    Notes: Abstract During early development in fish, phospholipase A2 (EC 3.1.1.4) regulates membrane lipid modifications, which relates to changes in environmental conditions and provision of fatty acids required for metabolic energy substrates and prostaglandin biosynthesis. A method to analyze phospholipase A2 in rat tissues has been modified to measure its activity in embryonic Atlantic halibut (Hippoglossus hippoglossus L.). Egg and embryo samples were collected during the 1994 spawning season. Enzyme activity was undetectable at fertilization but in 10-d embryos was 230 pmol mg−1 h−1 (at 20 °C) and increased by ∼120% at hatch (17-d). Significant alterations in the fatty acid composition of important phospholipids, phosphatidylcholine (PC) and phosphatidylethanolamine (PE), were also observed. The content of some critical polyunsaturated fatty acids, and the ratio of unsaturated/saturated fatty acids, declined significantly over development. Acyl-chain restructuring mediated through the activity of phospholipase A2, coupled with other observed lipid changes (significant increases in the PC/PE ratio and cholesterol content), would produce a decreased fluidity of membranes during embryonic development, coinciding with the predicted upward movement of larvae in the water column. Arachidonic acid (20:4n-6) removed from PE could serve as a precursor for biosynthesis of 2-series prostaglandins, and eicosapentaenoic acid (20:5n-3) from PC is a likely source for other prostaglandin types. Despite removal of polyunsaturated fatty acids, there was an overall increase in lipid and fatty acid concentration, which can be attributed to amino acid catabolism during early developmental stages.

    Notes: Abstract. The need to accurately, reliably and economically screen large populations of brood salmon for infectious pancreatic necrosis virus (IPNV) prompted a fresh approach to the use of ELISA and cell cultures. The result is an integrated procedure that recognizes the limitations of each method while building upon the strength of both in the way that they are brought together, ELISA tests can never be sensitive enough to detect the very low virus levels typical of carrier fish, but are able to detect virus in infected cell cultures, both specifically and objectively. Cell cultures provide the means to detect very low virus levels, but without specificity and only through subjective observation. Without a suitable ELISA, the need to keep cell monolayers in a condition in which viral effects can be observed imposes strict demands on inoculation and culture procedures. In contrast, the new procedure described in this paper provides a simpler culture method in which the inoculum is retained throughout a single, uninterrupted 14-day culture cycle. This culture duration allowed maximum opportunity for viral replication in healthy cells, even though the monolayers became disorganized. However, the cultures were tested in the standardized ELISA, eliminating the need for microscopic observation and virus neutralization tests. Thorough validation studies confirmed the system's adequacy in practice.

    Notes: The relationship between the choruses of Seneca's tragedies and the action of the plays in which they occur is one of the least understood and most controversial aspects of the Roman dramatist's work. It is often asserted that Seneca's choral odes are mere act-dividers, that their relationship with the play's action is loose and unconvincing. I would not care to assert that the handling of the chorus is flawless in all instances in Seneca's tragedies (or indeed in the works of any ancient tragedian), but in his best works it is, I believe, masterly. In this paper I propose to illustrate the close and complex interconnection between ode and action in Senecan tragedy through an analysis of the choruses of Thyestes.

    Notes: This paper is a brief review of thermally induced covalent modifications to proteins in foods, focussing mainly on the advanced glycation end-products (AGE) of the Maillard reaction. Most foods are subjected to thermal processing, either in the home or during their production/manufacture. Thermal processing provides many beneficial effects, but also brings about major changes in allergenicity. Far from being a general way to decrease allergenic risk, thermal processing is as likely to increase allergenicity as to reduce it, through the introduction of neoantigens. These changes are highly complex and not easily predictable, but there are a number of major chemical pathways that lead to distinct patterns of modification. Perhaps the most important of these is through the reaction of protein amino groups with sugars, leading to an impressive cocktail of AGE-modified protein derivatives. These are antigenic and many of the important neoantigens found in cooked or stored foods are probably such Maillard reaction products. A deeper understanding of thermally induced chemical changes is essential for more advanced risk assessments, more effective QC protocols, production of more relevant diagnostic allergen extracts and the development of novel protein engineering and therapeutic approaches to minimise allergenic risk.

    Notes: This paper is a review concerning the way in which heat treatment can modify the allergenicity of food proteins. Any food protein may be allergenic if it can be absorbed intact, or as substantial fragments, through the gut mucosa and then evoke an immune (allergic) response. The intrinsic properties of the protein, the overall composition of the food, and the past processing history (especially thermal processing) all have an effect on the allergic potential. When a protein is denatured by heat, most of the original tertiary structure is lost, so that many of the sites recognized by antibodies on the native molecule are destroyed. There are many examples of allergenicity being reduced, but not eliminated, by heating. But heat-denatured proteins can also present new antigenic sites, uncovered by the unfolding process or created by new chemical reactions with other molecules present in the food (e.g., β-lactoglobulin associating with a-lactalbumin in milk). We have found that heat-denatured β-lactoglobulin has at least one new epitope, not found in the native state. Therefore, thermal processing can be part of a procedure for making hypoallergenic food, but will rarely be sufficient on its own. Increased understanding will help in evaluating novel proteins and processes.
