    Notes: Summary Cells of the line Kc, derived fromDrosophila melanogaster embryos, extend long processes when exposed to ecdysteroid hormones. We have devised a quantitative assay for this morphological response, using the subline Kc-H. The assay was used to characterize the conditions required for the response. A halfmaximal response is elicited by approximately 10−8M 20-hydroxyecdysone; the response is saturated by 10−7M 20-hydroxyecdysone, which causes detectable elongation within a few hours, and a maximal response after 2–3 days. The response occurs substantially normally in the absence of serum, during growth in suspension, and in over-crowded cultures. It is not elicited by cyclic nucleotides, vertebrate growth factors, or a variety of other non-ecdysteroid reagents. Of 60 ecdysteroid compounds tested, only those which were active in other insect test systems elicited the response, and the concentrations required were approximately proportional to the concentrations active in other in vitro systems. We conclude that the response of Kc cells to 20-hydroxyecdysone retains basic features of the ecdysteroid response of intact tissues and therefore that Kc cells are a useful model system for studying ecdysteroid action.

    Notes: Abstract Segregation of the complement component, C4, was analyzed in six families that each included an individual who inherited an HLA haplotype where a crossover event had occurred in the region between HLA-B and HLA-DR. Two cDNA clones corresponding to the C4 gene were utilized as probes in Southern blot analysis of DNA from members of each family. Restriction fragment length polymorphisms (RFLP) were observed and were assigned to haplotypes. In one family RFLP, hybridizing with the C4 probes, segregrated with HLA-B, and in four families RFLP segregated with HLA-DR; one family was not informative in this respect. These analyses have made it possible to localize the genes for C4 between HLA-B and HLA-DR by molecular genotyping and to characterize three different genomic configurations of C4 genes by limited restriction mapping.

    Notes: Summary DiGeorge syndrome (DGS) is a human developmental defect of the structures derived from the third and fourth pharyngeal pouches. It apparently arises due to deletion of 22q11. We describe a strategy for the isolation of DNA probes for this region. A deleted chromosome 22, which includes 22q11, was flow-sorted from a lymphoblastoid cell line of a patient with cat eye syndrome and used as the source of DNA. A DNA library was constructed from this chromosome by cloning into the EcoR1 site of the vector Lambda gt10. Inserts were amplified by PCR and mapped using a somatic cell hybrid panel of this region. Out of 32 probes, 14 were mapped to 22q11. These probes were further sublocalised within the region by dosage analysis of DGS patients, and by the use of two new hybrid cell lines which we have produced from DGS patients. One of these lines (7939B662) contains the altered human chromosome segregated from its normal homologue. This chromosome 22 contains an interstitial deletion in 22q11, and will be useful for localising further probes to the DGS region.

    Notes: This paper uses the theory of atoms in molecules to investigate the origin of molecular dipole moments. The dipole moment is given by a sum over the net charge and first moment of every atom in a molecule. The first term leads to a charge transfer contribution μc, the second to an atomic polarization contribution μa. It is shown that both terms are, in general, of equal importance in determining both the static molecular dipole moment and the moment induced by a nuclear displacement. Models which imploy only point charges and corresponding bond moments which follow rigidly the nuclear framework, i.e., models which approximate μc and ignore μa, are shown to lead to results that are incompatible with the changes that are found to occur in a molecular charge distribution during a nuclear vibration. The dipole moment is shown to be another group property that is transferable between molecules in the normal hydrocarbons, This property, along with the net charge, the energy, the correlation energy (expressed as a functional of the charge density), and the atomic volumes are transferable for methyl and methylene groups because of a corresponding transferability of the distribution of charge over the basins of the atoms in these groups up to their surfaces of zero flux in the gradient vector of the charge density. Transferability of group properties is not a result of the transferability of individual orbitals or geminals, which necessarily have tails lying outside of the group in question. Atoms have no tails and their energy, as is known from experiment, can in certain instances be transferred between systems with changes of less than 1 kcal/mol. The properties of atoms in molecules can be determined experimentally and quantum mechanics predicts these properties, just as it predicts the properties of a total system.
