Notes: Multistep stress-relaxation experiments in torsion have been done on a single crystal of the long-chain hydrocarbon compound n-eicosane. It was found that at very small strains the behavior approaches that of a linear viscoelastic material, whereas at larger strains the behavior becomes increasingly nonlinear and time dependent. In the region where the stress-strain behavior becomes nonlinear, it is shown that the second-step response, in an experiment in which the magnitude of the second step is one half that of the first step, can be described very well using a recent constitutive equation proposed by Zapas.

Notes: Formalin-fixed, paraffin-embedded tissue sections from 26 malignant fibrous histiocytomas (MFH) and 61 benign fibrohistiocytic proliferations (BFHP) were evaluated immunohistochemically. An avidinbiotin-pcroxidase technique was used to determine immunoreactivity for alpha-1 antichymotrypsin, muramidase, HLA-DR, leucocyte common antigen, S-100 protein, vimentin, desmin, and keratin. MFHs were consistently positive for ACT and vimentin and inconsistently reactive for the other antigens. MFHs were negative for LCA suggesting a mesenchymal origin for these lesions. In the MFH histologic subtypes, antigen expression was not significantly different to be useful in their classification. Also no distinctive pattern emerged relative to immunoreactivity and tumor location. The benign lesions, giant cell tumor of tendon sheath, dermatofibroma, and oral benign fibrous histiocytoma differed from the MFHs in that they were often LCA positive, suggesting origin from hcmatopoetic mononuclear-macrophages. The immunoprofiles of peripheral fibromas and “giant cell” fibromas were felt to be consistent with origin from mesenchymal cells. Several of the antigens studied could be used to differentiate the benign lesions studied from other benign neoplasms. The antigens were, however, of little value in separation of benign and malignant lesions.

Notes: Blood vascular and lymphatic tumors were evaluated immunohistochemically by studying a spectrum of endothelial associated antigens. (1) UEA-I lectin reacted with the tumor cells of one patient with malignant angioendothelioma in the non-metastatic stage. However, when metastasis occurred, the binding sites of this lectin completely disappeared from the surface of the tumor cells in both original and metastatic lesions, suggesting the loss of blood group H antigen from the tumor cells could be used as an indicator of metastasis in this tumor. (2) Reaction with anti-HLA-A, B, C, intense in normal blood vessels, remained intensely positive in pyogenic granuloma and Kaposi's sarcoma, whereas it did not react with normal lymphatics and lymphangioma. This indicates that anti-HLA-A, B, C is useful in differentiating blood vascular structures from lymphatic structures in both normal and pathological conditions. (3) OKM5 reacted intensely with benign hyperplasius in pyogenic granuloma, while barely reacting with proliferating parts in Kaposi's sarcoma, suggesting the difference in staining patterns can be used to distinguish vascular proliferation or malignancy. (4) Reaction with anti-Type IV collagen and anti-laminin was intense in normal blood vessels, pyogenic granuloma and Kaposi's sarcoma, whereas reaction with these antibodies in normal lymphatics was patchy and irregular in its thickness.

Notes: Abstract Introduction: Determination of the risk of invasive bladder tumors progressing is still imprecise due to the heterogeneous biological behavior of this neoplasm. The goals of this study were to evaluate the patterns of expression of the epidermal growth factor (EGF) system in invasive bladder cancer and to assess its prognostic value. Methods: This immunohistochemical study was performed using fresh frozen tumor samples and a panel of monoclonal antibodies on a series of 43 invasive bladder cancers treated by cystectomy. Results: EGF was detected in 45% of the tumors and did not correlate with survival from bladder cancer. Transforming growth factor alpha (TGFα) was expressed by 60% of the tumors and correlated strongly with death from bladder cancer. Epidermal growth factor receptor (EGF-R) expression was seen in 86% of cases and had no prognostic significance. cerbB2 was expressed in 50% of cases and was inversely related to a poor prognosis. When EGF and TGFα were both expressed, there was little or no expression of c-erbB2. Conclusion: The accumulation of several growth factors and the relevant receptor are necessary for the progression of invasive bladder cancers. They could be used as indicators of tumor aggressiveness.

Notes: Abstract Kinetic index determined by enumeration of neoplastic cells positive for proliferative cell nuclear antigen (PCNA) in 70 breast carcinomas (avidin-biotin immunoperoxidase technique) was compared to synthesis-phase fraction (S-phase, or SPF) values obtained by flow cytometry (FCM) using a multiparametric, 2 color method (dual-label propidium iodide/cytokeratin-FITC). The percent PCNA positive tumor cells (12.5% mean, range 1–28%) was significantly greater in aneuploid tumors (14.2% mean, N=35) compared to diploid range tumors (10.7% mean, N=35) (p〈0.05), and was correlated with SPF derived from ungated DNA histograms (12.5% mean ± 5.5%, r=0.45, p〈0.001). Marginally stronger statistical correlations were observed between the PCNA index and SPF values calculated from cytokeratin-gated (15.8% mean, r=0.53, p〈0.001) DNA histograms or from SPF values obtained following linear baseline debris subtraction (mean=8.1%, r=0.48, p〈0.001). Significant associations were identified between PCNA index and prognostically important clinicopathologic parameters including nuclear grade (p=0.014), presence of necrosis (p=0.005), and angiolymphatic invasion (p=0.003). We conclude: 1) PCNA index is comparable to FCM SPF and correlates with factors of known prognostic importance in carcinoma of the breast; 2) baseline debris and contaminating events derived from non-epithelial cells both represent significant artifacts in proliferative fraction estimates derived from FCM DNA histograms; and 3) multiparametric analysis may represent one means of improving the specificity and clinical value of FCM SPF determinations.

Notes: Creep and creep rupture of PMMA at high stresses have been characterized and found to be relatable by use of reduced variables. It is shown that when the creep compliances can be correlated by a superposition principle for which the vertical shift is the ratio of the applied stress to a reference stress and when strain at failure is a constant, a commonly used failure criterion (that the product of the strain rate at failure and the time to failure is constant) becomes valid. The reduced variables approach is found to apply to two greatly different thermal histories. Consistent with the concept of physical aging, the response of a quenched sample is simply shifted along the log time axis to shorter times relative to the response of the aged sample.

Notes: The damping, resonant frequency, and dynamic elastic storage modulus of five polyamides have been investigated at resonant frequencies ranging from 100 to 2000 cycles/sec. at temperatures in the 100 to 450°K. region. Specimens of poly(hexamethyleneadipamide) containing 0, 0.9, 3.3, and 6.4 wt.-% water were studied and it was found that with increasing water content: the loss peak and associated modulus dispersion at 170°K. in the dry material decreased; the loss peak and modulus dispersion at 370°K. in the dry material shifted to lower temperatures reaching a value of 280°K. for 6.4% water; a third damping peak appeared at 240 to 245°K. and the modulus in the 150 to 250°K. region increased while above and below this range the reverse was true. A poly(hexamethylene adipamide) specimen containing 10.5 wt.-% methanol exhibited one broad peak at about 255 to 260°K. Dry specimens of poly(hexamethylene sebacamide), poly(decamethylene sebacamide), 17% N-methylated poly(decamethylene sebacamide), and 58% N-methylated poly(decamethylene sebacamide) were also investigated. For the latter three specimens the principal amorphous transition shifted from 340 to 285°K. and the area under the loss peak increased greatly in going from 0 to 58% N-methylation, while the low temperature transition remained at 160 to 170°K. essentially unchanged. Of the five polymers studied the 17% N-methylated poly(decamethylene sebacamide) exhibited the highest apparent modulus in the 110 to 160°K. region; and at all temperatures below the principal amorphous transition this material had a higher modulus than the unmethylated parent polymer.

Notes: An attempt has been made to determine what influence chain folds may have on the α and γ mechanical loss peaks in linear polyethylene. In so doing, one long-chain n-paraffin (C94H190) and two low molecular weight polyethylene fractions have been examined with mechanical relaxation, differential scanning calorimetry (DSC), and low-angle x-ray diffraction techniques. The data suggest that chain folds play a prominent role in both the α and γ processes but that other factors such as polydispersity and/or branching are also important.

Notes: The results of creep and recovery experiments are reported for a sample of ultrahigh-molecular-weight polyethylene at three different temperatures and creep times in the range from 10 s to 1.47 X 106s. In order to approximate constant-true-stress conditions all the experiments were carried out at the same value of applied stress (4 MPa) and the change in strain during creep was never more than about 2%. It was found that within the range of conditions used the creep and recovery data could be described quite well by a one-dimensional constitutive equation consisting of two terms, one a single integral hereditary term and the other a plasticity term. Further, it was shown that to a very good approximation the idea of time-temperature superposition can be applied to the description of the hereditary term Ĵ (σ, T, t). We believe that the results described here provide greater insight into what constitutes “mechanical preconditioning” of semicrystalline polymers and further provide a relatively simple method for describing the uniaxial creep and recovery behavior of such materials.
