (7.1 kb) by electroporation resulted in 1.5 × 106, 1.2 × 106 , and 5.2 × 104 transformants per μg DNA, respectively. Transformation was possible with plasmid DNA, which was religated after restriction endonuclease digestions. In addition, evidence is presented that indicates that B. licheniformis possesses DNA restriction and modification systems.

    Notes: Abstract Screening for the plasmid content of 94 strains belonging to ten species of thermophilic bacilli mainly referred toBacillus stearothermophilus was carried out. Twenty-seven of the strains tested were found to harbor one or more plasmids ranging in size from 1.7 to〉50 kb. The physical map of the smallest plasmid, pBC1, is reported. Southern hybridization experiments showed that pBC1 hybridized strongly with the other small plasmids, weakly with medium-sized plasmids, and not at all with the largest plasmids and chromosomal DNA. pBC1 was cloned into pHV14 and pJH101 vectors, and the chimeric plasmids obtained were used to transformEscherichia coli andBacillus subtilis.

    Notes: The optimal culture conditions for Bacillus stearothermophilus NUB3621 (BGSC 9A5) in chemostat were studied. The results obtained showed that the optimal culture conditions in terms of biomass concentration and maximum growth rate were 65°C, pH 6.8 to 7.2. Dissolved oxygen became growth limiting at pO2 levels below 10%. Furthermore, this strain was transformed with three new hybrid vectors (pPAM2, pPCH2, or pPLY2) constructed by cloning in pRP9, a plasmid based on the thermophilic replicon, pBC1, and three heterologous genes: the α-amylase gene from Bacillus licheniformis, the cholesterol oxidase gene from Streptomyces sp., and the lipase gene from Pseudomonas fluorescens. The influence of several fermentative conditions on segregational and structural stability of the recombinant B. stearothermophilus NUB3621 transformants was studied.The parameters of plasmid loss, that is, rate of plasmid loss (R) and specific growth rate difference (δμ), were calculated. B. stearothermophilus NUB3621 carrying pRP9 showed great segregational stability in all the assayed conditions, exceeding more than 300 generations without significant plasmid loss, whereas NUB3621 carrying pPAM2, pPCH2, or pPLY2 exhibited relatively low plasmid stability. The segregational instability of the recombinant constructs increased by increasing the fermentation temperature, decreased by increasing the dilution rate, and was not affected by the level of dissolved oxygen. On the other hand, plasmid maintenance decreased in minimal medium if compared with the results obtained in complex medium. Restriction analyses carried out on cultures of NUB3621 carrying pRP9, pPAM2, pPCH2, or pPLY2, grown for 200 generations on nonselective media, revealed that all the clones tested contained the parental plasmids. These results indicate that the heterologous inserts did not affect the structural stability of the recombinant plasmids. © 1997 John Wiley & Sons, Inc. Biotechnol Bioeng 53: 507-514, 1997.
