    Notes: Abstract The cortisol stress response to capture was investigated in two species of fish (Perca flavescens and Esox lucius) from sites polluted by high levels of polycyclic aromatic hydrocarbons (PAHs), polychlorinated biphenyls (PCBs), and mercury, and from reference sites in the St. Lawrence river system. Fish from the reference sites exhibited the normal elevation of serum cortisol in response to the acute stress of capture and had large pituitary corticotropes. In contrast, fish from the most polluted sites were unable to increase their serum cortisol in response to the acute stress of capture and their pituitary corticotropes were atrophied. These results suggest that a life-long exposure to chemical pollutants may lead to an exhaustion of the cortisol-producing endocrine system, possibly as a result of prolonged hyperactivity of the system.

    Notes: Abstract. Lipid peroxidation (LPO), measured as thiobarbituric acid reactive substances (TBARS), was evaluated in lungs of rats 24 h after intraperitoneal injection of 50, 250, and 1000 μg Cd/kg body weight as CdCl2. In order to gain some insight into possible causative factors responsible for these oxidative phenomena, the redox-active elements iron (Fe) and copper (Cu), and total lung protein content (an indication of pulmonary inflammatory processes) were also measured. Results obtained demonstrate a similar dose-related, non-linear evolution of total lung TBARS and total lung protein as a function of increasing lung Cd concentrations. Standardization of total lung TBARS to lung protein content further resulted in a linear relationship with lung Cd concentrations, thus suggesting a possible cause-effect relationship between these parameters. No statistically significant association was observed between the dose-related evolution of lung TBARS, and iron (Fe) and copper (Cu) after Cd exposure. The results obtained provide support for the possible involvement of inflammatory phenomena as the most likely events responsible for the generation of LPO in lung tissue following acute exposure to Cd salts.

    Notes: Abstract In order to determine the nature and extent of the problem caused by the presence of mercury in the environment of the James Bay territory a Mercury Agreement was signed in 1986 jointly by the Crees, the Govemment of Québec and Hydro-Québec. The objectives were to minimize potential hazards to human health arising from the presence of mercury in the environment, to alleviate negative impacts on the Crees, their way of life and their harvesting activities and to provide for remedial measures. Monitoring of mercury concentrations in fish show that, 15 years after impounding of La Grande-2 reservoir, the concentrations remain higher in reservoirs than in natural lakes but they are now decreasing in both predatory (Northern pike) and non-predatory (Whitefish) fishes. Monitoring of the mercury exposure among the Crees showed that concentrations in hair have decreased and are now stabilized, for most of the population, to concentrations that do not present a health risk. This decrease may be attributed to a reduction in fish consumption and/or a possible switch of the diet towards non-piscivorous fish, less contaminated by mercury.

    Notes: Summary The response of caudal neurosecretory cells in Salvelinus fontinalis to exposure to media in which one ion (Na, K, Mg, Ca and Cl) was either selectively enriched or depleted was evaluated by morphometric criteria. Morphological changes indicating stimulation of synthetic activity were observed: exposure to sea water with low potassium concentration was the most efficacious in inducing the increase in the average diameter of caudal cells and the number of nucleoli for both cells with lobed and with nonlobed nuclei. The proportion of cells with lobed nuclei was also increased. To a lesser degree and in decreasing order, experimental milieu with modified magnesium contents, either fresh water enriched in this ion or seawater depleted of the same ion, and fresh water enriched in calcium also resulted in significant increases in cell diameter and the number of nucleoli. The two cytological cell types, viz cells with non-lobed nuclei or cells with lobed nuclei, are interpreted as being linked to different levels of neurosecretory activity.

    Notes: Abstract The biological activity of natural and synthetic mineral fibers has been examined. Natural attapulgite [(Mg, Al)2Si4O10(OH).4H20], synthetic xonotlite [Ca3Si3O8(OH)2] and natural sepiolite [Mg2Si3O8.2H2O] were selected. Genotoxic effects were investigated by means of a well established cellular model based upon the measurement of unscheduled DNA synthesis (UDS) in rat hepatocytes in primary culture. The intrinsic capacity of the fibers (1 and 10 µ/ml) to induce UDS was first tested. None of the fiber types showed detectable UDS-eliciting activity. Also, the possible modulation of the cellular response to genotoxic agents by the materials was examined by exposing the cells to mixtures of 2-acetylaminofluorene (AAF) (0.05 and 0.25 µg/ml) and fibers (1 and 10 µg/ml). In these experiments, the UDS response was significantly diminished in the presence of xonotlite. This phenomenon may reflect changes in the uptake and/or metabolism of AAF or may result from an inhibition of DNA repair processes, the latter suggesting a possible cocarcinogenic potential for this synthetic silicate. These results point to the immediate necessity of studying more extensively the biological effects of fibrous materials that can be used as substitutes for asbestos.

    Notes: A quantitative time study of the incorporation of choline-3H, leucine-3H and galactose-3H in lung epithelial cells, was undertaken in vivo with electron microscopic autoradiography. Type II pneumocytes were selectively labeled with choline-3H, a specific precursor of phosphatidylcholine, which is the main component of pulmonary surfactant. Choline was initially localized in endoplasmic reticulum, then was rapidly transferred through the Golgi complex and stored in lamellar bodies. Previously undescribed small lamellar bodies are suggested as phospholipid carriers between Golgi complex and lamellar bodies. After initial incorporation in the endoplasmic reticulum, the leucine label migrated through the Golgi complex into lamellar bodies by fusion of multivesicular bodies, which are the carriers between the two structures. Galactose was modestly incorporated into lamellar bodies via the Golgi complex. Intra-alveolar myelin figures, recognized as excreted surfactant, were labeled 120 minutes after injection with the three precursors.These findings indicate that the synthesis of a secretory product by type II pneumocytes involves phospholipid, protein and polysaccharide precursors; the secretory product is segregated as lamellar bodies which are destined to be excreted into the alveolar cavity to become part of the lining layer.
