    Notes: Abstract: [3H]Acetylcholine efflux and Na+-K+ ATPase ion pump activity were measured concomitantly in rat cortical synaptosomes. Ouabain (500 μM), strophanthidin (500 μM). and parachloromercuribenzene sulfonate (500 μM) each inhibited ouabain-sensitive 86Rb uptake and elevated [3H]acetylcholine release independently of the external calcium concentration. Veratridine (10 μM), electrical field stimulation (60 V, 60 Hz, 5-ms pulse duration), or the calcium ionophore A23187 (10μg/ml) also inhibited ouabain-sensitive 86Rb uptake and released [3H]acetylcholine, but via a calcium-dependent process. Veratridine-induced [3H]acetylcholine release and ion pump inhibition were correlated over a wide range of drug concentrations and both effects were blocked by pre-treatment with tetrodotoxin (1 μM). The rate of [3H]acetyl-choline efflux from superfused synaptosomes was increased within 15 s of exposure to ouabain, strophanthidin, veratridine, A23187, or field stimulation, while ouabain-sensitive 86Rb uptake was significantly decreased within a similar interval. These results suggest that [3H]acetylcholine release is due at least in part to inhibition of Na+-K+ ATPase.

    Notes: The low-level leakage currents in thin silicon oxide films were measured before and after the oxides had been stressed at high voltages. Four components of current were identified. These components were the tunneling current, the capacitive current associated with the measurement sweep rate, a negative differential current associated with the voltage sweep through the changing oxide C-V characteristic, and an excess current that occurred after the high-voltage stress. The excess current was due to the charging and discharging of traps generated inside of the oxide by the high-voltage stress. The excess current was proportional to the number of traps generated in the oxide. The magnitude of the excess current could be changed by changes in the measurement procedures due to the charging and discharging of traps. A major portion of the stress-generated excess low-level leakage current may not be a current that flowed through the oxide, but may be a trap charging/discharging current. This paper will concentrate on describing the low-level pretunneling leakage currents and the measurement techniques needed to determine the properties of the excess leakage currents associated with the traps generated inside of the oxide.

    Notes: Abstract— Abstract-Tumors of the human nervous system were utilized to investigate the cellular distribution of N-acetyl-L-aspartic acid (NAA). Astroglial tumours contained about 0.144 μmol/g. Oligodendrogliomas and medulloblastomas contained somewhat larger amounts. However, the level of NAA in all gliomas studied was less than that of normal human white matter. NAA was undetectable in meningiomas and acoustic neurinomas. If these results may be taken as representative of normal tissue, they imply a predominantly neuronal localization for NAA.Substantial amounts of NAA were found in peripheral nervous tissues and retina. Neurons seem to vary widely in NAA content.

    Notes: —A method is described for the preparation of relatively pure cholinergic synaptosomes from bovine cervical sympathetic ganglia. After dispersion of the tissue by incubation with collagenase, differential centrifugation yielded a crude synaptosomal fraction (P2), which was further purified by centrifugation on sucrose or Ficoll-sucrose density gradients. Assay of acetylcholine and choline acetyltransferase, and electron microscopy confirmed that presynaptic nerve endings survived the incubation procedure, appeared in the P2 fraction and thereafter sedimented to a density of approx. 1.13 in sucrose and 1.06 in Ficoll-sucrose. The washed P2 fraction had an acetylcholine content of 2.3 nmol/mg of protein, which is at least three times higher than the corresponding fraction from brain.

    Notes: GABA at physiological concentrations has been shown to stimulate the synthesis of arginine phosphate in lobster walking nerves. This effect can be observed both in intact axons and in axonal homogenates. The stimulatory activity of GABA on the synthesis of arginine phosphate appears to be on the Vmax of arginine phosphokinase whereas in the reverse reaction the effect is referable to the Km of ADP.

    Notes: Abstract— A method is described for the isolation of relatively pure cholinergic synaptic vesicles by hypo-osmotic shock and density gradient centrifugation of a synaptosome fraction prepared from bovine superior cervical ganglia. Vesicles from this source were found to sediment to a density equivalent to 0·3–0·41 M-sucrose. The vesicle subfraction from the gradient had an acetylcholine (ACh) content of 4.4 nmol/mg of protein and were subject to leakage of ACh. By a ‘tagging’ technique, the vesicles were counted under the electron microscope and their numbers related to their ACh concentration. After correction for leakage, an ACh content of 1630 molecules/vesicle was estimated.
