    Notes: Summary The production of the K4 polysaccharide was obtained for the first time extracellularly from a strain of Escherichia coli. The set up of the fermentation conditions led to the maximum fermentation yield, as extracellular K4, after 20 h. Purification and characterization of this K4 resulted in 200 mg/L of highly purified K4.

    Notes: Summary α-Galactosidase (Ec 3.2.1.22) was obtained from the yeast C. javanica grown on a mineral culture medium supplemented with melibiose plus raffinose. The cell-bound α-galactosidase and the preparations from the culture filtrate and from the cells exhibited maximum activity at pH 4 and 70°C. After 15 min at 70°C, 70% of the α-galactosidase activity was recovered, and after 15 min at 80°C complete inactivation was observed. The Michaelis constant (Km) of α-galactosidase on pNPG and on melibiose was about 11 mM. Galactose as a reaction product had no effect on α-galactosidase activity of the preparation from the cells, whereas glucose (2 and 10 mM) increased the activity. On the basis of polyacrylamide gel electrophoresis, the preparation from the cell extract, obtained after ultrafiltration and ethanol precipitation, showed only an active band which stained with Coomassie-blue but not with Schiff-periodate. The α-galactosidase from C. javanica is α-galactosidase A, it lacks invertase and it reduces group B specificity of erythrocytes.

    Notes: Summary d-Xylanase (1,4-β-xylan xylanohydrolase, EC 3.2.1.8) was obtained from mycelial submerged culture of the mushroom Schizophyllum radiatum, grown on wheat straw pretreated with “steam explosion” as the substrate. The enzyme was purified 192-fold (specific activity 455 IU mg-1 protein), with 37% yield with respect to total d-xylanase activity. Polyacrylamide electrophoresis of the d-xylanase peak showed a single band of protein whose molecular weight, calculated by electrophoretic mobility, was 25 700. The enzyme exhibited maximum activity at pH 4.9 and 55°C. d-Xylanase was stable from pH 5.0 to 7.5; its half-life was 12 h at 45°C. The Michaelis constant was 9.5 mg ml-1 and V max 0.37 μ mole min-1. End-product analysis of the d-xylan hydrolysate showed the presence of d-xylobiose, d-xylotriose, d-xylotetraose, and d-xylopentose showing the mode of action of an endo-type enzyme.

    Notes: Abstract The morphological, cultural, and physiological characteristics are described of a yeast, LI70, which uses methanol as its source of energy and carbon; these characteristics have made it possible to identify the strain as Candida boidinii Ramirez. The identification was confirmed by a DNA-DNA genetic homology of 99.43% with the type strain of C. boidinii. Strain LI70 is not pathogenic.

    Notes: A continuous enzymatic hollow fiber reactor (HFR), obtained by immobilizing cellobiose active cells into the shell side of hollow-fiber modules, was studied. The HFR yield was monitored by glucose analysis resulting from hydrolysis of cellobiose. The residence time of substrate in the bioreactor to obtain convenient hydrolysis yields was calculated from tests carried out by varying the reactor dilution rate in the range 0.001-0.004 L/min. The glucose yield was measured for 300 h (continuous substrate flux). The yield decreased from 40 to 15%. This decrease was due to the loss of specific activity in the operating conditions and to the pressure drop increase from 0.2 to 1.7 atm. The pressure drop increase is in turn dependent on the cell loading (0.2-2.1 g dry cell) and the substrate flux.
