    Notes: The structure of the ADP complex of the enzyme 3-phosphoglycerate kinase (PGK, E.C. 2.7.2.3) from Bacillus stearothermophilus NCA-1503 has been determined by the method of molecular replacement. The structure has been refined to an R factor of 0.16 for all data between 10.0 and 1.65 Å resolution, using data collected on the Hendrix–Lentfer imaging plate at the EMBL outstation in Hamburg. The r.m.s. deviations from stereochemical ideality are 0.010 and 0.011 Å for bonds and planes, respectively. Although crystallized in the presence of the nucleotide product MgATP, the high-resolution structure reveals the bound nucleotide to be MgADP reflecting the low intrinsic ATPase activity of PGK. Although the two domains of this enzyme are found to be some 4.5° closer together than is found in the yeast and horse-muscle apo-enzyme structures, this structure represents the `open' rather than the `closed', catalytically competent form, of the enzyme.

    Notes: The laccase from the fungus Coprinus cinereus has been prepared and crystallized in a form suitable for X-ray diffraction analysis. Small plate-like crystals of an enzymatically deglycosylated form of the enzyme have been grown by the hanging-drop method using polyethylene glycol as precipitant. These crystals diffract to at least 2.2 Å. They belong to the space group P212121 with cell dimensions a = 45.4, b = 85.7, c = 143.1 Å with a single molecule of laccase in the asymmetric unit.

    Notes: The structure of the catalytic core of the endoglucanase V (EGV) from Humicola insolens has been determined by the method of multiple isomorphous replacement at 1.5 Å resolution. The final model, refined with X-PLOR and PROLSQ, has a crystallographic R factor of 0.163 (Rfree = 0.240) with deviations from stereochemical target values of 0.012 Å and 0.037° for bonds and angles, respectively. The model was further refined with SHELXL, including anisotropic modelling of the protein-atom temperature factors, to give a final model with an R factor of 0.105 and an Rfree of 0.154. The initial isomorphous replacement electron-density map was poor and uninterpretable but was improved by the use of synchrotron data collected at a wavelength chosen so as to optimize the f′′ contribution of the anomalous scattering from the heavy atoms. The structure of H. insolens EGV consists of a six-stranded β-barrel domain, similar to that found in a family of plant defence proteins, linked by a number of disulfide-bonded loop regions. A long open groove runs across the surface of the enzyme either side of which lie the catalytic aspartate residues. The 9 Å separation of the catalytic carboxylate groups is consistent with the observation that EGV catalyzes the hydrolysis of the cellulose, β(1→4) links with inversion of configuration at the anomeric C1 atom. This structure is the first representative from the glycosyl hydrolase family 45.

    Notes: The temperature dependence of threshold current between 130 and 310 K of 1.65 μm In0.53Ga0.47As–InP bulk lasers grown by chemical beam epitaxy has been measured. Comparison with a calculation of the spontaneous recombination current at threshold allows one to determine the proportion of current loss over this temperature range. It is found that the loss can be described using an Auger recombination component of the form Rauger=C'n3 exp(−Ea/kBT) where n is the carrier population density in the undoped active region. The activation energy Ea is found to be 39±5 meV which is in excellent agreement with the theoretical value for the conduction to heavy hole band/split-off to heavy hole band Auger process. The values obtained for the Auger coefficient C over the temperature range are in close agreement with published values obtained by time resolved photoluminescence. © 1995 American Institute of Physics.

    Notes: We report the growth of nominally undoped InP by molecular beam epitaxy (MBE) from metallic indium, trimethylindium, or triethylindium and phosphine. We find significantly reduced acceptor incorporation when metalorganic sources are used, with exciton-dominated photoluminescence at 4.2 K and electron mobilities up to 47 500 cm2 V−1 s−1 at 77 K. The 4.2 K photoluminescence indicates that the residual shallow acceptors are either Ca, Mg, or Be. The carbon incorporation appears to be minimal. Interestingly, there is also little difference in electrical and optical properties of InP grown by metalorganic molecular beam epitaxy (MOMBE) using trimethyl or triethylindium, in marked contrast to the situation for GaAs MOMBE growth using trimethyl or triethylgallium.

    Notes: Herbage and faeces samples were retained from an intake and in vivodigestibility trial using material harvested at three different stages from five indigenous hill plant communities (those dominated by Agrostis-Festuca., Nardus stricta., Molinia caerulea., Eriophorum vaginatum and Tri-chophorum caespitosum) and from sown swards of ryegrass or white clover. Samples of the herbages and of their separate components, together with extrusa samples of the same herbages recovered from pen-fed sheep and cattle oesophageal fistulates, were digested in vitrousing rumen liquor. Measurements were made also of faecal nitrogen concentration (FN) and of indigestible acid-detergent fibre (IADF) using the samples from the original trial.The range of in vitrodisappearance values of the herbages (IVOMD; 0809-0278) was slightly wider than those of in vivodigestibility (OMD; 0-796-0-37I). Differences between OMD and IVOMD were greatest at low quality and relationships were best described by two separate linear regressions of OMD on IVOMD for (a) ryegrass, white clover, Agrostis-Festucaand Nardus(RSD; 0 0185) and, (b) Molinia, Tricho-phorumand Eriophorum(RSD; 00246). In vitrodisappearance values of extrusa were higher than those of herbages offered by 00503 and 00156 units for sheep and cattle respectively, partly because of greater levels of ensalivation, especially in the sheep. The relationships of OMD to IADF and to FN were poor, though inclusion of faeces output and fitting parallel lines for communities in the regression of OMD on FN reduced the RSD to 0020. We conclude that in vitrodigestion of samples of extrusa, using appropriate standards, is the best method of diet digestibility prediction for sheep and cattle grazing these communities.

    Notes: Summary. A survey of barley crops on 166 west of Scotland farms in 1969 indicated a more widespread distribution of Avena fatua L. than had previously been recognized. Whilst only 0·8% of the barley area was infested with A. fatua, the weed was found on 16% of the known barley-growing farms. A. fatua was associated with arable farming situations and with fields where there was a recent history of cereal cropping. Only on four of the sample farms where A. fatua was found had any attempt been made to control the weed. It is concluded that there is a strong case for eradicating or at least containing the weed before it becomes a major problem.The distribution of Avena strigosa Schreb. and Agropyron repens (L.)     Notes: The diffusion of Be from buried Be-doped layers in GaInAs has been studied for temperatures between 600 and 700 °C. An interstitial-substitutional model is proposed for the diffusion mechanism, which is dependent on growth conditions and consistent with the data presented. Under growth conditions where Be transport is minimized GaInAs junction field-effect transistors have been produced with transconductances in excess of 200 mS mm−1 for a 1-μm gate length.

    Notes: Insulated gate Ga0.47In0.53As/Al0.48In0.52As heterojunction field-effect transistors have been prepared using SiOx and SrF2 insulation and dilute acid and O2 plasma surface cleaning. The leakage current-gate voltage characteristics at different temperatures are analyzed by thermionic and field emission theories to extract the effective barrier heights. The transistor pinch-off characteristics and effective barrier height results are compared with elemental depth profiles obtained from Auger electron spectroscopy. It is concluded that O2 plasma cleaning followed by SiOx gate insulation, is the most suitable technique for fabricating devices and leads to an effective barrier height of 0.3±0.1 V. With refinement this fabrication route should lead to satisfactory room-temperature operation.
