    Notes: Summary We have studied the embryonic and adult phenotypes of genetic combinations between Polycomb (Pc), Regulator of bithorax (Rg-bx) and the genes of the Bithorax complex (BX-C) and the Antennapedia complex (ANT-C). The products of Pc and Rg-bx genes act antagonistically, their mutant combinations leading to the ectopic expression of genes of the BX-C and ANT-C. The genetic analysis of the Pc locus suggests it is a complex gene. Pc+ products behave as members of a “regulatory set” that negatively control the expression of BX-C and ANT-C genes. Genetic combinations between different doses of Pc, Rg-bx and the genes of the BX-C and ANT-C have phenotypes which may be interpreted as resulting from ectopic derepression of posterior selector genes repressing selector genes of anterior segments. The transformation phenotypes of certain genetic combinations differ in embryos and adults. A model of regulation of the BX-C and the ANT-C genes during the imaginal cell proliferation is presented, in which the specification state is maintained by self-activation of a given selector gene and down modulation of other selector genes in the same cell.

    Notes: Summary Exposure to ether of wild-type embryos of different strains ofDrosophila melanogaster causes phenocopies of different alleles of thebithorax system. Clonal analysis of the phenocopy spots has shown that the transformation caused by the treatment is maintained by cell heredity. Embryos heterozygous for several recessive mutant alleles ofbithorax show the same frequency of phenocopies as wild-type homozygous sib controls. The same holds for embryos heterozygous for the dominant mutant allelesCbx andUbx 1 which are point mutants in thecis-regulatory region of the system. However, for dominant mutants which have breakpoints in this region (Ubx 80,Ubx 130 andHm) the frequency of phenocopies is about twice that of their sib controls. Embryos with increasing numbers of copies (from 1 to 4) of thebithorax system show a decreasing frequency of phenocopies. A model is proposed that explains “bithorax” phenocopies as resulting from disturbances in the distribution of positional information signals for segments (inductor molecules) which compete with the product of a regulator gene (repressor) and thecis-regulatory region of thebithorax system. On this model, the initiation of a metathoracic developmental pathway would result from the derepression of thebithorax system.

    Notes: Abstract. Genetic engineering, coupled with spectro scopic analyses, has enabled the metal binding proper ties of the α and β subunits of mouse metallothionein 1 (MT) to be characterized. A heterologous expression system in E.coli has led to high yields of their pure zinc-complexed forms. The cadmium(II) binding properties of recombinant Zn4-αMT and Zn3-βMT have been studied by electronic absorption and circular dichroism. The former binds Cd(II) identically to α fragments obtained from mammalian organs, showing that the recombinant polypeptide behaves like the na tive protein. Titration of Zn3-βMT with CdCl2 results in the formation of Cd3-βMT. The addition of excess Cd(II) leads to Cd4-βMT which, with the extra loading of Cd(II), unravels to give rise isodichroically to Cd9-βMT. The effect of cadmium-displaced Zn(II) ions and excess Cd(II) above the full metal occupancy of three has been studied using Chelex-100. The Cd3-βMT species is stable in the presence of this strong metal-chelating agent.

    Notes: Summary We have studied the genetic properties and developmental effects of several mutations, in eight different loci, which alter the specification of embryonic segments. All of the changes are related to the transformations caused by mutants of the bithorax complex. Several properties: the interactions of these mutants with different mutants of the bithorax complex, the interactions between themselves, the effect of changing dosages of their wildtype alleles and their response to ether induction of phenocopies permit one to distinguish between those mutations which affect activation of the bithorax genes in early embryogenesis and those which affect expression of the bithorax genes during development. This paper deals mainly with the Rg-bx locus and its interactions with other loci which affect segment specification. In particular two loci show genetic and developmental characteristics which seem to conform to those expected for a repressor coding gene (Polycomb) and for an inducer synthesizing gene (Rg-bx) active in a negative control system of the genes in the bithorax complex. A model for the interaction of the wildtype products of these genes to determine the segmental characteristics of both the thorax and abdomen is proposed.

    Notes: The complex [Pt2(μ-H)(μ-S)(dppe)2](PF6) undergoes a displacive order–disorder transformation at ca 230 K. The low-temperature structure is ordered with one cation–anion pair as the asymmetric unit in space group P21/n. At room temperature the b axis is halved and the space group is P2/n, imposing crystallographic twofold rotation symmetry on both ions; the anion shows major disorder and there is probably minor disorder in the cation, but its internal geometry remains essentially unchanged. The heteronuclear complex [PdPt(μ-H)(μ-S)(dppe)2](PF6) is isostructural with the Pt2 complex at room temperature. All three structures have been determined crystallographically and both complexes have been extensively characterized by NMR spectroscopy, unambiguously confirming the genuine heteronuclear nature of the mixed-metal complex and the presence of the bridging hydride ligand.
