    Notes: Abstract Muropeptide composition of peptidoglycan from the Gram-negative bacteria Aeromonas sp., Acinetobacter acetoaceticus, Agrobacterium tumefaciens, Enterobacter cloacae, Proteus morganii, Pseudomonas aeruginosa, Pseudomonas putida, Vibrio parahaemolyticus Yersinia enterocolitica and Escherichia coli, was analyzed by HPLC In all instances peptidoglycan was built up from the same subunits. A wide disparity in the relative abundance of muropeptides and all structural parameters was observed. The contribution of LD-A2pm-A2pm cross-linked muropeptides was extremely variable; from 1 to 45% of cross-linked muropeptides. Muropeptides with the dipeptides Lys-Lys or Arg-Lys, indicative of murein-bound (lipo)proteins, were detected in all instances although abundance was very variable.

    Notes: Abstract Murein synthesis in Escherichia coli can be partially inhibited by d-methionine without concomitant alterations in growth and morphology. d-Methionine-treated cultures grow steadily for an indefinite time, therefore murein surface density should be reduced. Determination of this parameter in control and d-methionine-treated cells showed a severe reduction in the latter. Murein surface density increases drastically in resting cells, irrespective of the presence of d-methionine. Mutants in ponB are hypersensitive to d-methionine. Analysis of ponB strains revealed an important reduction in murein surface density. An approximately two-fold reduction in average surface density is apparently compatible with normal growth and division.

    Notes: Abstract Certain d-amino acids can be incorporated into the murein sacculus of Escherichia coli apparently through a direct transpeptidation reaction independent of the normal biosynthetic pathway. Investigation of this process is important because it could lead to the identification of hitherto unknown enzymes involved in murein metabolism. However, a serious drawback is the lack of an appropriate in vitro assay. We have analysed the suitability of a system based on the incorporation of a radioactive substrate (S-[3H]methyl-d-cysteine) by ether-treated cells, a method successfully applied before to the study of murein biosynthesis. The results reported here indicate that ether-treated cells are indeed proficient in the incorporation of d-amino acids, matching closely the properties of the reaction in growing cells.

    Notes: Three selected peptidogycan monomers isolated from muramidase-digested Escherichia coli-sacculus were characterized by positive- and negative-ion 252Cf plasma-desorption mass spectrometry (PDMS). Dut to the high affinity of sugar moieties towards alkali cations multiple alkali attachment in PDMS can be a problem. Electrospray, nitrocellulose, nitrocellulose sandwich anthracene-9-carboxylic acid 3-(3-Pyridyl)acrylic acid deposition techniques were evaluated for the degree of (de)proltonation and cationization of the molecular species by sodium and/or potassium. Electrosprayed and nitrocellulose-deposited samples mainly exhibited intense cationized molecular ion species whereas anthracene-9-caboxylic acid-deposition sample show a similar pattern but with much lower intensity. An extension of the nitrocellulose deposition technique, the so-called nitrocellulose sandwich sample preparation reduced the aallkali attachment problem for these hydrophilic compounds. Application of the small molecule matrix 3-(3-pyridyl)acrylic acid yielded mass spectra with abundant [M+H]+ and [M—H]- ions in the positive- and in the negative-ion modes, respectively.

    Notes: Abstract. A variant form of mouse Chromosome (Chr) 17, the t-haplotype, contains several loci responsible for transmission ratio distortion in males. Sperm carrying the responder locus (Tcr) have a high probability of fertilizing eggs at the expense of wild-type sperm, provided that distorter loci (Tcd-1 to Tcd-5) are expressed during spermatogenesis. Tcr has been mapped to the Leh66b region within a maximum of 155 kb. In the search for genes in the genomic region Leh66EI, we have identified the mouse homolog of human ribosome S6 kinase 3 (RSK3) on cosmid DNA. The complete mouse Rsk3 gene is encoded in the region Leh66a of t-haplotypes and Leh66EI of the wild-type chromosome. It consists of at least 13 exons spanning over more than 120 kb. Rsk3 is expressed in embryos and in several adult organs including testis. Cosmids covering 100 kb of the Leh66b region or 120 kb of the Leh66a region were isolated. Rsk3 covers about 65 kb of the Leh66b region and appears to be incomplete at its 5′-end. A correlation of the physical map provided here with the genetic mapping of Tcr reported previously suggests that Tcr is most likely encoded within a fragment of 30 kb upstream or 20 kb downstream of Rsk3. These data will facilitate the isolation of Tcr, a prerequisite for understanding transmission ratio distortion in mouse.
