    Notizen: Abstract Somatic embryogenesis was achieved in callus cultures derived from immature cotyledonary explants ofHardwickia binata Roxb., a multipurpose leguminous tree, on semisolid modified Murashige and Skoog's (mMS) medium containing 2900 mg/l potassium nitrate (KNO3) supplemented with 4.64 µM kinetin (Kn) and 5.37µM a-naphthaleneacetic acid (NAA). Somatic embryos proliferated rapidly after transfer to MS basal medium supplemented with 2052.6 µM L-glutamine and 0.084 µM gibberellic acid (GA3). Maturation of somatic embryos was achieved on half-strength MS basal medium supplemented with 1.23 µM IBA and 2% (w/v) sucrose. Histological studies confirmed different developmental stages of somatic embryogenesis inHardwickia binata.

    Notizen: Abstract An experiment was conducted to optimise rooting in stem cuttings of the mangrove tree species Bruguiera parviflora, Cynometra iripa, Excoecaria agallocha, Heritiera fomes and Thespesia populnea. Cuttings were girdled or non-girdled and treated with the auxins indole-3-acetic acid (IAA), indole-3-butyric acid (IBA) and naphthalene-acetic acid (NAA) and amylase, IAA-oxidase, peroxidase and polyphenol oxidase activities were measured. The best rooting performance (in terms of root number) was in stem cuttings of E. agallocha followed by C. iripa and H. fomes when treated with IBA + NAA. IAA and IBA combinations were more effective in T. populnea and B. parvifolia. Variations in rooting response were markedly reflected in amylase, IAA-oxidase, peroxidase and polyphenol oxidase activities. Sharp increases in amylase and polyphenol-oxidase activities were associated with enhanced rooting in E. agallocha, C. iripa and H. fomes. On the contrary, lowered IAA-oxidase and peroxidase activities favoured rooting in T. populnea and B. parviflora. A higher number of roots occurred in pre-girdled cuttings in comparison to non-girdled cuttings.

    Notizen: Abstract Vegetative propagation through rooting in stem cuttings in five tree mangroves namely Bruguiera parviflora, Cynometra iripa, Excoecaria agallocha, Heritiera fomes, and Thespesia populnea using IAA, IBA and NAA was reported. Spectacular increase in the root number was noted in the cuttings of H. fomes and C. iripa treated together with IBA (5000 ppm) and NAA (2500 ppm). The highest number of roots was obtained with IBA (2500 ppm) and NAA (500 ppm) in E. agallocha. B. parviflora and T. populnea responded better to IAA and IBA treatment. The species specific variation in the rooting response to exogenous application of auxins was reflected in the metabolic changes during initiation and development of roots in cuttings. Biochemical analysis showed increase of reducing sugar in the above-girdled tissues at initiation as well as subsequent development of roots which was further enhanced by the use of auxins. Decreases in the total sugar, total carbohydrate and polyphenols and increase in total nitrogen were recorded in the girdled tissues and the high C/N ratio at the initial stage helped in initiation of roots in all the species. Interaction of IBA and NAA promoted starch hydrolysis better than IAA and IBA during root development and subsequently reduced the C/N ratio and increased the protein-nitrogen activity during root development which suggest the auxin influenced mobilization of nitrogen to the rooting zone.

Clumps of somatic embryos were separated and grown into complete plantlets when transferred to 1/2 MS medium+37 μM adenine sulphate with 2% (w/v) sucrose.
