    Notes: We recently found that alternative transcripts of tissue transglutaminase (tTG or TG2) were present in hippocampal brain regions of Alzheimer's disease (AD), but not in control, non-demented, age-matched brains. Since antecedent non-severe trauma has been implicated in AD and other neurodegenerative diseases, such as Parkinson's disease (PD) and amyotrophic lateral sclerosis (ALS), we were interested in whether alternative transcripts might be detected in a model of neurotrauma, controlled-contusion spinal cord injury (SCI) in the rat. Implicated in diverse roles from growth and differentiation to apoptotic cell death, only bifunctional tTG, of the nine member TG family, has dual catalytic activities: guanine trinucleotide (GTP) hydrolyzing activity (GTPase), as well as protein cross-linking. These functions imply two physiological functions: programmed cell life and death. These may have profound roles in the nervous system since studies in cultured astrocytes found tTG short (S) mRNA transcripts induced by treatment with injury-related cytokines. In the developing rat spinal cord, tTG activity is concentrated in ventral horn alpha motoneurons, but neither studies of spinal cord tTG gene expression, nor evaluation of the GTP-regulated isoforms in tissues, have been reported. We now report increased tTG protein and gene expression occurring rapidly after SCI. In parallel, novel appearance of a second, short form transcript, in addition to the normal long (L) isoform, occurs by 8 h of injury. Up-regulation of tTG message and activity following neural injury. with appearance of a truncated GTP-unregulated S form, may represent new approaches to drug targets in neurotrauma.

    Notes: In shoots of the garden pea (Pisum sativum L.), the main bioactive gibberellin (GA) is GA1, which is synthesised from GA20 by 3β-hydroxylation. Gibberellin A20 is produced from GA19, as part of the process known as GA 20-oxidation. Because these steps are thought to be negatively regulated by GA1, we compared the metabolism of labelled GA19 and GA20 in mutants deficient in GA1, with that observed in isogenic wild-type (WT) plants. There was a large and specific increase in the 3β-hydroxylation of labelled GA20 in the GA1-deficient (dwarf) mutants, compared with the WT. Metabolism experiments did not provide convincing evidence for feedback regulation of 20-oxidation, possibly because GA19 akppears to be metabolised rapidly, even in WT pea shoots. Both 3β-hydroxylase and 20-oxidase transcript levels were markedly higher in the mutants than in isogenic WT lines. The results sukpport previous suggestions that both biosynthetic steps are feedback-regulated by GA1 in pea.

    Notes: Field and laboratory studies suggest that different types of ooids form during quiet and agitated water conditions. Both types have been synthesized in the laboratory. Quiet water types exhibit a radial orientation of carbonate crystals, whereas in those formed in agitated conditions, a tangential orientation is prevalent.Successful laboratory formation of quiet water ooids was accomplished in supersaturated seawater solutions containing humic acids. Negative results were obtained from strictly inorganic solutions, and from those containing simple amino acids, single proteins, mixtures of proteins or mucopolysaccharides, soil and sediment extracts. Partly successful results were obtained using an organic extract from Bahamian ooids. The organic parameters most important in quiet water ooid formation are molecular weight, the presence of carboxyl groups and an ability to participate in hydrophobic/hydrophilic interactions, all of which are critical to membrane formation. Membranes form concentric shells which act as growth surfaces for carbonate and also induce the periodicity in carbonate precipitation.Ooids exhibiting a tangential orientation of batten-like crystals have been synthesized under conditions of agitation, supersaturation and without the intervention of organic processes during the precipitation. Complete growth may be divided into agitation, resting and sleeping stages In the agitation stage, quartz nuclei induce an inorganic, heterogeneous nucleation from a supersaturated solution, which finally ceases as a result of Mg2+ and possibly H+ poisoning of the carbonate surfaces. No further precipitation occurs until the crystal surfaces are reactivated by removal of Mg2+ and H+ during the resting stage. Following a series of agitation and resting stages, precipitation is inhibited by a degree of poisoning which is not totally removed during the resting stage. For further growth, a new substrate is required and is provided by the development of organic membranes around the grains. This occurs when the grains are buried in the subsurface, the period of organic growth constituting the sleeping stage. Only 2% of an ooid's life is spent growing in the agitated environment, while 95% of its life is spent accreting organic membranes in the subsurface.Our experimental work indicates that ooids of Bahamian type are inorganic precipitates. The tangential arrangement of battens is the result of suspension in an environment where the degree of turbulence is sufficient to induce grain to grain contact of sufficient strength and frequency to inhibit any crystal growth other than tangential. The role of organics is to provide a substrate for further growth after precipitation has slowed to a point when no further accretion is occurring.

    Notes: DNA-based genetic markers are now widely used by geneticists to locate genes for quantitative traits, and may also serve as a valuable tool for dissecting complex physiological phenomena. Van den Berg et al. (1996a QTL analysis of potato tuberization. Theor Appl Gen 93: 307–316), using restriction fragment length polymorphism (RFLP)-mapped populations of potato, detected eleven quantitative trait loci (QTLs) for tuberization. Taylor et al. (1992 Expression and sequence analysis of cDNAs induced during the early stages of tuberisation in different organs of the potato plant [Solanum tuberosum L.]. Plant Mol Biol 20: 641–651) have identified one of the genes associated with tuberization as that for the enzyme S-adenosylmethionine decarboxylase (SAMdc), an enzyme of the polyamine biosynthetic pathway. Chromosomal loci for SAMdc and arginine decarboxylase were established on the potato and tomato chromosomal maps, respectively, by hybridizing cDNA probes for these genes to RFLP digests. The polyamine content of leaves from an RFLP-mapped potato population was analyzed by fluorescence detection following HPLC, with quantitation using an internal standard. The data were analyzed by the ‘qGene’ statistical program, and QTLs for polyamines were detected on seven chromosomes. At least six QTLs were found for spermine, two for spermidine, and two for putrescine. A spermidine QTL was on chromosome 5 linked to marker TG441, very close to the place where SAMdc mapped. There was some congruence between QTLs for spermine and those previously detected for tuberization and dormancy, but relationships were not consistent.
