    Notes: Abstract Due to mathematical ill-conditioning there exists a lack of confidence in the uniqueness of the solutions derived by the application of Laplace inversion techniques to photon correlation spectra. Criteria based on the use of multi-angle measurements of the diffusion coefficient are suggested for narrowing the range in the multitude of permitted solutions. Although some results on calibrated, monodisperse latices are included the method is mainly employed to calculate size distributions for casein micelle suspensions providing a ‘real’ system evaluation of the recently developed exponential sampling technique of Pike and Ostrowsky (Ostrowsky et al. (1981) Optica Acta 28: 1059–1070). Notwithstanding the use of exponential sampling software (POLY-BAS) here, the criteria apply equally well to solutions generated using the CONTIN procedure of Provencher [Provencher (1979) Makromol. Chem. 180: 201–209].

    Notes: The binding curves of 1-, 2-, 3-, and 9-aminoacridine and proflavine on native DNA and the circular dichroism (CD) spectra of the bound cations have been determined under the same conditions. The variation of the CD spectra with the amount (r) of aminoacridine bound per DNA phosphorus was of two main kinds: (1) the rotational strength of those aminoacridines which possess a 3-amino group depended markedly on r and decreased to relatively small values (or zero) at zero r; or, (2) the rotational strength changed relatively little with r and tended to a finite value at zero r. The relevance of these observations is discussed with respect to interelation models of the complexes and with respect to possible explanations of the basis of this induction of optical activity.

    Notes: The Ca2+-induced association of αs-casein has been studied using the methods of stopped-flow light-scattering and stopped-flow turbidity. The methods used are described, and the analysis of the results to give plots of Mw against time in the time-scale 0-15 sec is demonstrated. The validity of the method is discussed, and it is shown to be applicable to the association kinetics of aggregating proteins whose molecular-weight averages lie between 107 and 109. The method was used to study the association of bovine αs-casein in the presence of Ca2+, and results obtained are briefly discussed in terms of possible association mechanisms, and a mechanism for the overall reaction of αs-casein with Ca2+, and the subsequent precipitation of the caseinate is proposed.

    Notes: The binding isotherm of 2, 7-di-tert-butyl proflavine on calf thymus DNA has been measured by dialysis equilibrium. The CD spectra of complexes of the dye and DNA have been measured, and the variation of the induced circular dichroism of the dye with the amount of dye bound (r) has been found. The results show that di-tert-butyl proflavine binds to DNA in a completely different manner from proflavine itself, since both the visible and ultraviolet CD spectra of complexes of the two dyes with DNA differ markedly. The conformation of the nucleic acid is not affected by the binding of di-tert-butyl proflavine. It is possible that these results may allow determination, by using CD spectroscopy, of whether molecules intercalate into DNA.

    Notes: Complexes of proflavine with DNA and deoxyribonucleohistone from calf thymus show different optical activity in the visible and the ultraviolet. Although the visible CD spectra of both complexes arise from interactions of dye molecules, the variation in the optical activity with the amount of dye bound suggests that a lesser conformational mobility exists in DNH. This is confirmed by the ultraviolet CD spectra of the complexes, and it is suggested that the conformation of DNA within nucleohistone is altered by separation of the base pairs by a greater extent than occurs in DNA in free solution. Even if the protein is unequally distributed along the DNA, the conformation of all of the DNA is altered by its incorporation into the nucleoprotein complex, since no evidence could be detected to show that DNA in a “free” conformation existed.

    Notes: The circular dichroism spectra of complexes of the antibiotics daunomycin, nogalamycin, chromomycin, and mithramycin with calf thymus DNA have been measured over a range of drug/DNA ratios. The similarity of the CD spectra of bound chromomycin and mithramycin suggests that they have very similar binding sites, which produce strong effects on the CD spectra of the bound drugs, and remove the differences arising from local stereochemistry in the free drugs. It was found that it was not possible to predict whether the antibiotics intercalated, from studies of the CD spectra alone, even when comparisons were made with the CD spectra of aminoacridine-DNA complexes with intercalating or nonintercalating ligands.

    Notes: The title reactions have been investigated in a fast flow system at pressures of about 2 torr and temperatures between 12 and 132°C. The following Arrhenius equations are derived for reaction (2) where the units of k2 are l/mole sec and of E2, cal/mole, and the limits are the 95% confidence limits assuming random errors.These equations are in good agreement with those which can be derived from previous investigations.
