    Notes: The influence of animal feed quality on lipid and cholesterol oxidation in whole milk powder was investigated. Powders from a herd receiving a ‘supplemented’ diet showed reduced PV (p 〈 0.01) and TBARS (p 〈 0.09) compared to a ‘restricted’ herd, after storage in both vacuum and sachet-packs and less (p 〈 0.003) cholesterol oxidation products (COPs). High pre-heating temperatures resulted in higher levels of PV, TBARS and COPs in fresh whole milk powders than low pre-heat temperatures, but after storage the reverse occurred. Superior animal feed quality and proper control of processing and storage conditions enhanced oxidative stability of whole milk powder. Lipid and cholesterol oxidation were positively correlated (p 〈 0.001).

    Notes: The effects of grass dry matter (DM) allowance and dietary supplements of full fat rapeseeds on levels of cis-9,trans-11 octadecadienoic (CLA) acid in bovine milk were investigated. Grass allowance of 16 kg/(cow*day) resulted in reduced (p〈0.05) milk fat CLA levels (3.91 mg CLA/g fat) compared to 20 kg/(cow*day) after 19 wk treatment. CLA levels increased in milk fat from cows on a high-rapeseed-supplemented diet (p〈0.001) (1650 g/(cow*day) full fat rapeseed) compared to the control (pasture) and low rapeseed (p〈0.01) (825 g/(cow*day) full fat rapeseed) supplemented diets. The variation in milk fat CLA levels among individual cows over both trials was 1.5–16 mg/g.

    Notes: Summary This study was concerned with the effect of insulin and catecholamines on the rate limiting enzymes of cholesterol metabolism in rat hepatocytes. Insulin was found to increase the activity of 3-hydroxy-3-methyl glutaryl coenzyme A reductase and to have no effect on the activity of acyl-coenzyme A: cholesterol-o-acyltransferase. Noradrenaline and isoprenaline increased the activities of both 3-hydroxy3-methyl glutaryl coenzyme A reductase and acyl-coenzyme A: cholesterol-o-acyltransferase. The effect of noradrenaline or isoprenaline in the presence of insulin was that of a lower stimulatory response on 3-hydroxy-3-methyl glutaryl coenzyme A reductase but comparable to that found with either catecholamine alone. The combination of either catecholamine with insulin had no effect on the activity of acyl-coenzyme A: cholesterol-o-acyltransferase. These observations suggest that the activities of 3-hydroxy-3-methyl glutaryl coenzyme A reductase and acyl-coenzyme A: cholesterol-o-acyltransferase are regulated independently by insulin in the presence or absence of catecholamines. By contrast, catecholamines appear to regulate both enzyme activities in a similar fashion.

    Notes: Summary Serum lipoproteins and key hepatic and intestinal enzymes regulating cholesterol synthesis, esterification and catabolism, namely 3-hydroxy-3-methylglutaryl coenzyme A (HMGCoA) reductase, acyl coenzyme A: cholesterol-o-acyltransferase (ACAT) and cholesterol 7α-hydroxylase respectively, were compared in two hypercholesterolaemic rabbit models — the cholesterol-fed animal and the hypercholesterolaemic diabetic animal. Hypercholesterolaemia in the cholesterol-fed animals was reflected in the VLDL and LDL fractions, whereas VLDL and HDL2 cholesterol levels were elevated in the diabetic animals. The lipoproteins of the cholesterol-fed animals were enriched with cholesterol but the lipoprotein fractions in the diabetic animals were enriched with triacylglycerol. While hepatic HMGCoA reductase activity was significantly reduced in both groups, the activities of hepatic ACAT and cholesterol 7α-hydroxylase were significantly increased in the cholesterol-fed animals and significantly reduced in the diabetic animals compared with controls. In the intestine, the activity of HMGCoA reductase was increased and ACAT reduced in the diabetic animals. By contrast, in the cholesterol-fed group, HMGCoA reductase activity was lower and ACAT activity was higher in comparison with the control group. These differences in lipoproteins and cellular cholesterol metabolism between the hypercholesterolaemic rabbit models may explain the differences in susceptibility to atherosclerosis, previously reported in these two animal models.

    Notes: Abstract Suspension cultures of isolated rat hepatocytes were used to investigate whether 7-ketocholesterol and cholestane-3β,5α,6β-triol exert oxidative stress in cells as manifested by increased lipid peroxidation and the induction of the antioxidant enzymes catalase, glutathione peroxidase and superoxide dismutase. The oxysterols were found to increase the levels of both superoxide dismutase and catalase and to have variable effects on glutathione peroxidase activity. Increased lipid peroxidation was not observed, indicating that the endogenous antioxidant defense system was capable of protecting against any oxidative stress that might otherwise by exerted by 7-ketocholesterol or cholestane-3β,5α,6β-triol. Covi-ox, a natural tocopherol blend reduced the effects of both oxysterols on the antioxidant enzymes. A concurrent reduction in the production of thiobarbituric acid-reactive substances in Covi-ox-treated cells is indirect evidence that reactive oxygen species were produced by oxysterols in hepatocyte suspension cultures.
