    Notizen: Abstract: Laboratory cultures of marine Synechococcus sp. WH 7803 were grown under conditions of restricted iron availability. The culture medium was adjusted to restrict iron availability: (i) by adding the iron chelator EDDA; (ii) by omitting iron; and (iii) by omitting both iron and EDTA. An adaptive response was observed to these iron-restricted conditions, including a decrease in cellular phycoerythrin and synthesis of a 36 kDa polypeptide in [35S]methionine radiolabelled whole cell lysates separated by SDS-PAGE. The polypeptide was synthesized within 48 h of transferring exponential phase cells to the iron-restricted medium. The protein was localized to the cell membranes and not the cytoplasmic fraction.

    Notizen: Cyanophycin is a high molecular weight co-polymer of aspartate and arginine, hitherto considered characteristic of all cyanobacteria. Chemical analysis and radioisotopic incorporation procedures have shown that precipitable cyanophycin is absent from the five species of Synechococcus tested, including the marine Synechococcus WH7803 (type DC2). The implications with respect to the role of cyanobacterial nitrogen reserves are discussed.

    Notizen: During phosphate-limited growth the marine phycoerythrin-containing picoplanktonic cyanobacterium Synechococcus sp. WH7803 synthesizes novel polypeptides, including two abundant species of 100 kDa and 32kDa. The 32kDa polypeptide was localized to the cell wall, although in a related strain, Synechococcus sp. WH8103, it could be detected in both the cell wall fraction and the periplasm. The gene (designated pstS) encoding this polypeptide was cloned and shown to be present in a single copy. The deduced amino acid sequence indicated a polypeptide consisting of 326 amino acids with a calculated Mr of 33763. Comparison of this sequence with that obtained by microsequencing the N-terminus of the 32kDa polypeptide showed that the mature protein was synthesized as a precursor, the first 24 amino acid residues being cleaved between two alanine residues at positions 24 and 25. The amino acid sequence of the mature polypeptide showed 35% identity and 52% similarity to the periplasmic phosphate-binding protein (PstS) from Escherichia coli, including three regions of much stronger homology which, by comparison with E. coli PstS, are directly involved in phosphate binding. Northern blot analysis revealed a pstS transcript of 1.2 kb in RNA extracted from cells grown in Pi-replete conditions and one of 1.4 kb in considerably increased abundance under Pi-depleted conditions. Homologues of the pstS gene were detected in other marine phycoerythrin-containing Synechococcus strains, but not in freshwater or halotolerant species.

    Notizen: Abstract The response of eight species of unicellular cyanobacteria to growth under high and low CO2 regimes was measured by RuBisCO activity and the extent of inorganic carbon concentration. No significant increase in RuBisCO activity in CO2-limited batch cultures was observed. The main feature of the data presented is the absence of a bicarbonate concentrating process in the oceanic, phycoerythrin-containing species examined, this is discussed in light of bicarbonate supply in freshwater and ocean environments.

    Notizen: Abstract Three constitutive forms of Superoxide dismutase activity have been demonstrated in the cyanobacterial marine picoplankter Synechococcus sp. WH 7803 using polyacrylamide gel activity staining techniques. A protein which gave a positive non-haem iron stain on native polyacrylamide gels exhibited N-terminal similarity to both the iron Superoxide dismutase and the manganese Superoxide dismutase of Escherichia coli. The metal prosthetic group of each of the three activity bands was characterised by analysing their differential sensitivities to 5 mM H2O2, 2 mM cyanide and 2 mM of the copper chelator diethyldithiocarbamate. Three distinct Superoxide dismutase activities were observed, an iron Superoxide dismutase, a copper/zinc Superoxide dismutase and a third form which has not been identified. Growth of Synechococcus cells in ASW medium containing no added iron resulted in no alteration in the activity of the iron Superoxide dismutase. Growth of cultures in the absence of copper or zinc resulted in differential changes in the activities of the copper/zinc Superoxide dismutase and the unidentified Superoxide dismutase.

    Notizen: Abstract Fifteen strains of cyanobacteria, mainly Nostoc and Anabaena species, were screened for plasmids using five rapid procedures. Two of these methods, based on alkaline extraction and phenol extraction of cleared lysates respectively, were successful with a total of ten species, the latter method proving more sensitive. Plasmids ranging from less than 2.6 to at least 30 mD were isolated; most of the strains examined possessed one or two plasmids, while five lacked detectable plasmid DNA.

    Notizen: Abstract Addition of the arginine analogue, canavanine, to cultures of nitrogen-fixing Anabaena cylindrica at the onset of akinete formation, resulted in the development of akinetes randomly distributed within the filament, in addition to those adjacent to heterocysts. The total frequency of akinetes increased up to five-fold. A feature of akinetes is their increased content of cyanophycin granules (an arginine-aspartic acid polymer) and addition of canavanine to cultures at an earlier stage resulted in entire filaments becoming agranular and containing agranular akinetes. The effects on akinete pattern appeared to be specific for canavanine since other amino acid analogues, although increasing the frequency of akinetes (approximately two-fold), had no effect on their position relative to heterocysts. In ammonia-grown, stationary phase cultures of A. cylindrica, akinetes were observed adjacent to proheterocysts and in positions more than 20 cells from any heterocyst. These observations indicate that nitrogen fixation and heterocysts are not essential for akinete formation in A. cylindrica, although the availability of a source of fixed nitrogen does appear to be a requirement. These results suggest that during exponential growth some aspect of the physiology of vegetative cells suppresses their development into akinetes and that the role of the heterocyst may not be one of direct stimulation of adjacent vegetative cells to form akinetes, but the removal or negation of the inhibition within them. A model for akinete formation and the involvement of canavanine is given.

    Notizen: Abstract The effect of irradiance on the growth rate, macromolecular composition and photosynthetic carbon metabolism of Amphidinium carteri was studied in batch culture. Growth rate increased linearly with increasing irradiance up to a maximum growth rate of 0.04 h-1 at an irradiance of 80 μEm-2s-1. In contrast to a number of other studies on both prokaryotic and eukaryotic microorganisms, ours showed that cellular content of RNA, DNA, protein and carbohydrate of A. carteri were invariant with growth rate over the range μ=0.04 to 0.007 h-1. The invariant macromolecular composition was correlated with a constant modal cell volume. Chlorophyll and lipid per cell increased with decreasing irradiance. The distribution of [14C]-bicarbonate in the major end products of photosynthesis after incubation with isotope for 14% of a doubling time showed that the percentage carbon in the chloroform (lipids and pigments) fraction increased with decreasing irradiance while that of the trichloroacetic acid soluble (carbohydrate) fractions decreased. The percentage of isotope in the trichloroacetic acid insoluble (protein) fraction and methanol: water fraction (metabolites) remained constant. Because this species, under light-limited growth, differs from other organisms so far studied, more species must be similarly examined before nucleic acid content is used as an index growth rate in the field.

    Notizen: Abstract The glucose-6-phosphate dehydrogenase (EC 1.1.1.49) gene (zwf) of the cyanobacterium Synechococcus PCC 7942 was cloned on a 2.8 kb Hind III fragment. Sequence analysis revealed an ORF of 1572 nucleotides encoding a polypeptide of 524 amino acids which exhibited 41% identity with the glucose-6-phosphate dehydrogenase of Escherichia coli.
