    Notes: Summary A histological comparison was made between normal mouse tails and those treated with crude coal tar, and the effect of crude coal tar on the keratin profile of the living cells of treated animals was examined. The prophylactic effect of crude coal tar on the neonatal mouse tail is described. The variation in the anatomical site of prekeratin of the dorsal and tail epidermis of the mouse is reported. These results are discussed with reference to the use of the mouse tail as a model for screening drugs for the treatment of psoriasis.

    Notes: Abstract It has been postulated that patients with Malassezia furfur associated dermatoses have a deficient cell-mediated immune response to M. furfur. This study examined the cell-mediated immune responses to M. furfur serovars A, B and C of 10 patients with pityriasis versicolor and 10 age- and sex-matched controls; and 10 patients with seborrheic dermatitis and 10 age- and sex-matched controls. The responses to each serovar of M. furfur were assessed using the lymphocyte transformation assay and the leukocyte migration inhibition assay. The lymphocyte transformation responses of the patients with pityriasis versicolor to M. furfur serovars A, B and C (O/10, 6/10 and 5/10 respectively) were not significantly different from those of controls (0/10, 2/10 and 1/10). However, for patients with seborrheic dermatitis, significantly more patients lymphocytes responded to serovars B and C (6/10 and 6/10 respectively) than those of controls (1/10 and 1/10). No patient or control responded to serovar A. In the leukocyte migration inhibition assay, the leukocytes from a greater proportion of patients with pityriasis versicolor (5/7) responded to serovar B than controls (2/10); and the leukocytes from a greater proportion of patients with seborrheic dermatitis (4/10) responded to serovar C than controls (0/9). Thus, this data did not indicate the presence of any cell-mediated immune deficiency to M. furfur in patients with pityriasis versicolor or seborrheic dermatitis, as measured by the lymphocyte transformation assay or the leukocyte migration inhibition assay. The greater responsiveness of T lymphocytes from patients may indicate that T lymphocytes might be involved in the pathogenesis of these diseases.

    Notes: Abstract Patient and normal volunteer sera were used as probes in two-dimensional PAGE of P. acnes culture supernatant fluid and cell extracts to determine whether specific P. acnes polypeptides were associated with the immune reaction in acne. Eight polypeptides, Mr 20 to 131 × 103, pi 4.7 to 6.5 in the cell extract, and 7 polypeptides Mr 10 to 24 kD, pI 4.8 to 7.5 in the culture supernatant fluid were specifically highlighted by patient sera and not volunteer sera. These polypeptides were not related to described extracellular enzymes of P. acnes. It is possible that these polypeptides are involved in the induction of acne.

    Notes: Abstract: The mechanisms involved in the development of inflammation in acne vulgaris have yet to be elucidated. Previous studies have shown that the initial cellular infiltrate in early inflammatory lesions is mononuclear, predominantly CD4 positive T cells. The aims of this study were to investigate the pattern of expression of adhesion molecules and HLA-DR in evolving acne lesions. Forty-nine patients with moderate to severe acne were biopsied following lesion mapping. Lesions were classified according to their duration of inflammation as up to 6 h, from 6 to 24 h, from 24 to 48 h and from 24 to 48 h and from 48 to 72 h. The cellular infiltrate was determined using monoclonal antibodies to CD1, CD3, CD4 and CD8. The expression of ICAM-1, E-selectin, VCAM-1 and HLA-DR was determined. Early (6 h) lesions had perivascular CD3 positive T-cell infiltrates which were predominantly CD4 positive. This was associated with vascular expression of ICAM-1, E-selectin, VCAM-1 and HLA-DR. Periductal infiltrates were present in 70% of the early lesions (up to 6 h). The cells were predominantly CD4 positive and associated with a high level of HLA-DR and ICAM-1 expression. Periductal infiltration increased with time and persisted to 72 h. ICAM-1 and HLA-DR were expressed epidermally in early and late lesions. CD1 positive cells were a minor, but consistent element in the perivascular and periductal infiltrates of early and late lesions. There was no statistically significant difference in the levels of expression of E-selectin, VCAM-1, ICAM-1 or HLA-DR for lesions of different duration. The pattern of HLA-DR and adhesion molecule expression plus the nature of the cellular infiltrate supports the hypothesis that inflammation in acne is mediated by CD4 positive T cells.

    Notes: Abstract Leg ulcers present a common and recurring problem in older people creating discomfort and distress for the patient and a great cost to the health care services. Cultured keratinocyte grafts have been used by many investigators to stimulate healing of chronic venous ulcers. It has been proposed that they may do this by producing cytokines which modulate the healing process. However, the types and levels of cytokines in the leg ulcer fluid before and during healing arc not known. Wound fluid was collected from venous leg ulcers in 18 patients beneath occlusive Tega-derm™ dressing for 4 to 6 h. The leg ulcers were divided on clinical criteria into ‘healing’ and ‘non-healing’. PDGF-AB, GM-CSE IL-1α, IL-1β. IL-6 and bFGF were measured by ELISA and the levels of IL-1α, IL-1β. and IL-6 were also measured using biological assays. The effect of leg ulcer wound fluid on fibroblast and keratinocyte proliferation was measured indirectly by 3H-thymidine incorporation and MTT assay. Total protein, albumin levels, fibronectin degrading activity and collagenase activity, both active and latent were measured. No statistically significant differences in the levels of cytokines or collagenase were identified between healing and non-healing leg ulcers in the sample of leg ulcers studied. However, this study does give valuable information concerning the levels of cytokines and collagenase in chronic leg ulcer wound fluid.

    Notes: We describe two simple, reproducible scoring systems for assessing acne severity, and we emphasize the technical problems which could invalidate either technique. Constant baseline data is desirable for any clinical trial, and our data clearly show that acne patients should ideally be off all treatment for at least 2 months before the start of a therapeutic trial.

    Notes: Investigations on fifteen patients with acne showed that at the end of 1 and 2 months of treatment 5% benzoyl peroxide increased the sebum excretion rate by 22.5%. This rise in sebum excretion is probably due to the comedolytic activity of benzoyl peroxide which will influence the pooling of sebum in the upper reaches of the pilosebaceous duct.
