    Description / Table of Contents: Summary An electron microscopic study of the submandibular salivary gland lesion occurring in the NZB/NZW F1 hybrid mice was performed, and the findings were discussed and correlated to the autoimmune salivary gland disease observed in patients with Sjögren's syndrome. Within the submandibular salivary gland of this mouse strain focal infiltrates of mononuclear inflammatory cells were observed. Lymphoid cells were observed inside the basal lamina of acini and ducts, leading to destruction of these glandular units. The typical duct cell proliferation found in the autoimmune sialoadenitis of Sjögren's syndrome with the formation of the typical epimyoepithelial islands was not found in the submandibular glands of NZB/NZW mice.
    Notes: Zusammenfassung Eine elektronenmikroskopische Untersuchung der bei den NZB/NZW F1 hybriden Mäusen vorkommenden Veränderungen in den submandibularen Speicheldrüsen wurde durchgeführt, und die Befunde wurden diskutiert und korreliert mit dem autoimmunen Speicheldrüsenleiden, das bei Patienten mit Sjögrens-Syndrom beobachtet wird. In der submandibularen Speicheldrüse dieses Mäusestammes wurden begrenzte Infiltrationen mononuklearer inflammatorischer Zellen beobachtet. Die mononuklearen Zellen wurden hauptsächlich als lymphoide Zellen identifiziert. Außerdem wurden innerhalb der Lamina basilaris von Drüsen und Drüsengängen lymphoide Zellen beobachtet, die zur Zerstörung dieser Drüseneinheiten führen. Darüber hinaus wurde der typische Zuwachs der Drüsengangszellen, der bei der autoimmunen Speicheldrüsenentzündung des Sjögrens-Syndroms vorkommt und die Form von typischen epimyoepithelialen Inseln annimmt, in den submandibularen Drüsen der NZB/NZW-Mäuse nicht vorgefunden.

    Notes: Summary Four different local anaesthetic agents were tested on the tympanic membrane of the rat in order to detect any structural changes. Specimens were obtained for examination under the light microscope 10 min, 24 h, 1 week, and 2 weeks after exposure to each anaesthetic. The histological changes observed were, in general, rather similar for all agents tested, although certain differences were noted. The initial change observed was damage of the outer epithelial layers, later also accompanied by changes in the corresponding endothelial cover. No perforations were observed macroscopically. The healing process was characterized by hyperplasia of the connective tissue layers of the drum. However, the most prominent feature of this process was the hyperplastic epithelium, ingrowing from the periphery of the membrane to cover denuded areas. During certain stages of the healing process the thickness of the membrane amounted to 40 times that of the normal drum. The initial changes appeared severe both with the clinically more potent and effective anaesthetics (phenol and Bonain's liquid), and with Xylocaine spray.

    Notes: Summary Immunohistochemistry was used to study the localization of lysozyme (LZ) and lactoferrin (LF) in the human sinus mucosa during recurrent and chronic sinusitis. Serous cells of submucosal mixed glands and polymorphonuclear leukocytes both displayed a strongly positive staining reaction to both LZ and LF in the normal mucosa. A positive though weak staining for LZ and LF could also be found occasionally within goblet cells. In the mucosa from patients with recurrent or chronic sinusitis, the staining reaction to LZ appeared to intensify in goblet cells. Furthermore, an increased immunoreactivity of glands vis-à-vis LZ and LF was also noted occasionally. Atypical glands were frequently found in mucosa from patients with chronic sinusitis. The epithelium of these latter glands often showed an intense staining reaction to LF, but a rather weak reaction to LZ. The results of the present study suggest that the observed increase in LZ and LF secreting activity of goblet cells, epithelial cells and newly formed atypical glands may play a part in the defense mechanism of the sinus mucosa during the course of chronic sinusitis.

    Notes: Abstract In order to study morphological effects on the nasal and sinus mucosa, New Zealand White rabbits underwent either unselective, regional sectioning of sensory and parasympathetic nerve branches or topical treatment of the mucosa with capsaicin. Ten days after treatment, mucosal specimens were analyzed by light and electron microscopy. Immunohistochemistry was used to evaluate neuropeptides present, in particular substance P, calcitonin gene-related peptide, vasoactive intestinal peptide and neuropeptide Y. In surgically denervated rabbits, mucosal glands were found to be enlarged and contained an increased number of zymogen granules having a bipartite substructure. Topical capsaicin application caused localized epithelial changes in the sinus mucosa and maxilloturbinal region of the nose, including clotting of cilia and an increased number of goblet cells. Reduced amounts of all neuropeptides investigated were found in the surgically denervated animals, while topical capsaicin treatment had only marginal effects on the mucosal neuropeptide content. The morphological changes observed after surgical denervation suggest an imbalance between neural stimulation and secretory capacity of the mucosal glands. These findings could explain the difference in clinical effect noted between sectioning of the vidian nerve and topical treatment with capsaicin in patients with perennial rhinitis.

    Notes: Zusammenfassung Isolierte Submandibularisdrüsen des Meerschweinchens wurden in physiologischer Pufferlösung mit sekretionsstimulierenden Substanzen inkubiert. Dopamin, Noradrenalin, Adrenalin und 5-HT induzieren die Sekretion von Peroxydase und Amylase, während der Serotoninantagonist BOL 148 (2-Bromo-d-Lysergsäure Dietylamid) eine beinahe vollständige Sekretionshemmung hervorruft. BOL 148 hat sich also nicht als spezifisch 5-HT-hemmend an der Meerschweinchenspeicheldrüse erwiesen.

    Notes: Summary As visualized in the electron microscope, the parotid gland of the rabbit has a dual innervation. Both adrenergic and cholinergic nerves are equally distributed in the parenchyma and often run together within the same nerve bundle. Nerve terminals are observed not only subjacent to the basement membrane but interposed between the latter and the acinar cells, where they establish a close membrane to membrane contact with the latter.
