    Notes: Summary Oxprenolol, 40, 80 and 160 mg, was administered orally to seven healthy volunteers. Over the following eight hours repeated measurements were made of the plasma concentrations and effects on heart rate, myocardial contractility (PEPC) and systolic and diastolic blood pressure in recumbency, in the upright position and during physical effort at a work load of 120 watts on a bicycle ergometer. The maximum plasma levels and the area beneath the plasma concentration curves increased roughly in proportion to the dosage increment. No evidence of first-pass inactivation in the liver was found. The half-life of the drug in plasma was approximately 80 minutes, irrespective of the dose administered. Oxprenolol slowed heart rate, prolonged PEPC and lowered systolic blood pressure, by comparison with values recorded after a placebo. The effects were generally least marked in the recumbent position and most marked during effort, when a clear-cut dose-response relation was found. The pharmacodynamic effects of oxprenolol were compared with its concentration in plasma. Marked beta-receptor blockade still persisted eight hours after dosing, although at this time, after doses of 40 and 80 mg, the drug could not be detected in plasma.

    Notes: Summary 9 unilateral, nephrectomised dogs became hypertensive by injecting SiO2-suspension into the renal artery. The chronic renal hypertension ceased in 6 dogs by shunting the renal venous blood into the portal vein. 2 animals died after the shuntoperation, 1 dog was used as control-animal. The intrarenal lesions caused a permanent reduction of the excretory renal function for 30–50% (C In ,C PAH ). It is concluded that the experimental, chronic renal hypertension is based on a humoral agent, which was changed by a depressing effect of the liver.

    Description / Table of Contents: Zusammenfassung Nach einer kurzen Beschreibung des von Jensen, Schmidt u. Garamella angegebenen Verfahrens zur Korrektur der Trichterbrust wird über die Operationsspätergebnisse bei 26 nach dieser Methode operierten Patienten, von denen 14 nachuntersucht werden konnten, berichtet. Die besten funktionellen und kosmetischen Resultate fanden sich nicht, wie bisher im Schrifttum mitgeteilt, bei Kindern und Jugendlichen, sondern bei Erwachsenen jenseits des Wachstumsalters. Aufgrund dieser Untersuchungen sollte die Indikation zur Korrektur einer Trichterbrust anders als bisher gestellt werden: Die Operation sollte erst nach abgeschlossenem Wachstum durchgeführt werden, da nur dann dauerhafte funktionelle und kosmetische Erfolge zu erwarten sind.
    Notes: Summary After a brief description of the method which has been reported by Jensen, Schmidt and Garamella for correction of funnel chest, the author reports on the late results of this operation in 26 patients who underwent surgical procedures according to this method and of whom 14 could be re-examined. The best functional and cosmetics results were not found, as has hitherto been reported in the literature, in children and juveniles, but in adults beyond the age of growth. On account of these studies the indication for correction of funnel chest should be redefined: The operation should only be carried out after growth has been completed since only then can permanent functional and cosmetic results be expected.

    Description / Table of Contents: Zusammenfassung Nach eingehenden Voruntersuchungen am Hund war zu vermuten, daß ein intrarenal bedingter Hochdruck des Hundes durch Ableitung des Nierenvenenblutes in Gestalt venöser renoportaler Anastomosen in die Leber zu beeinflussen ist. - Um dieses Problem methodisch zu erarbeiten, wurden zuerst gezielt einige Voruntersuchungen angestellt, die sich folgendermaßen gestalteten 1. Kurz- und Langzeitinfusionen von Angiotensinlösung in die untere Hohlvene und die Pfortader bewirkten unterschiedliches Blutdruckverhalten: Der Blutdruckanstieg bei Infusion in die Pfortader war geringer als bei solchen in die Hohlvene. Diese Untersuchungsergebnisse deuteten darauf hin, daß in der Leber pressorische Substanzen vergleichsweise vermehrt abgebaut werden. 2. Durch das Blutdruckverhalten der Ratte konnte geklärt werden, daß die Angiotensinasewirksamkeit im Plasma des Pfortaderblutes und peripheren venösen Blutes von Hunden und Menschen gleich ist. Damit war eine Hypothese widerlegt, nach welcher im Pfortaderblut angiotensinasehemmende Stoffe sind, die den Abbau pressorischer Substanzen verhindern könnten. Es müßte demnach die direkte Ableitung des Nieren. venenblutes über eine renoportale Anastomose genügen, um eine Drucksenkung bei Hunden mit Drosselungshochdruck herbeizuführen. Eine suprarenale kavoportale Transposition wäre dazu nicht unbedingt erforderlich. 3. Durch Ableitung des Nierenvenenblutes über eine venöse renoportale Anastomose gelang es erwartungsgemäß bei linksseitig nephrektomierten Hunden einen Drosselungshochdruck zu verhindern bzw. zu senken. 4. Chronische intrarenale, hypertensinogene Läsionen konnten durch Injektion von Quarzstaubsuspension in die Nierenarterie mittels einer besonderen Injektionstechnik erzeugt werden. Alle vier Versuchstiere starben infolge einer Überdosierung des verabreichten Siliciumdioxyds an Urämie. Auf Grund der in den Voruntersuchungen gesammelten methodischen Erfahrungen gelang es, in den Hauptversuchen bei 9 linksseitig nephrektomierten Bastardhunden einen chronischen nephrogenen, intrarenal bedingten Hochdruck zu erzeugen, der 65 bis 110 Tage erhalten wurde und in 6 Fällen durch Ableitung des Nierenvenenblutes über eine renoportale Anastomose in die Leber gesenkt werden konnte. Ein Tier blieb 160 Tage hyperton. Zwei Tiere gingen nach der Anastomosenoperation ein. Die blutig gemessenen (Carotisschlinge) Drucke stiegen durchschnittlich von Werten um 13590 auf solche um 180120 mm Hg. Einige Tage nach der Anastomosenoperation sanken die Blutdrucke teilweise sogar unter die Ausgangswerte und verblieben dort. Die begleitenden Nierenfunktionsproben (Harnstoff und Kreatinin) und vor allem die Inulin- und PAH-Clearance blieben nach der SiO2-Injektion in der hypertonen Periode und auch nach Anlegen der renoportalen Anastomose — in der wieder normohypertonen Periode — bis zum Versuchsende erheblich verändert. Die Werte der Ausgangsclearance zeigten eine bleibende Reduktion um 30 bis 50%, wobei die Inulinclearance etwas stärker betroffen war. Die funktionelle Insuffizienz der restlichen Nephren drückte sich konstant in einer kompensierten Retention harnpflichtiger Substanzen aus. Der renale Hochdruckmechanismus wurde durch die Ableitung des Nierenvenenblutes in die Leber abgewandelt. Der Hypertonus war zu beeinflussen, wobei die Beeinträchtigung der exkretorischen Nierenfunktion bestehen blieb. Diese Art des nephrogenen Hochdruckes — hervorgerufen durch intrarenale Läsionen - war funktionell dem renovasculären Hochdruck nicht vergleichbar, weil in Verbindung mit der exkretorischen Störung Wasser- und Elektrolytverluste im Sinne einer Salzverlustniere beobachtet wurden.

    Notes: Abstract The purpose of this work was to compare mitochondrial DNA restriction endonuclease patterns in strains of the yeast Kluyveromyces lactis, from different sources, to see how conserved is the organization of this organellar genome. The mitochondrial DNA of five independently-isolated strains and one of unknown origin were compared. Strains NRRL Y-1205, NRRL Y-8279 and NRRL Y-1140 gave identical patterns. Strain NRRL Y-1564 showed an insertion, with respect to the other three, of approximately 1250 bp. Strain W600B had also an insertion with extra restriction sites for EcoRI, HpaI, HaeIII, HincII and XbaI. On the other hand, strain Y-123 showed a restriction pattern quite different from the others. Sequences putatively encoding apocytochrome b, ATPase subunit 9 and ribosomal RNA large subunit, were localized on the physical maps of three strains. Results demonstrated that the order of these three genes shows a common feature in strains W600B and WM37 (auxotroph of Y-1140) but a different distribution in WM27 (auxotroph derived from Y-123). All these facts explain the extensive intraspecific polymorphism observed in the mtDNA of this yeast.

    Notes: Summary The mitochondrial ATPase from oligomycin-resistant mutants which map on different regions of an extrachromosomal DNA (01 and 011 class mutants) showed an increased resistance to oligomycin and venturicidin when assayed in vitro as compared to the sensitive strains. The resistance to oligomycin of the isolated mitochondrial ATPase from 01 class mutants was higher than that of the 011 class mutants. Cross resistance of the oligomycin-resistant mutants to the antibiotics peliomycin and ossamycin, which also inhibit phosphoryl transfer reactions in mitochondria (Walter et al., 1967), was observed, 01 mutants being more resistant to ossamycin than 011 class mutants. At the concentrations of peliomycin studied, no difference in sensitivity among both groups of oligomycin-resistant mutants could be detected. Mitochondrial respiration and isolated mitochondrial ATPase activity are sensitive to venturicidin, suggesting that the previously observed (Brunner et al., 1977) in vivo venturicidin resistance of K. lactis is probably due to an impairment of the influx of the drug at the level of the plasma membrane.
