    Notes: Abstract  Fourteen deep dives were conducted along the southern slope of the Côte d’Ivoire–Ghana marginal ridge. Detrital rock samples reveal Early Cretaceous synrift sediments. In situ observations and microtectonic analysis identify the nature and the timing of three successive episodes of deformation. Clay layers are affected by early diagenetic deformational structures such as slump microfolds and normal microfaults. Most of the clastic deposits display a second deformational episode. These structures result from a layer-parallel extension and are related to extensional synrift tectonics. The last stage of deformation is identified by right lateral wrench folds associated with brittle joints and slaty cleavage. It is attributed to the syntransform evolution of the Côte d’Ivoire–Ghana marginal ridge.

    Notes: Microbiological contamination was determined in core samples from the surface of the ham, shoulder, belly and back of skinned and scalded pork carcasses. Aerobic plate counts of mesophilic organisms indicated that ham and shoulder areas of scalded carcasses had higher bacterial numbers than skinned carcasses. Contamination on the back of the carcass was similar for skinned and scalded carcasses, while skinned carcass bellies had slightly higher counts than did scalded bellies. Numbers of psychrotrophic bacteria were negligible in all cases. Weights of primal cuts were compared in skinned and scalded carcasses by alternately skinning one side of each of 23 pork carcasses, while the other side was left skin-on. Belly, picnic shoulder and ham yields were less for skinned sides, while the loin and Boston shoulder were not affected.

    Notes: The numbers of psychrotrophic bacteria in prerigor ground pork were slightly greater than in postrigor ground pork throughout an 11-day storage period at 2°C. Freezing both types of samples and subsequent thawing before storage at 2°C reduced the initial psychrotrophic counts but the freezing effects did not persist beyond 8 days of storage. Differences in psychrotrophic counts between prerigor and postrigor ground pork were not sufficiently great in any comparison to limit the use of prerigor grinding of pork.

    Notes: Alternaria alternata, A. solani, Colletotrichum coccodes, Fusarium oxysporum, Geotrichum candidum, Rhizopus stolonifer and Drosophila flies were added to tomato and fruit purees and nectars at 0.1 - 10 mg (dry weight)/2g sample, hydrolyzed at 121°C in potassium hydroxide (KOH) and analyzed colorimetrically for glucosamine. Glucosamine varied with product, quantity of mycelium, hydrolysis time, KOH concentration, age (older cultures gave highest values), and mold species (R. stolonifer gave maximum and G. candidum, minimum values). When processed foods spiked with molds were analyzed, the mold concentration was linearly correlated to glucosamine but not to Howard mold count. Drosophila were barely detectable in hydrolyzed foods. Hydrolysis may be the critical step in this procedure.

    Notes: Abstract: Ornithine decarboxylase (ODC), the rate-limiting enzyme in the biosynthesis of polyamines, was measured in the brain and the liver of adrenalectomized rats after an acute S.C. treatment with glucocorticoids. The effects of corticosterone and dexamethasone were compared in three brain areas, the cerebral cortex, hippocampus, and cerebellum. These structures have similar concentrations of cytosolic glucocorticoid receptor, as measured by an in vitro exchange assay using a specific glucocorticoid ligand, [3H]RU 26988, but contain different amounts of mineralocorticoid receptor. Corticosterone and dexamethasone increased ODC activity in the liver and brain areas in a dose dependent manner, dexamethasone being more active than corticosterone in all tissues. Moreover, estradiol, progesterone, and testosterone were inactive. Aldosterone, at high doses, increased brain ODC activity. Glucocorticoids, selected for their weak binding, or lack of binding to the mineralocorticoid receptor, were tested and found to be highly active in inducing brain and liver ODC, thus showing that ODC induction by steroids is specific for glucocorticoids. These results are among the first to suggest biochemically a central action of glucocorticoids following an acute treatment and confirm that the brain is a glucocorticoid target organ.

    Notes: Abstract: Upon addition of the cardiac glycoside ouabain to cultured cerebellar granule cells, an immediate increase in intracellular free sodium is evoked mediated by two pathways, a voltage-sensitive channel blocked by tetrodotoxin and a channel sensitive to flunarizine. Ouabain induces a steady plasma membrane depolarization in low Ca2+ medium; whereas in the presence of Ca2+, a distinct discontinuity is observed always preceded by a large increase in intracellular free Ca2+ ([Ca2+]c). The plateau component of the increase can be inhibited additively by the L-type Ca2+ channel antagonist nifedipine, the spider toxin Aga-Gl, and the NMDA receptor antagonist MK-801. Single-cell imaging reveals that the [Ca2+]c increase occurs asynchronously in the cell population and is not dependent on a critical level of extracellular glutamate or synaptic transmission between the cells. A prolonged release of glutamate is also observed that is predominantly Ca2+ dependent for the first 6–10 min after the evoked increase in [Ca2+]c. This release is four times as large as that observed with 50 mM KCl and is predominantly exocytotic because release was inhibited by tetanus toxin, the V-type ATPase inhibitor bafilomycin, and Aga-Gl. It is proposed, therefore, that ouabain induces a period of membrane excitability culminating in a sustained exocytosis above that observed upon permanent depolarization with KCl.

    Notes: Abstract : The recycling of synaptic vesicles in nerve terminals involves multiple steps, underlies all aspects of synaptic transmission, and is a key to understanding the basis of synaptic plasticity. The development of styryl dyes as fluorescent molecules that label recycling synaptic vesicles has revolutionized the way in which synaptic vesicle recycling can be investigated, by allowing an examination of processes in neurons that have long been inaccessible. In this review, we evaluate the major aspects of synaptic vesicle recycling that have been revealed and advanced by studies with styryl dyes, focussing upon synaptic vesicle fusion, retrieval, and trafficking. The greatest impact of styryl dyes has been to allow the routine visualization of endocytosis in central nerve terminals for the first time. This has revealed the kinetics of endocytosis, its underlying sequential steps, and its regulation by Ca2+. In studies of exocytosis, styryl dyes have helped distinguish between different modes of vesicle fusion, provided direct support for the quantal nature of exocytosis and endocytosis, and revealed how the probability of exocytosis varies enormously from one nerve terminal to another. Synaptic vesicle labelling with styryl dyes has helped our understanding of vesicle trafficking by allowing better understanding of different synaptic vesicle pools within the nerve terminal, vesicle intermixing, and vesicle clustering at release sites. Finally, the dyes are now being used in innovative ways to reveal further insights into synaptic plasticity.

    Notes: The haemodynamic effects of propofol and ketamine were studied in two groups of eight randomly allocated elderly patients (mean age 85.8 years) anaesthetised for hip replacement. Group 1 patients received propofol 1 mg/kg by intravenous bolus for induction and 0.1 mg/kg/minute by continuous infusion for maintenance. Group 2 patients received ketamine 1.5 mg/kg by intravenous bolus as induction dose and 50 μg/kg/minute by continuous infusion for maintenance. All patients breathed spontaneously via a facemask at FIO2 1.0. Haemodynamic status was established before induction and at 1, 3, 5, 10 and 15 minutes after induction. Arterial pressure and cardiac output decreased slightly in group 1 but heart rate, right atrial pressure and pulmonary arterial pressure remained unchanged. Myocardial oxygen consumption showed a significant decrease of 27%. There was a significant increase in blood pressure and pulmonary capillary wedge pressure (by 97%) in group 2. Cardiac output and systemic vascular resistance remained unchanged whereas myocardial oxygen consumption showed a very significant increase of 100%.

    Notes: A disease of Populus nigra‘Italica’ associated with foliar yellowing, sparse foliage, stunting, dieback, and decline was observed in south-western Germany; a witches’ broom disease of Populus alba that is known in other countries was also detected in Hungary and Germany. The aetiology of the diseases was studied by fluorescence microscopy and polymerase chain reaction (PCR) amplification. Using fluorescence microscopy, phytoplasmas could be detected only in P. alba. However, most diseased trees of P. nigra‘Italica’ tested phytoplasma-positive by PCR. In some of the trees the phytoplasma numbers were so low that nested PCR was required to detect the infection. Very low phytoplasma numbers were also observed in diseased Populus tremula. The identity of phytoplasmas from P. nigra‘Italica’ sampled in Germany and France, P. alba and also P. tremula was examined by restriction fragment length polymorphism (RFLP) analysis of PCR-amplified ribosomal DNA. In all poplars, phytoplasmas of the aster yellows group were detected. However, three different RFLP groups were identified that consisted of (1) French strains from P. nigra‘Italica’, (2) German strains from P. nigra‘Italica’ and (3) strains from P. alba and P. tremula. The profile observed in the last group was probably the result of sequence heterogeneity in the two 16S RNA genes.
